Cisplatin Ototoxicity In The Guinea Pig: An Electrophysiological, Morphological, And Biochemical Study, Sarah E. Barron

The purpose of this study was to determine changes induced by cisplatin on hair cell (HC) morphology, compound action potential (CAP), Preyer reflex (PR), and Na,K-ATPase activity in the cochlear lateral wall. Guinea pigs were treated with either 0.42, 0.83, 1.17, 1.50 mg/kg cisplatin, i.p. or sterile water for 12 consecutive days and tested on day 13. HC condition was visualized with scanning electron microscopy (SEM), CAP was initiated with a 1 volt click, and Na,K-ATPase activity was measured by the method of Fiske and SubbaRow. HC damage was linear with dose and occurred throughout the turns of the cochlea …

Twenty tortoises, 10 adult males and 10 adult females, were tagged with radio transmitters. Numbers were painted on each carapace to aid in visual sighting. The tortoises were released, and their movement was monitored over a one year period.
The percent recovery of tagged tortoises along with the numbers of non-tagged tortoises counted in sections of the study area, were used to determine tortoise population movement and location.
Weather data were collected and observations were made concerning the vegetation. These observations were compared with those from previous studies to see what impact seasonal climatic conditions and food availability had on …

Modification Of Chromium(Vi)-Induced Dna Damage By Glutathione And Cytochromes P-450 In Chicken Embryo Hepatocytes., Doreen Y. Cupo, Karen E. Wetterhahn

The role of glutathione and cytochrome P-450 in the production of DNA damage by chromium(VI) was examined in chicken embryo hepatocytes by the alkaline elution technique. Cellular levels of glutathione and cytochrome P-450 were altered by treating the hepatocytes with N-acetyl-L-cysteine, buthionine sulfoximine, isopentanol, or beta-naphthoflavone. A dramatic increase in chromium(VI)-induced DNA strand breaks was observed after increasing glutathione levels in the cells. Chromium(VI)-induced DNA strand breaks were even more numerous when the level of cytochrome P-450 was also increased. Upon depletion of glutathione levels and induction of cytochrome P-450 or cytochrome P-448, little or no DNA strand breaks or …

The microbial characteristics of a mixed population from the Passaic Valley Sewerage Commissioners (PVSC) Wastewater Treatment Plant in Newark were examined. This plant handles about 250 million gallons per day of mixed sewerage, with a large industrial component. The microbial species were characterized using standard plating techniques, morphology, microscopic observation, and biochemical tests. These characteristics were determined for the fresh liquor, and after successive batch exposure to phenol (100 ppm) and 2-chlorophenol (20 ppm). Predator/prey, gram positive to gram negative bacteria ratios, and the fungi and protozoan populations were also determined. These results were compared with those previously obtained using …

Safety procedures for performing geotechnical testing with hazardous and toxic materials, excluding radioactive wastes, are developed for the Flexible Wall Permeameter and the Atterberg Limits Tests. The tests were evaluated in detail to determine the critical points at which the technician would be exposed to the hazardous and/or toxic permeant. It was found that the following safety measures should always be taken; 1) the apparatus and all testing should be undertaken in a sliding sash hood using the proper face velocity, 2) air monitoring using a personal dosimeter and/or a detector tube is required to assure that the concentration of …

The Kinetics And Hepatotoxicity Of Benzyl Chloride Vapor In Rats, Hari V. Rao

Male Sprague Dawley rats (250g) were exposed to benzyl chloride vapor at a concentration of 220 ppm for 6 hours. During inhalation, the blood benzyl chloride concentration increased to reach a steady state level of 9-10 ug/ml between 4 and 6 hours of exposure. A small concentration of benzyl chloride (0.86 ug/ml) was present in the blood four hours after the termination of exposure. The absorption half life was 0.53 hours (0.39-0.81) and the elimination half life was l.12 hour (0.93-l.40).
The absorbed benzyl chloride was largely retained in the blood and some of it (0.22 ug/g) was sequestered into …

DTIC and BCNU have been demonstrated to have enhanced chemotherapeutic effects in mice bearing Bl6 mouse melanoma cells. The order of administration seems to be important in maintaining optimum effects. This was shown is a study by Hill et al. (unpublished).
In the present study, mouse melanoma cells, Bl6CL4, were treated with either DTIC or BCNU. The drug was allowed to remain on the cells for 2, 3, or 4 days. Morphine and exogenous opioid peptides alter the development of central noradrenergic neurons damaged by neonatal treatment with the neurotoxin 6-hydroxydopa. Transection of the dorsal noradrenergic bundle (DNB) in neonatal rats produces nearly the same alteration in the developmental pattern. It was of interest to determine whether morphine or an exogenous opioid peptide (metenkephalin) was able to modify the recovery of noradrenergic neurons after neonatal surgical transection of the DNB. Neonatal rats were divided into two groups. Both groups received saline (0.85%) or naloxone (2.0 mg/kg) by the intraperitoneal (i.p.) route. A number of immune abnormalities have been found in epileptic patients treated with antiepileptic drugs (AED). The alterations seen range from mild suppression of immunoglobulins to severly impaired humoral and cellular immunities. There is evidence for both drug effects and genetic or acquired factors as contributors to these abnormalities. In order to examine the basis for immune abnormalities in patients with epilepsy, a number of experimental designs were employed: clinical studies, in vitro studies, and use of an animal model.
Peripheral blood mononuclear cells (PBMC) isolated from epileptic patients currently receiving AED were found to have a reduced OKT4+/0KT8+ ratio. …

Based on an interpretation of the literature of phencyclidine (PCP) together with an examination of the relevant models or prototype structures, this research has attempted to design and synthesize a rigid analog of the phencyclidine structure. The parent ring systems suggested for this study were the 1-arnino and 2-arninornethyltetralins, 10 and, 12.
Several organic compounds, based on this design, have been prepared by classical examples of reductive alkylation reactions of the corresponding 2-hydroxyciethylene-1-tetralones, 11, and by the condensation and subsequent reductive alkylation of the 1-arninotetralin, 9. The Virginia State Water Control.Board, in cooperation with the Department of Chemical Oceanography of the Virginia Institute of Marine Science has recently conducted periodic chemical surveys of organic pollutants in the sediments of the upper James River. The August 1983 survey showed elevated concentrations of polycyclic aromatic hydrocarbons (PAH) at Stations ·4 and 7 (Figure 1 and Table 1). Although the reported concentrations were approximately one order· of magnitude lower than those reported in contaminated areas of the Elizabeth River, they were deemed high enough to warrant_ further study based on reported toxicity of Elizabeth River sediments (Hargis et al. …
