There has been growing interest in using N-methyl-D-aspartate (NMDA) receptor antagonists as treatments for mood disorders, but there is still much to learn about their cognitive effects. Research shows NMDA receptors can affect decision-making, and the antagonist MK-801 has had varying effects in rodents. Specifically, some have reported impairments in working memory while foraging behaviors remained intact, while others have demonstrated changes in choice behavior related to delay or risk in behavior tasks. This study examined the impact of Type 1 Diabetes Mellitus (T1DM) on executive function using a series of operant conditioning based tasks in rats. Sprague Dawley rats were randomized to either non-diabetic (n = 12; 6 male) or diabetic (n = 14; 6 male) groups. Diabetes was induced using multiple low-dose streptozotocin injections. All diabetic rodents were insulin-treated using subcutaneous insulin pellet implants. At week 14 of the study, rats were placed on a food restricted diet to induce 5 - 10% weight loss. The contactin-associated protein-like 2 gene, CNTNAP2, is a highly penetrant gene thought to play a role in the genetic etiology of neurodevelopmental disorders such as autism spectrum disorder (ASD). Despite its link to ASD, the field lacks a complete understanding of the role CNTNAP2 plays in the hallmarks of ASD: repetitive behaviours and abnormalities in social interaction, language, and sensory processing. Therefore, this thesis first examines if a loss-of-function mutation in the CNTNAP2 gene in the rat (SD-Cntnap2tm1Sage) is sufficient to cause alterations in social interactions, stereotypic behaviour, and sensory processing. The hApoE4 allele is one of the strongest genetic risk factors for Alzheimer’s disease (AD). It underlies amyloid-bdeposits and neurofibrillary tangles, the two hallmarks associated with AD pathology, and is subsequently associated with AD symptomology. Despite its importance, no rat animal studies to date use hApoE4 knock-ins. In addition to this deficit in the field of AD literature, the vast majority of AD studies focus on memory, even though executive function deficits may precede memory impairments in AD, and may be a predictor of AD development. The carotid bodies (CB), the primary peripheral chemoreceptors, respond to changes in blood gases with neurotransmitter release, thereby increasing carotid sinus nerve firing frequency and ultimately correcting the pattern of breathing. It has previously been demonstrated that acute application of the adipokine leptin caused perturbations of intracellular calcium and membrane ion movement in isolated CB Type I cells (Pye et al, 2015) and augmented the response of the intact CB to hypoxia (Pye et al, 2016). The synapse from the perforant path to the dentate gyrus has been widely used successfully to demonstrate long-term potentiation, a cellular model underlying learning and memory. Caffeine is one of the most widely consumed psychoactive stimulants in the world. Caffeine consumption increases in alertness, improvements in motor skills, and neurological functions, and these effects have promoted its use throughout history. Ischemic strokes account for 87% of all strokes and can have debilitating effects on language, sensory, and motor skills. Currently, tPA is the only medication approved by the FDA for the treatment of ischemic stroke, but the window of time to administer the drug is very small. In this thesis, we investigate the use of a simvastatin and fluoxetine drug combination (FS) as a possible treatment for ischemic stroke victims. Chronic or repeated exposure to stressful stimuli can result in several maladaptive consequences, including increased anxiety-like behaviors and altered peptide expression in brain structures involved in emotion. Among these structures, the bed nucleus of the stria terminalis (BNST) has been implicated in emotional behaviors as well as regulation of hypothalamic-pituitary-adrenal (HPA) axis activity. However, all of these studies were conducted in anesthetized, decerebrated, or spinalized mammals. Impulsive choice is defined as the preference for a small immediate reward over a larger delayed reward. Individual variablity in impulsive choice correlates with many socially relevant behaviors. Although forms of impulsive choice have been studied in both behavioral ecology and psychology, the exchange of knowledge between these fields is just beginning. Drawing from both of these fields will improve our research methods allowing for a more detailed understanding of this complex behavior. As the use of rodents in the study of the development and organization of segmental spinal cord circuitry increases, a complete account of the peripheral encoding and central connectivity of rodent muscle proprioceptors is necessary. The aim of this study is to investigate the long-term physiological consequences of early-life seizures in rats of different genetic backgrounds. Rats bred to be prone (Fast) or resistant (Slow) to amygdala kindling were induced with status epilepticus (SE) on postnatal day (PND) 10 by injecting 3 mg/kg i.p. kainic acid; SE consisted of seizures for 2 h, including stage 5 seizures (lying down with four limb tonic-clonic convulsions). Rats given ~24 h access to a 4% sucrose solution every 3rd day (E3DA) consume about 100 g more solution than rats with continuous, everyday access (EDA). Under the same EDA-E3DA conditions rats will consume similar amounts of a more concentrated 8 or 16% sucrose solutions (Eikelboom, Hewitt, & Adams, Unpublished). It maybe that with these more concentrated solutions rats hit a satiety limit that prevents a difference between EDA and E3DA consumption from being evident.
