Sensitive And Specific Detection Of Sars-Cov-2 In Saliva Using Reverse Transcriptase Loop-Mediated Isothermal Amplification, Casey Juntila

All NMU Master's Theses

A novel coronavirus called SARS-CoV-2 has caused the emergency release of the most well-known molecular assay, the CDC N1 RT-PCR assay, causing reports of poor analytical performance resulting in false-negative results (20, 26), and inconsistent testing kits received (23, 32). An immediate and dire need for a rapid and reliable SARS-CoV-2 testing workflow specifically designed for a university setting is the purpose this project is aiming and intended to fulfill. The workflow design uses a less invasive saliva sample for rapid screening using colorimetric RT-LAMP detection of three SARS-CoV-2 gene regions for Orf1ab, envelope, and nucleocapsid. Purified SARS-CoV-2 genomic RNA …

Development Of A Rapid Diagnostic Assay For The Colorimetric Detection Of Bacterial And Viral Pathogens Using Loop-Mediated Isothermal Amplification Coupled With A Peptide Nucleic Acid And Gold Nanoparticle Reporter System, William Brown Jul 2018

Development Of A Rapid Diagnostic Assay For The Colorimetric Detection Of Bacterial And Viral Pathogens Using Loop-Mediated Isothermal Amplification Coupled With A Peptide Nucleic Acid And Gold Nanoparticle Reporter System, William Brown

All NMU Master's Theses

Detection of pathogenic agents remains pivotal to the control and prevention of infectious disease. While diagnostic methods continue to break barriers, the need for accurate, sensitive, and rapid methods persists. Current methods for pathogen detection, including culture, immunochemical, or molecular-based techniques, are subject to significant limitations that restrict their clinical utility. Point-of-care (POC) tests have received attention for the diagnostic screening of infectious disease, with benefits of simplicity, affordability, and convenience. This study describes the development of a novel loop-mediated isothermal amplification (LAMP) reaction followed by a peptide nucleic acid (PNA) probe and gold nanoparticle (AuNP) detection system that results …

A Rapid Colorimetric Peptide Nucleic Acid Loop-Mediated Isothermal Amplification Assay For The Detection Of The Idh1 Mutation In Glioblastoma, Edward J. Raack Jul 2017

A Rapid Colorimetric Peptide Nucleic Acid Loop-Mediated Isothermal Amplification Assay For The Detection Of The Idh1 Mutation In Glioblastoma, Edward J. Raack

All NMU Master's Theses

The IDH1 mutation is an important diagnostic and prognostic biomarker used to characterize glioblastoma (GBM). Patients harboring the IDH1 mutation have improved overall survival following maximal resection. Knowledge of the IDH1 mutation status allows the surgeon to modify the surgical plan; however, no existing molecular test can provide this information intraoperatively. We designed a novel colorimetric peptide nucleic acid loop-mediated isothermal amplification (PNA-LAMP) method that rapidly detects the IDH1 R132H mutation in GBM. PNA-LAMP amplifies target DNA under isothermal conditions with high specificity and speed. The PNA prevents amplification of wild-type IDH1 DNA, while allowing amplification of the R132H variant …

Phylogenetic Studies Of The Madagascan Freshwater Crabs (Potamoidea, Potamonautidae, Deckeniinae), Rainee Stevens Apr 2017

Phylogenetic Studies Of The Madagascan Freshwater Crabs (Potamoidea, Potamonautidae, Deckeniinae), Rainee Stevens

All NMU Master's Theses

Relationships within the Madagascan freshwater crab fauna were examined based on unstudied specimens from all parts of Madagascar. This study allowed the examination of the validity of existing genera and species, and identified potential new taxa. The new specimens provided phylogenetic and evolutionary data, as well as new insights into the distribution patterns of the Malagasy freshwater crab fauna. In addition, the large number of new localities from previously unsurveyed areas of Madagascar allowed the construction of updated distribution maps.

The present analysis included 62 unidentified specimens plus 13 identified species of Malagasy freshwater crab taxa that had already been …
