Chitin is a naturally abundant polymer that also happens to be biodegradable. Chitin can be used in a variety of different products such as biodegradable plastics, papers, medical products, foods, and medical treatments. Protein, DNA, and RNA methyltransferases have an ever-expanding list of novel substrates and catalytic activities. Even within families and between homologs, it is becoming clear the intricacies of methyltransferase specificity and regulation are far more diverse than originally thought. In addition to specific substrates and distinct methylation levels, methyltransferase activity can be altered through formation of complexes with close homologs. This work involves the N-terminal methyltransferase homologs NRMT1 and NRMT2. NRMT1 is a ubiquitously expressed distributive trimethylase. The Influences Of Feeding Broilers On Graded Inclusion Of Sunflower Meal With Or Without Avizyme On Growth, Protein And Energy Efficiency, Carcass Traits, And Nutrient Digestibility, Mahmoud Alagwany, Adel Attia, Zenat Ibrahim, Mohamed Abd El-Hack1, Muhammed Arif, Mohamed Emam

The present study aimed to investigate the substituting effect of sunflower meal (SFM) for soybean meal (SBM) with or without supplemental Avizyme on growth performance, protein and energy efficiency ratios, carcass traits, and nutrient digestibility of broiler chickens. A 4 × 2 factorial design experiment was conducted with four levels of SFM replacing SBM (0%, 25%, 50%, 75%) and two levels of supplemented Avizyme (0 or 0.1 g/kg diet). Results revealed that increasing SFM substitution for SBM up to 50% was associated with an increase in body weight (P < 0.01). Daily weight gain was statistically (P < 0.01) enhanced with increasing SFM up to 50% through 22 to 42 and 7 to 42 days of age. Feed conversion ratio improved (P < 0.01) in groups given SFM at 25% and 50% compared to the control. No carcass values were affected by SFM inclusion or Avizyme. The inclusion of 25% or 50% SFM increased (P < 0.01) protein efficiency at periods of 7?21 and 22?42 days of age. Energy efficiency ratio improved (P < 0.01) due to SFM incorporation at levels of 25% or 50% in all periods. The interaction effect between SFM and Avizyme was not significant on a majority of studied parameters. The digestion coefficients of crude fiber, N-free extract, and organic matter increased (P < 0.05 or P < 0.01) with increased levels of SFM. In conclusion, the incorporation of SFM as a substitute for SBM in broiler diets improved the growth performance of broilers. The use of SFM up to 50% without Avizyme supplementation could be recommended in broiler diets from 7 to 42 days of age.

Gross Responses And Apparent Ileal Digestibility Of Amino Acids And Minerals In Broiler Chicken Fed Vegetable-Based Starter Diets Supplemented With Microbial Enzymes, Mohammad Abul Hossain, Paul Ade Iji, Al-Farooq Mohammad Fakhrul Islam

This study was conducted to investigate the effects of diet and exogenous enzymes on growth responses and micronutrient digestibility of broilers. Day-old broilers (n = 256) were distributed into a 2 x 2 factorial design and were fed on two basal diets: soybean (T1) or canola (T2) meals as such, or supplemented with enzymes up to 21 days. Feed intake (FI) and live weight (LW) to 21 days on the T2 diet was higher (P < 0.001) than that on T1. Feed conversion ratio (FCR) was better (P < 0.01) on T1 than on T2 at 21 days. Enzymes generally improved (P < 0.001) FI, FCR, and LW. The digestibility of histidine was higher (P < 0.05) on T2, but lysine digestibility was higher (P < 0.01) on T1 at 21 days. Histidine, threonine, lysine, valine, isoleucine, and phenylalanine digestibility was improved (P < 0.05) in the supplemented diets. Mineral digestibility was unaffected (P > 0.05) by diet, but enzymes increased (P < 0.05) digestibility of P, K, Mn, and Cu at 21 days. The digestibility of Cu, Zn, and Mg was higher (P < 0.05) on T2, whereas Ca digestibility was greater (P < 0.05) on the T1 diet. Broilers' growth responded positively to enzyme diets, probably due to improvement in nutrient digestibility.

Phospholipase D (PLD) is a major virulence determinant of Corynebacterium pseudotuberculosis. Various studies have focused on the expression of recombinant PLD (rPLD) enzyme in different Escherichia coli strains, but generally a high yield of insoluble protein was reported. The aims of this study were to express soluble rPLD by different methods in E. coli. Severe morbidity and mortality consequences result from irreversible inhibition of human acetylcholinesterase by organophosphates (OPs). Oxime-based reactivators are currently the only available treatments but lack efficacy in the central nervous system (CNS) where the most damage occurs. Computational docking and molecular dynamics (MD) simulations reveal complex structural barriers that may reduce oxime efficacy. These results may guide future drug designs of more effective countermeasures.

Biofilm formation by Pseudomonas fluorescens Pf0-1 requires the cell surface adhesin LapA. We previously reported that LapG, a periplasmic cysteine protease of P. fluorescens, cleaves the N terminus of LapA, thus releasing this adhesin from the cell surface and resulting in loss of the ability to make a biofilm. The activity of LapG is regulated by the inner membrane-localized cyclic-di-GMP receptor LapD via direct protein-protein interactions. Here we present chelation and metal add-back studies demonstrating that calcium availability regulates biofilm formation by P. fluorescens Pf0-1. The bacterial dinucleotide second messenger c-di-GMP has emerged as a central molecule in regulating bacterial behavior, including motility and biofilm formation. Proteins for the synthesis and degradation of c-di-GMP and effectors for its signal transmission are widely used in the bacterial domain. Previous work established the GGDEF-EAL domain-containing receptor LapD as a central switch in Pseudomonas fluorescens cell adhesion. LapD senses c-di-GMP inside the cytosol and relays this signal to the outside by the differential recruitment of the periplasmic protease LapG. Cystic fibrosis transmembrane conductance regulator (CFTR) inhibitory factor (Cif) is a virulence factor secreted by Pseudomonas aeruginosa that reduces the quantity of CFTR in the apical membrane of human airway epithelial cells. Initial sequence analysis suggested that Cif is an epoxide hydrolase (EH), but its sequence violates two strictly conserved EH motifs and also is compatible with other alpha/beta hydrolase family members with diverse substrate specificities. To investigate the mechanistic basis of Cif activity, we have determined its structure at 1.8-A resolution by X-ray crystallography. The cationic molecule thrombin-induced platelet microbicidal protein 1 (tPMP-1) exerts potent activity against Staphylococcus aureus. We previously reported that a Tn551 S. aureus transposon mutant, ISP479R, and two bacteriophage back-transductants, TxA and TxB, exhibit reduced in vitro susceptibility to tPMP-1 (tPMP-1(r)) compared to the parental strain, ISP479C (V. The influence of 2 enzyme supplements for diets based on wheat with high or low viscosity on performance and some intestinal parameters of male broiler chickens were investigated. The experiment was of 2 x 3 factorial design, with 2 types of wheat (high or low viscosity) and 3 enzyme treatments (none, Avizyme 1300-xylanase or Avizyme 1500-amylase). Ileal digesta viscosity was significantly (P < 0.029) higher in birds fed with high viscous wheat than with low viscous wheat. Ileal digesta viscosity (P < 0.001) and dry matter content (P < 0.002) were significantly reduced by both enzyme preparations. Body weight gain, feed intake, dry matter intake, feed conversion ratio, DM retention and wet or dry excreta output were not significantly (P > 0.05) affected by all treatments. There were no significant interactions (P > 0.05) between the effects of wheat type and enzyme supplementation. Polysaccharidase genes from rumen bacteria were transferred to and expressed in ruminal and non-ruminal Gram-positive bacteria. The transformation efficiency and genetic stability of polysaccharidase genes in bacteria from different habitats were investigated. PCR amplification of cloned polysaccharidase genes from Escherichia coli, Lactococcus lactis, Enterococcus faecalis, Streptococcus sanguis and S. bovis strain 26 showed that rearrangement of plasmid and the gene fragment did not occur. The DNA band sizes from all primers agreed with the expectations of some transformants of S. bovis JB1 being rearranged. The xylanase and \beta(1,3-1,4)-Glucanase activities encoding gene from the cellulolytic rumen anaerobe bacterium was cloned into Escherichia/Streptococcus shuttle vector for introduction into Gram-positive bacteria, including the rumen facultative anaerobe bacterium Streptococcus bovis. Activities due to the cloned gene decreased in the stationary phase in batch cultures of S. bovis, reflecting the sensitivity of the cloned enzymes to inactivation in the presence of accumulated lactic acid. Cloned gene activity was detected in the culture supernatant, indicating recognition of the cloned gene signal peptide by Gram-positive bacteria.

The study was carried out to investigate the effects of sunflower meal with an enzyme mixture containing \alpha-amylase, ß-glucanase, cellulase, lipase and protease substituted for soybean meal in broiler diets on performance, intestinal weight, viscosity and pH, as well as the frequency of sticky droppings and abdominal fat. This experiment was conducted to study the effects of different enzyme supplementation to wheat-and barley-based diets on the live weight gain, feed efficiency, dressing percentage and ileal pH of broiler chickens. Two hundred fourtyfive, day-old Ross-PM3 chicks were randomly assigned to a control and four treatment groups. Starter (1 to 21 days old) and grower (22 to 42 days old) diets were given without (control) or with different enzyme preparations, which are Avizyme 1200 (1000 mg/kg), Kemzyme Dry (500 mg/kg), Roxazyme-G (200 mg/kg) and Wheatzyme (700 mg/kg). Interieukin-1, a mediator of inflammation, or tumor promoting phorbol esters induce transcription of stromelysin, a metalloproteinase that degrades extracellular matrix molecules and that is overexpressed in diseases such as rheumatoid arthritis. Sequences required for induction of transcription of the human stromelysin promoter are contained on a 46 base pair fragment. This fragment contains a sequence with a high degree of similarity to the binding site for the transcription factor activator protein-1 (AP-1) and indeed, the AP-1 sequence of this fragment is necessary but not sufficient for the maximal response to phorbol 12-myristate 13-acetate (phorbol) or interleukin-1. Simian virus 40 (SV40) deletion mutants dlA2459 and dlA2475 express T antigens that lack the normal carboxy terminus. These mutants are called host range/helper function (hr/hf) mutants because they form plaques at 37 degrees C on BSC-1 and Vero monkey kidney cell lines but not on CV-1p monkey kidney cells. Wild-type SV40 can provide a helper function to permit growth of human adenoviruses in monkey kidney cells; the hr/hf mutants cannot. Regulation Of Queuine Insertion Into Transfer Rna: Effects Of Tumor Promoters, Bonnie Jean Brooks

The purpose of this study was to treat normal human fibroblasts with known tumor promoters and observe queuine modification of tRNA. The queuine insertion enzyme, tRNAguanine ribosyltransferase, was studied in vivo and in vitro. Tumor promoter-treated human fibroblast cultures exhibited variable queuine-insertion rates with a transient inhibition that correlated with variable levels of queuine modified tRNA over time in culture from passages 3 — 8. In contrast the in vitro studies showed that phorbol cetera and saccharin actually increased insertion with strong evidence indicating phosphorylation as a positive modulating force of the enzyme activity.
