Lectins are substances composed of proteins or glycoproteins that specifically bond to carbohydrates on the cell surfaces or inside the cell. Lectins are used to identify the carbohydrate types in the cellular membrane or cytoplasm. The aim of the study was to identify the location and distribution of the four types of lectins PNA (Peanut agglutinin- Arachis hypogea), WGA (Wheatgerm Agglutinin- Triticum vulgaris), Con A (Conconavalin A-Canavalia ensiformis), and SBA (Soybean Agglutinin-Glycine max) in dog testes and epididymites. The testes and the body of 10 adult dogs were collected from an animal shelter after castration. Following the standard histological procedures, …

Pet animals raised by humans do not have the chance to choose their own food. Therefore, mistakes made by humans in feeding dogs and cats will adversely affect the health of the animal. There are applications at high temperatures at various stages during the production of dog and cat foods. It is predicted that acrylamide (AA) can be formed as a result of a possible Maillard reaction since the cat and dog foods contain starch in its structure. The aim of this study is to determine the AA residue immuno-enzymatically in dry cat and dog feeds from different companies available …

Toxoplasma gondii is known to develop extracellular traps from neutrophils in some animal species such as mice, cattle, sheep, cats, and donkeys. This study aimed to investigate the extracellular trap structures formed in dog polymorphonuclear leukocytes (PMNs) to T. gondii tachyzoites in vitro. Dog PMN was isolated using Percoll dilutions (45%, 54%, 63%, and 72%). After incubation with tachyzoites, the extracellular traps originating from the dog PMNs were observed in the extracellular areas. Histones (H3), myeloperoxidase (MPO), and neutrophil elastase (NE), the characteristic features of NETosis reaction, were detected in extracellular areas. The tachyzoites were observed between the extracellular trap …

Shotgun metagenomics applied to archaeological feces (paleofeces) can bring new insights into the composition and functions of human and animal gut microbiota from the past. However, paleofeces often undergo physical distortions in archaeological sediments, making their source species difficult to identify on the basis of fecal morphology or microscopic features alone. Here we present a reproducible and scalable pipeline using both host and microbial DNA to infer the host source of fecal material. We apply this pipeline to newly sequenced archaeological specimens and show that we are able to distinguish morphologically similar human and canine paleofeces, as well as non-fecal …

Globally, epilepsy is a common serious brain disorder. In addition to seizure activity, epilepsy is associated with cognitive impairments including static cognitive impairments present at onset, progressive seizure-induced impairments and co-morbid dementia. Epilepsy occurs naturally in domestic dogs but its impact on canine cognition has yet to be studied, despite canine cognitive dysfunction (CCD) recognised as a spontaneous model of dementia. Here we use data from a psychometrically validated tool, the canine cognitive dysfunction rating (CCDR) scale, to compare cognitive dysfunction in dogs diagnosed with idiopathic epilepsy (IE) with controls while accounting for age. An online cross-sectional study resulted in …

Beta-amyloid (Aβ) immunotherapy is a promising intervention to slow Alzheimer’s disease (AD). Aging dogs naturally accumulate Aβ and show cognitive decline. An active vaccine against fibrillar Aβ 1–42 (VAC) in aged beagles resulted in maintenance but not improvement of cognition along with reduced brain Aβ. Behavioral enrichment (ENR) led to cognitive benefits but no reduction in Aβ. We hypothesized cognitive outcomes could be improved by combining VAC with ENR in aged dogs. Aged dogs (11–12 years) were placed into 4 groups: (1) control/control (C/C); (2) control/VAC (C/V); (3) ENR/control (E/C); (4) ENR and VAC (E/V) and treated for 20 months. …

Dogs remain the main non-rodent species in preclinical drug development. Despite the current dearth of new drug approvals and meagre pipelines, this continues, with little supportive evidence of its value or necessity. To estimate the evidential weight provided by canine data to the probability that a new drug may be toxic to humans, we have calculated Likelihood Ratios (LRs) for an extensive dataset of 2,366 drugs with both animal and human data, including tissue-level effects and Medical Dictionary for Regulatory Activities (MedDRA) Level 1–4 biomedical observations. The resulting LRs show that the absence of toxicity in dogs provides virtually no …

Predicting Human Drug Toxicity And Safety Via Animal Tests: Can Any One Species Predict Drug Toxicity In Any Other, And Do Monkeys Help?, Jarrod Bailey, Michelle Thew, Michael Balls
Jarrod Bailey, PhD
Animals are still widely used in drug development and safety tests, despite evidence for their lack of predictive value. In this regard, we recently showed, by producing Likelihood Ratios (LRs) for an extensive data set of over 3,000 drugs with both animal and human data, that the absence of toxicity in animals provides little or virtually no evidential weight that adverse drug reactions will also be absent in humans. While our analyses suggest that the presence of toxicity in one species may sometimes add evidential weight for risk of toxicity in another, the LRs are extremely inconsistent, varying substantially for …

Animal use continues to be central to preclinical drug development, in spite of a lack of its demonstrable validity. The current nadir of new drug approvals and the drying-up of pipelines may be a direct consequence of this. To estimate the evidential weight given by animal data to the probability that a new drug may be toxic to humans, we have calculated Likelihood Ratios (LRs) for an extensive data set of 2,366 drugs, for which both animal and human data are available, including tissue-level effects and MedDRA Level 1–4 biomedical observations. This was done for three preclinical species (rat, mouse …
