The large T antigen encoded by simian virus 40 (SV40) plays essential roles in the infection of permissive cells, leading to production of progeny virions, and in the infection of nonpermissive cells, leading to malignant transformation. Primary mouse embryo fibroblasts (MEFs) are nonpermissive for SV40, and infection by wild-type SV40 leads to immortalization and transformation of a small percentage of infected cells. We examined the ability of an extensive set of mutants whose lesions affect SV40 large T antigen to immortalize MEFs. We found that immortalization activity was retained by all mutants whose lesions are located upstream of codon 346. …

Economic efficiency of beef production to weaning and to slaughter was estimated in three groups, different in milk available (low, medium, and high) to the calves but the same in growth potential. Efficiency was the ratio of income to expenses. Income was derived from cull cows and calves at weaning or carcasses of calves fed to slaughter. Feed and non-feed expenses for the cowherd and for calves to weaning or to slaughter were included. Efficiencies were estimated assuming observed reproductive rates and energy requirements for maintenance, as well as for equal reproductive rates and equal requirements for maintenance in the …

Performance testing of beef bulls was initiated to improve productivity of Arkansas beef cattle through breeding. This cooperative testing program provides cattlemen with reliable information for identifying animals with superior breeding value for the traits of production measured in the test. By using the information as a basis for herd sire selection, a breeder can greatly increase the chances of obtaining a bull that will sire rapid-gaining, more efficient, highquality calves. Such calves can increase profits for both the breeder and the feeder. This testing program provides information that is useful to financial institutes in arranging their lending programs and …

Nearly 2,400 plant species have been documented ethnographically as having medicinal value among Native Americans. Therefore, it is not surprising that evidence of medicinal plants appears in some of the over 1,000 coprolites analyzed from the southwestern U.S. Three medicinal species identified in pollen analysis of coprolites are discussed. Willow (Salix) is the most common analgesic in the Native American pharmacopoeia. Prehistoric use of this plant is documented in coprolites from Bighorn Cave in the Black Mountains of Arizona and also in a burial from the Mimbres Valley, New Mexico. Perforin lyses cells by binding to the target cell membrane, where it polymerizes into large nonspecific pores. It is shown here that the first 34 amino acids of the N-terminal region of either human or murine perforin are soluble in aqueous medium and spontaneously insert into membranes. The N-terminal peptides lyse liposomes and nucleated cells, and they form ion channels in planar bilayers, some of which are comparable to those previously described for perforin. The lytic activity of the N-terminal domains does not require calcium, is independent of the lipid headgroup composition, and can be inhibited by heparin. Tumor cells …

T antigen is able to transactivate gene expression from the simian virus 40 (SV40) late promoter and from several other viral and cellular promoters. Neither the mechanisms of transactivation by T antigen nor the regions of T antigen required for this activity have been determined. To address the latter point, we have measured the ability of a set of SV40 large T antigen mutants to stimulate gene expression in CV-1 monkey kidney cells from the SV40 late promoter and Rous sarcoma virus (RSV) long terminal repeat (LTR) promoter. [3H]-methotrexate binding at pH 5.0 and pH 7.2 by the cytosol of tumor tissues and the surrounding normal areas of the gastrointestinal tract of patients suffering from colon or gastric cancer has been used to identify in these cells the presence of a binder of methotrexate having low-affinity for this drug in addition to the enzyme dihydrofolate reductase. Scatchard analysis of the [3H]-methotrexate binding data by a colon tumor sample also reveals that there are two binders of this drug present in the cytosol of these cells. Monoclonal antibody SF8/5E11, which recognizes the transmembrane protein (TMP) of simian immunodeficiency virus of macaque monkeys (SIVmac), displayed strict strain specificity. It reacted with cloned and uncloned SIVmac251 but not with cloned SIVmac142 and SIVmac239 on immunoblots. This monoclonal antibody neutralized infection by cloned, cell-free SIVmac251 and inhibited formation of syncytia by cloned SIVmac251-infected cells; these activities were specific to cloned SIVmac251 and did not occur with the other viruses. Site-specific mutagenesis was used to show that TMP amino acids 106 to 110 (Asp-Trp-Asn-Asn-Asp) determined the strain specificity of the monoclonal antibody. Genetic and antigenic variation may be one means by which lentiviruses that cause AIDS avoid elimination by host immune responses. Genetic variation in the envelope gene (env) was studied by comparing the nucleotide sequences of 27 clones obtained from two rhesus monkeys infected with molecularly cloned simian immunodeficiency virus. All 27 clones differed from each other and differed from the input clone in the gp120 (SU) portion of the envelope gene. Nucleotide substitutions were shown to accumulate with time at an average rate of 8.5 per 1,000 per year in SU. Surprisingly, the majority of nucleotide substitutions (81%) resulted in …

An alternative approach to structurefunction analysis of vesicular stomatitis virus (VSV) gene products and their interactions with one another during each phase of the viral life cycle is described. We showed previously by using the vaccinia viruslT7 RNA polymerase expression system that when cells expressing the nucleocapsid protein (N), the phosphoprotein (NS), and the large polymerase protein (L) of VSV were superinfected with defective interfering (DI) particles, rapid and efflicient replication and amplification of DI particle RNA occurred. Interieukin-1, a mediator of inflammation, or tumor promoting phorbol esters induce transcription of stromelysin, a metalloproteinase that degrades extracellular matrix molecules and that is overexpressed in diseases such as rheumatoid arthritis. Sequences required for induction of transcription of the human stromelysin promoter are contained on a 46 base pair fragment. This fragment contains a sequence with a high degree of similarity to the binding site for the transcription factor activator protein-1 (AP-1) and indeed, the AP-1 sequence of this fragment is necessary but not sufficient for the maximal response to phorbol 12-myristate 13-acetate (phorbol) or interleukin-1. Viruses with defective genomes have been identified in association with virtually every major family of viruses and have been widely utilized as tools for investigating virus functions in animal cell culture systems (Perrault, 1981). It is generally thought that defective interfering viruses (DIs) arise through deletion, rearrangement, or recombination of a competent viral genome. DIs tack the ability for independent existence relying on their parental helper viruses to supply factors required for replication, maturation, and/or encapsidation (Huang and Baltimore, 1977). It is correctly asserted that the intensity of the current debate over the use of animals in biomedical research is unprecedented. The extent of expressed animosity and distrust has stunned many researchers. In response, researchers have tended to take a strategic defensive posture, which involves the assertation of several abstract positions that serve to obstruct resolution of the debate. Those abstractions include the notions that the animal protection movement is trivial and purely anti-intellectual in scope, that all science is good (and some especially so), and the belief that an ethical consensus can never really be reached between the parties.

Five monoclonal antibodies specific for bovine respiratory syncytial virus were characterized by Western immunoblotting, radioimmunoprecipitation, and epitope mapping assays. The monoclonal antibodies were found to be specific for the fusion protein, and there were at least two antigen binding sites, one of which was neutralizing.

Currently there is an unprecedented interest in ethological studies of nonhuman animals. Much of this interest is motivated by a desire to learn more about animals themselves. For scientists assuming this stance, a secondary goal is to use this knowledge to assess the place of humans in the natural order of things, stressing continuity or discontinuity depending on one's views. Others, however, study animals primarily to apply this knowledge to human behavior. We argue that behavioral research demands the rigorous application of methods that are minimally harmful to the animals being studied. We argue for a moderate, but rigorous and …

Four coprolites were excavated with Burial 5 at Ventana Cave. a partially mummified five-year-old child. Two coprolites were granular and dark in color and two were fibrous and light in color. The coprolites are remains of the child's intestinal contents and were submitted for dietary and parasitological analysis. No parasites were found. The fibrous coprolites proved to be remains of highly masticated mesquite pods (Prosopis). The granular coprolites consist of seeds of saguaro cactus (Carnegiea gigantea). Pollen analysis reveals two dietary pollen types, both derived from cactus. No evidence of cultivated plants except for a trace amount of …
