Targeting Ribosome Biogenesis For Pancreatic Cancer Treatment, Carlos Perez Iii Jul 2022

Targeting Ribosome Biogenesis For Pancreatic Cancer Treatment, Carlos Perez Iii

Theses and Dissertations

Pancreatic cancer (PanCa) is one of the leading causes of cancer-related death in the United States. Currently, PanCa is one of the recalcitrant cancers that has very limited therapeutic options available for its treatment. The current standard care of PanCa is gemcitabine (GEM) alone or in combination with FOLFIRINOX, nab-paclitaxel, erlotinib, or 5-FU PanCa, which often show poor response. Therefore, new treatment strategies are required for the prevention and treatment of cancer. Ribosome biogenesis process is dysregulated in most of the cancer types of results in production of more ribosomes and synthesis of oncoproteins which lead to the induction, progression, …

Influence Of Paclitaxel Nanomedicine On The Pancreatic Tumor Immune Components, Godwin Peasah-Darkwah Jul 2022

Influence Of Paclitaxel Nanomedicine On The Pancreatic Tumor Immune Components, Godwin Peasah-Darkwah

Theses and Dissertations

Pancreatic cancer (PanCa) is one of the leading causes of cancer-related mortalities in the U.S due to ineffective therapeutic options. Pancreatic tumors are highly desmoplastic and inhibit efficient uptake of therapeutic payloads. Paclitaxel (PTX) has been tested in PanCa therapy with marginally better clinical outcomes, but remain limited by its poor hemocompatibility, biodistribution and intracellular accumulation in tumor cells. Thus, we synthesized a next generation nanoparticle system for PTX to improve its pharmacokinetics and pharmacodynamics (PKPD) in treating PanCa. We also examined ability of the nano formulation to potentiate gemcitabine (GEM) activity in combating chemoresistance in the pancreatic tumor microenvironment …

Characterization Of Biomarkers For Alzheimer’S Disease And Hiv-1 Associated Neurocognitive Disorders, Armando Garces Iii Dec 2021

Characterization Of Biomarkers For Alzheimer’S Disease And Hiv-1 Associated Neurocognitive Disorders, Armando Garces Iii

Theses and Dissertations

Alzheimer’s disease is a neurodegenerative disorder that is characterized by progressive cognitive decline and the accumulation of amyloid beta and neurofibrillary tangles in regions of the brain. These protein deposits are known to generate multiple effects on the brain that lead to neurodegeneration. It has been established that (Human Immunodeficiency Virus) HIV-1 accelerates the aging process of people living with HIV-1. Moreover, there is significant clinical evidence indicating a potential link between the neurodegeneration developed by those with an HIV-1 infection and AD. HIV-1 viral infection causes cognitive impairment known as …

Development Of A Novel Strategy To Improve Checkpoint Immune Response In Pancreatic Cancer, Poornima Devi Shaji Dec 2021

Development Of A Novel Strategy To Improve Checkpoint Immune Response In Pancreatic Cancer, Poornima Devi Shaji

Theses and Dissertations

Pancreatic Cancer is the 3rd lethal causing cancers in United States with a survival rate less than 5-7%. In advanced unresectable pancreatic cancer, treatment options are restrained to surgery because of its extreme aggressiveness. Immunotherapy, one of the current advanced treatments has shown promising response in other cancers. However, this therapy is limited in pancreatic cancer due to desmoplasia and fibrotic tumor microenvironment (TME).

Our superparamagnetic iron oxide nanoparticles (SPIONS) of curcumin (Curcuma longa, principal curcuminoid of turmeric) have potential ability to inhibit desmoplasia and tumor stroma with an increased bioavailability. This would soften up the tumors for …

Relationship Between Cognitive Performance, Physical Activity, And Socio-Demographic/ Individual Characteristics Among Aging Americans, Imtiaz Masfique Dowllah Jul 2021

Relationship Between Cognitive Performance, Physical Activity, And Socio-Demographic/ Individual Characteristics Among Aging Americans, Imtiaz Masfique Dowllah

Theses and Dissertations

Despite the attenuation of association following adjustments for covariates, participants who engaged in 3–6 hr/wk of vigorous- and > 1 hr/wk of moderate-intensity PA scored significantly higher in tests that assessed executive function and processing speed domains of cognition compared to inactive peers (η2 = 0.005 & 0.007 respectively, p < 0.05). Also, after adjustment, the effects of 1–3 hr/wk of vigorous-intensity PA became trivial for the delayed recall memory domain test scores (β = 0.33; 95% CI: –0.01, 0.67; η2 = 0.002; p = 0.56). There was no clear dose-response relationship between the cognitive test scores and weekly moderate-intensity PA. Interestingly, higher handgrip strength and higher late-life body-mass-index were associated with a higher performance across all cognitive domains. Observed associations provide evidence linking habitual PA with superior cognition health among older adults. Furthermore, increased muscle strength and higher late-life adiposity may …

Understanding An Inflammatory Pathway In Diabetic Retinopathy, Reanna Raye Rodriguez May 2021

Understanding An Inflammatory Pathway In Diabetic Retinopathy, Reanna Raye Rodriguez

Theses and Dissertations

Diabetic Retinopathy (DR) is the leading cause of blindness in the U.S. However, not much is known of its molecular pathway and how it attributes to increases in inflammatory response in the eye. One avenue we will investigate is the transforming growth factor beta (TGFB) signaling pathway and its effect of vascular endothelial growth factor (VEGF) secretion and cell viability. VEGF is the hallmark that exacerbates DR progression in prolonged diabetes. Some major concern that have arisen are the underlying effects of oxidants and antioxidants in elevating VEGF secretion in diabetes. In attempt to learn more, we evaluated how an …

Investigating Bmp7 Expression In Glioblastoma Multiforme, Yajaira Janett Macias May 2021

Investigating Bmp7 Expression In Glioblastoma Multiforme, Yajaira Janett Macias

Theses and Dissertations

The PI3K/AKT/mTOR pathway regulates important cell processes such as growth, survival, motility, inflammation, proliferation, and apoptosis. In Glioblastoma multiforme (GBM) the PI3K/AKT/mTOR pathway is aberrant as it is almost always active. This results in the deregulation of downstream molecules and ultimately leads to cancer progression and maintenance in GBM tumors. In this study, we used RNA-sequencing to identify differentially expressed genes (DEGs) in U87MG GBM cells treated with NVP-BEZ235, a dual inhibitory drug targeting PI3K and mTOR. A total of 7,803 differentially expressed genes were identified via RNA-seq. GEPIA2 online tool was used to assess differential gene expression …

Vertical Transmission Of Zika Virus In An Opossum Model, Dionn Carlo Silva May 2021

Vertical Transmission Of Zika Virus In An Opossum Model, Dionn Carlo Silva

Theses and Dissertations

This study will identify Brazilian ZIKV infection in the offsprings of mated, inoculated Monodelphis domestica females. Unlike non-human primates (NHP) and transgenic mice, M. domestica provides an opportunity to study ZIKV infection in a novel animal model possibly useful for in-utero studies. M. domestica has also been used in cancer and dietary research with varying degrees of success. ZIKV infection will be identified using antibody staining and in situ hybridization. The expected result of this study is that ZIKV will be identified in various tissues and reveal that ZIKV is transmitted vertically in this animal model. The significance of this …

The Long-Term Persistence And Tissue Tropism Of Brazilian Zika Virus In Monodelphis Domestica, Marisol Morales Aug 2020

The Long-Term Persistence And Tissue Tropism Of Brazilian Zika Virus In Monodelphis Domestica, Marisol Morales

Theses and Dissertations

Zika virus (ZIKV) rose as a major public health concern due to the congenital abnormalities of infants born to infected mothers during the 2015 Latin American outbreak. While animal models such as transgenic mice and nonhuman primates are used to study ZIKV pathogenesis, they display various limitations. Additionally, the persistence and tissue distribution of long-term ZIKV infection remains unknown. We describe the laboratory opossum, Monodelphis domestica, as a potential new model for ZIKV pathogenesis research to address these issues. We utilized immunohistochemistry, RT-PCR, and ELISA to show that: infant animals inoculated intracerebrally were susceptible to infection, ZIKV infection persisted …

Using Genetic Diversity To Differentiate Mosquito Populations, Rachel Christine Malampy Jul 2020

Using Genetic Diversity To Differentiate Mosquito Populations, Rachel Christine Malampy

Theses and Dissertations

Experiment 1: Little is known about Aedes aegypti females sharing oviposition sites. We examined four different oviposition sites in McAllen, TX. A mitochondrial gene was used to analyze the variation within the oviposition site to understand how many egg-laying females are sharing oviposition sites. These results will aid in the understanding of Aedes. aegypti oviposition behavior and contribute to more effective mosquito control techniques.

Experiment 2: Mosquito flight patterns and movement from habitat to habitat can drastically influence the spread of diseases. The anthropophilic Aedes aegypti mosquito has historically been identified as having a very limited flight range. Studies also …

Vertical Transmission And Venereal Transmission Of Zika Virus In A Population Of South Texas Aedes Aegypti, Jeremy Marshall Jul 2020

Vertical Transmission And Venereal Transmission Of Zika Virus In A Population Of South Texas Aedes Aegypti, Jeremy Marshall

Theses and Dissertations

Zika virus has become a greater global public health concern from the occurrence of outbreaks in 2007 and its expansion to the Americas in 2015. The presence of Aedes aegypti in the Lower Rio Grande Valley and evidence of local transmission of Zika in Brownsville, Texas provides a need to research transmission dynamics of local populations. In these experiments, a locally acquired, laboratory colony of Aedes aegypti mosquitoes were infected with a blood meal featuring a Mexican isolate of Zika virus to display vector competency, vertical transmission, venereal transmission. Here, we infected 39 females, with 34 of them displaying evidence …

Characterization Of Plasmids Harbored By Vibrio Cholerae Isolates Endemic To The Rio Grande Delta, Viviana Trevino Dec 2019

Characterization Of Plasmids Harbored By Vibrio Cholerae Isolates Endemic To The Rio Grande Delta, Viviana Trevino

Theses and Dissertations

Cholera is a diarrheal disease responsible for considerable morbidity and mortality globally. Epidemics along the Rio Grande Delta were reported during the 1850s, and the etiological agent Vibrio cholerae, remains endemic, thus presenting a unique opportunity for a study of historical and biological relevance. V. cholerae strains were isolated from two locations: the Rio Grande River under a bridge linking Brownsville to Matamoros (site 21) and where the river empties into the Gulf of Mexico (site 42). Two strains from site 21, and one from site 42 were selected to isolate, subclone, sequence, assemble and annotate extra-chromosomal elements. Results …

Temperature Influence On Insecticide Resistance In Aedes Aegypti And Aedes Albopictus Mosquitoes From South Texas, Wendy Soledad Salinas Aug 2019

Temperature Influence On Insecticide Resistance In Aedes Aegypti And Aedes Albopictus Mosquitoes From South Texas, Wendy Soledad Salinas

Theses and Dissertations

Aedes aegypti and Aedes albopictus are vectors for several emerging arboviruses including Zika, chikungunya and dengue. Both mosquitoes are found along the Rio Grande River in South Texas, along the border between Mexico and the United States of America. These mosquito species typically reside near human settlements. A preventative measure public health official use to help halt the spread of diseases is by controlling vector population with the use of insecticides, but as temperature vary, and with the constant exposure to commonly used insecticides, mosquitoes from South Texas may be developing a resistance to the insecticides. Resistance to insecticides may …

Survey Assessment Of Community Knowledge And Prevention Of Chagas Disease And Its Vector In Hidalgo And Cameron Counties, Adam Dennis Wiejaczka Aug 2018

Survey Assessment Of Community Knowledge And Prevention Of Chagas Disease And Its Vector In Hidalgo And Cameron Counties, Adam Dennis Wiejaczka

Theses and Dissertations

Chagas disease, caused by the parasite Trypanosoma cruzi, kills thousands of people annually and is the dominant cause of infectious myocarditis worldwide. Kissing bugs are vectors that carry T. cruzi . and are found in the Lower Rio Grande Valley (LRGV). There is no vaccination nor cure for Chagas. The best mechanism of prevention is to control vector infestations. Little research is available regarding the level of understanding local communities have about Chagas.

Our goal was to test the knowledge of several communities in the LRGV on Chagas disease. Our hypothesis states that local communities do not have sufficient …

Elementary Educators' Perspective Of Physical Education And The Physical Education Teacher, Jennifer M. Garcia May 2016

Elementary Educators' Perspective Of Physical Education And The Physical Education Teacher, Jennifer M. Garcia

Theses and Dissertations

Physical education continues to have the reputation it has had for years of being a subject of marginalization and of least importance in the school curriculum. The purpose of this study is to gain a better understanding of the perceptions of educators of physical education and the physical education teacher in the Rio Grande Valley. The researcher attempts to find, understand, and describe the perceptions of the educator’s view of this subject matter. Statistical significance was set at the .05 level for both tests of analyses. The researcher offers the null hypothesis of no difference between educator’s perception of PE …

Activation Of Microglia In The Rotenone Lymnaea Stagnalis Model Of Parkinson's Disease, Xiomara Galvan Dec 2015

Activation Of Microglia In The Rotenone Lymnaea Stagnalis Model Of Parkinson's Disease, Xiomara Galvan

Theses and Dissertations

Parkinson’s disease (PD) is the most common irreversible neurodegenerative movement disorder affecting approximately ten million people worldwide. In the central nervous system (CNS), PD is known by the selective loss of Dopamine (DA) cells of the substantia nigra pars compacta. Although the disease has been very widely studied, the underlying mechanisms are not yet understood. Several hypotheses have arisen as result of injury of the mitochondria, some experiments include microglia activation, microtubule damage, and reactive oxidative species, which all result in the death of DA cells. The theory of microglia activation upon inflammation was explored in this study. Using the …
