The etiology of insulin resistance (IR) development in type 1 diabetes mellitus (T1DM) remains unclear; however, impaired skeletal muscle metabolism may play a role. While IR development has been established in male T1DM rodents, female rodents have yet to be examined in this context. Resistance exercise training (RT) has been shown to improve IR and is associated with a lower risk of hypoglycemia onset in T1DM compared to aerobic exercise. Additionally, the molecular mechanisms mediating RT-induced improvements in insulin sensitivity remain unclear. Regular aerobic exercise promotes physiological cardiac growth, which is an adaptive response thought to enable the heart to meet higher physical demands. Cardiac growth involves coordination of catabolic and anabolic activities to support ATP generation, macromolecule biosynthesis, and myocyte hypertrophy. Although previous studies suggest that exercise-induced reductions in cardiac glycolysis are critical for physiological myocyte hypertrophy, it remains unclear how exercise influences the many interlinked pathways of metabolism that support adaptive remodeling of the heart. Substantial evidence exists regarding how skeletal muscles use energy and how this affects muscular performance. What remains unclear is how characteristics of muscle energetics affect whole-body energetics during daily living, and what effects this may have on mobility. Muscle fatigue is defined as the reduced capacity to produce torque or power in response to contractile activity. Previous work has observed greater fatigue in older than young adults in response to high-velocity contractions, but similar fatigue with age in response to moderate-velocity contractions. The mechanisms for these age-related differences is not clear. One potential explanation is a greater accumulation of putative agents of fatigue (e.g., Pi, H2PO4-, and H+) in older than young muscle during high- but not moderate-velocity contractions. This study examined the effect of an acute session of low-load high-volume resistance training versus a more traditional high-load low-volume session on energy balance (EB). Five recreationally active males (age: 24±3 y; BMI: 25.8±1.5 kgm-2) completed three different sessions: 1) high-load (90% 1RM); 2) low-load (30% 1RM); and 3) CTRL (no exercise). Gas exchange (V̇O2), blood lactate, and subjective appetite perceptions were measured before each session, as well as at 0, 1, and 2 h post-exercise. A single high-fat meal (HFM) can increase systemic inflammation (postprandial inflammation; PPI) and may be attenuated by recent exercise. However, the effect of preprandial exercise on PPI in older adults (OA) is not clear. Purpose: To determine if preprandial exercise attenuates PPI in YA and OA. Methods: 12 YA (23.3±3.9 years n= 5 M/7 F) and 12 OA (67.7±6 years, n= 8 M/4 F) completed two HFM challenges in a randomized order. Introduction: Investigations of lipid metabolism commonly utilize an oral fat tolerance test (OFTT) to elicit a dramatic increase in serum triglycerides (TG). Although this is common practice there does not currently exist an agreed upon formula for an OFTT. Nor has there been in investigation as to the reliability among commonly used OFTT. The purpose of this study was to measure hormonal and metabolic markers in humans to see if simulating dehydration would emulate measures that are seen in prediabetic humans. 60 volunteers were divided into equal categories by sex and body mass index of normal or overweight. Participants completed two oral glucose tolerance tests (OGTT) after being intravenously infused with a isotonic or hypertonic saline solution for 120 minutes in a counter balanced order. Metabolic inflexibility is an emerging physiological marker indicative of metabolic dysfunction and associated with type 2 diabetes (T2D) and obesity. Exercise is a potent stimulus to improve metabolic health, however, not much is known about the acute effects of exercise on metabolic flexibility (METFLEX). Previous research has shown an inverse relationship between age and the relative intensity at which ventilatory threshold (VT) occurs in adolescent boys. However, no study has examined the effect of maturity status on VT in the differences in boys from the onset of puberty, adolescents. Introduction: Eighty percent restriction of normal dietary methionine (MR) intake has been shown to increase energy expenditure and attenuate the rate of adiposity gain in rodents, despite a paradoxical increase in energy intake. Energy expenditure in rats was shown to increase, even though physical activity level stays the same. This observation suggests that metabolic mechanisms account for the majority of increased energy expenditure measured in methionine restricted animals. At any given running speed, weight support with a lower body positive pressure (LBPP) device (i.e. Alter-G treadmill) reduces V̇O2. However, it is unknown how application of LBPP during running impacts lactate metabolism, specifically lactate threshold. Purpose: To determine if body weight unloading with the Alter-G treadmill alters lactate threshold. Methods. Maximal aerobic capacity (V̇O2max) and lactate threshold (LT) was determined in 8 male subjects on an Alter-G treadmill at 100% and 80% body weight loading at 0% grade in a randomized crossover design. INTRO: High fat diets (HFD) ingested over long periods of time have been shown to cause diet induced obesity leading to excessive adiposity and chronic low grade inflammation. Here we have shown how differing levels of saturated fats (SF), which are common storage fats, affect markers of inflammation and mitochondrial biogenesis. IL-1â and TNF-alpha are pro-inflammatory cytokines that have been shown to increase with a HFD, while molecular signalers, PGC-1alpha, cyt c, and SIRT1, related to mitochondrial biogenesis have been shown to decrease. Recently, whole-body vibration (WBV) exercise has been gaining interest in the health/fitness community for its reported beneficial outcomes. However, most of these have not been evaluated scientifically leading to some reservation in promoting this new exercise modality. The purpose of this dissertation was to assess the viability of WBV exercise to enhance several selected indices of health.

One possible explanation could be free radical production varies among individuals and this variation determines the aging process and the progression of disease. The purpose of this study was to test whether nonagenarians have a relatively low metabolic rate when compared to younger individuals and whether this low metabolic rate is associated with lower levels of oxidative stress and less incidence of disease.
