The Biobehavioral Effects Of Embryonic Exposure To Neural Inflammation And Oxidative Stress In Zebrafish, Dalton J. Anderson

The purpose of this research is to improve understanding of the neurodevelopmental effects of embryonic exposure to elevated inflammation and oxidative stress induced by the antipyretic drug acetaminophen (APAP). Our study was the first to examine the interactive effects of APAP and inflammation in zebrafish embryos and how the treatments affect brain development and larval behavior. Experimental groups of zebrafish larvae were exposed to lipopolysaccharide (LPS) to induce inflammation, APAP, or LPS + APAP and larval behavior was analyzed using Ethovision automated behavioral tracking software. The retina is made up of many different essential parts that work together to allow for vision of many different species. Some of these crucial cells and proteins include ganglion cells, bipolar cells, photoreceptors, synaptic markers, such as VGLUT, and receptors that bind glutamate. The basis of this project is to use immunohistochemistry to identify these important aspects of the retina in zebrafish via monoclonal and polyclonal antibodies. The goal being to compare specific cells in a control group retina to an experimental group of zebrafish.

Hundreds of millions of people are affected by diabetes worldwide. Whether they are diagnosed with prediabetes or Type I or II diabetes, there are a variety of mechanisms in the pathogenesis of diabetes. Diabetes is a disease which consists of recurring states of hyperglycemia that can be difficult to manage due to either lack of insulin production or improper utilization of insulin. While these mechanisms of action differ, complications induced by diabetes occur in both poorly regulated Type I and II. I tested the hypothesis that recombinant leptin protein can be introduced to zebrafish in vivo through non-invasive soaking in a solution containing the protein. One way to study various molecules’ effects in vivo is through intraperitoneal or intracerebroventricular injections during the embryonic or larval stage, which is invasive, difficult to administer, and can have a high mortality rate. 48 hours post fertilization (hpf) zebrafish were soaked in a His-tagged recombinant leptin protein solution at 10 nM and 100 nM concentrations (produced by Genscript). After soaking, zebrafish larvae were washed extensively to remove all recombinant protein on their exterior before homogenization. …

With an increase in popularity among young adults using vaping devices (electronic cigarettes) instead of cigarettes, the effects of these vapors combined with other substances, such as oral contraceptives, is not widely studied. Women who smoke traditional cigarettes while taking oral contraceptives have a greater risk for cardiovascular disease, stroke, and thromboembolism. Due to similarities between cigarettes and e-cigarettes, further studies need to examine the effects of e-cigarettes while taking oral contraceptives. Hyperekplexia is a neurological disorder affecting infants and can be characterized by excessive startle reactions in response to unexpected stimuli. This disease can be modeled in zebrafish in the bandoneon (beo) mutant, where the glrbbmutation results in glycine receptor (GLYR) loss of function glycine receptors and ultimately, improper inhibitory signaling. Though hyperekplexia is a glycinergic-based signaling disorder, disease symptoms are often alleviated with GABA-agonistic benzodiazepines. Hyperalgesia is the increased sensitivity to pain which can present itself to many people no matter age or ethnicity. Having the ability to alleviate pain whether it be acute or chronic pain without the use of addictive medication can have the potential to change the course or modern medicine known today. Through the use of zebrafish, this thesis provides a preliminary understanding in pain signal blocking. Amphetamines are frequently prescribed to young children for attention deficit hyperactivity disorder (ADHD). Due to the recent rise in ADHD diagnoses and the resulting increase in amphetamine intake in children, we investigated how amphetamine exposure impacts the development of stereotyped behaviors and the neural circuit that governs these behaviors. The zebrafish (Danio rerio) was used as our model organism because of the extensively studied developmental milestones these fish provide.
