Callisaurus draconoides, also known as the Zebra-Tailed lizard, belongs to the family Phrynosomatidae family (Pianka, et al. 1972). C. draconoides is a widespread desert lizard found western North America. In California, this species can be found in the Mojave and Colorado Deserts. There are currently several populations that reside in the San Bernardino basin on the cismontane side of the Transverse and Peninsular ranges. These mountain ranges have the potential to have isolated the cismontane populations from their typical desert ranges. In addition, geological passes have the potential to serve as migration corridor between the Deserts and cismontane regions. The …

Transcriptomics is a modern technique in genomics that utilizes RNA sequences to get a snapshot of genetic expression. This is a powerful tool in non-model species lacking a reference genome. Thus, the application of comparative transcriptomics has the potential to help us elucidate the evolutionary mechanisms that facilitate species invasion. Carpobrotus edulis is a prolific and widespread invasive succulent plant belonging to the Aizoaceae family. A native to South Africa, this species has become a dominant invader of many Mediterranean coastal areas. One of the current major issues in the control of vector-borne disease is resistance to insecticides. Resistance to one or more insecticides has appeared in over 580 different arthropod species. Widespread resistance has led to the field of Insecticide Resistance Management (IRM). The goal of IRM is to develop insecticide-use strategies that prevent or slow down the evolution of resistance. Computer models have shown that more effective IRM strategies rely on the assumption that resistance carries a fitness cost in the absence of insecticide use.

Approximately 10% of all bacterial genomes sequenced thus far contain a secondary replicon. This considerable genetic reservoir contains many potentially mobilizable elements, allowing for the formation of many unique secondary replicons. This property of bacterial populations vastly increases the genomic diversity available to species that effectively take up and maintain these replicons. Members of the genus Variovorax have extensive heterogeneity in genome architecture, including sequenced isolates containing plasmids, megaplasmids, and chromids. Using available Illumina data on the NCBI database, we have completed these assemblies using 3rd generation sequencing methods on 17 members of this genus. We have sequenced, assembled, …

Studies on population genetics examine the relationship and effects of population structure, migration, gene flow and demographic history, and are therefore important in the conservation of endangered species. Astragalus jaegerianus, a critically federally endangered species found in a geographically restricted range is investigated to determine population structure and genetic variation. Previous research on A. jaegerianus focused on DNA sequence data for cpDNA and nrDNA showed no variation. Further research on A. jaegerianus utilizing AFLP’s on the whole genome indicated substantial gene diversity and population structure consistent with geographically widespread species. Atribacteria (OP9), candidate phylum with no representatives in pure culture, is found in various anaerobic environments worldwide. “Caldatribacterium”, a lineage within Atribacteria that is predicted to be a strictly anaerobic sugar fermenter based on cultivation-independent genomic analyses, is currently being maintained in lab enrichment cultures with fucose as its sole growth substrate. Metagenomics and 16S rRNA gene tag sequencing indicated that the fucose culture was a co-culture of “Caldatribacterium” and an uncultivated member of the genus Thermodesulfobacterium. Due to failed attempts to isolate “Caldatribacterium” by dilution-to-extinction and plating, it was hypothesized that “Caldatribacterium” is dependent in some way on the …

‘Aigarchaeota’, a deeply branching lineage in the domain Archaea with no cultivated representatives, includes both thermophilic and hyperthermophilic microorganisms that reside in terrestrial and marine geothermal environments. The ‘Aigarchaeota’ consists of at least nine proposed genus-level groups that have been confirmed via 16S rRNA sequencing, with ‘Aigarchaeota’ Group 1 (AigG1) being the focus of this study. Based on cultivation-independent genomic data available from several AigG1 members in Great Boiling Spring (GBS), NV, and Yellowstone National Park, 22 different types of growth media were designed and tested for their ability to support growth of AigG1. Rhinichthys osculus (Cyprinidae), the speckled dace, is the most widely distributed freshwater fish in the western United States. The southern California populations of R. osculus are identified as the Santa Ana speckled dace (SASD), though the SASD has not yet been formally recognized as a distinct taxon. Current mtDNA analysis performed in the Metcalf Lab has shown a reciprocally monophyletic relationship among three California regions; southern, central coast, and Owens Valley. Similarly, microsatellite genotyping has shown significant levels of geographic population structure. “Captorhinikos” chozaensis is a multiple-tooth-rowed captorhinid reptile from the Lower Permian Clear Fork Group, undivided formation. Upon re-examination of the materials associated with the species from both the Chicago Field Museum of Natural History, and the Smithsonian United States National Museum, I reaffirm their affinity and collective identity as a valid taxon. “Captorhinikos” chozaensis does not, however, belong with either of the two members of its genus, C. valensis or “C.” parvus, instead occupying its own branch on the phylogenetic tree of the Captorhinidae. The purpose of this genetic study of the Santa Ana Speckled Dace Rhinichthys osculus was three-fold. The first goal was to characterize the molecular structure of the mtDNA control region of R. osculus. An 1143 base-pair region of the mitochondrial DNA genome, which included the complete control region was sequenced for all individuals. Analysis of the sequence data revealed that the molecular structure of the speckled dace control region was similar to the molecular structure described for other vertebrate taxa. In this study, I sequenced the mitochondrial cytochrome b gene to elucidate the extent and pattern of genetic variations among and within populations of Rhinichthys osculus (Santa Ana Speckled Dace) found in the different watersheds in Southern California, Central California Coast and Eastern California Desert. I described and analyzed the structural characteristics and pattern of base sequence substitutions in the cytochrome b gene to understand the molecular evolution of the gene. Rhinichthys osculus, the Speckled Dace, is one of the most ubiquitous fish in western North America. Within the Southern California region, the local taxon is known as the Santa Ana Speckled Dace. The purpose of this study was to characterize and identify polymorphic microsatellite markers for R. osculus in which twenty-three were identified through Illumina pair-end sequencing. A site within San Timoteo Canyon was revisited, 13-14 years after treatment, to look at long-term effects of Arundo donax removal. The data obtained were used to determine whether A. donax had re-invaded, other non-native species had established the area, or if native species were able to grow in place of the removed A. donax. The previous removals included a combination of grinding large patches of A. donax and then foliar spraying, foliar spraying of uncut plants, and direct spraying of hand cut stems, depending upon the location and size of the plant. This study investigated responsibility attributions for a partner's emotional infidelity and for a partner's sexual infidelity, and the likelyhood that the victim or partner would end the relationship. This study found a significant relationship between responsibility attributions for a romantic partner's unfaithfulness and the likelihood the relationship would end: the stronger the attributions of personal responsibility the more likely the relationship would dissolve.

Basal synapsids comprise one of the best represented groups of early tetrapods and demonstrate a classic example of a macroevolutionary transition. Differences in osmoregulatory behaviors were studied in two stream-dwelling tree frogs (Pseudacris cadaverina and P. regilla) with different natural histories. This study supports the idea that the natural history of a species has a strong effect on behavior associated with osmoregulation.

This study lends strong support to the idea that members of Bromeliaceae have undergone a recent adaptive radiation, and therefore show that, at least in part, diversity in the tropics is due to a fast speciation rate and that the tropics can be a "cradle" for new diversification and exploitation of varying ecological niches through the diversification of ecophysiological traits within a lineage.

Uses mitochondrial DNA sequence data to examine the degree of genetic differentiation among sky island populations of two subspecies of pygmy nuthatch (Sitta pygmaea melanotis and Sitta pygmaea leuconucha) occurring in Southern California. Assesses the hypothesis that sky islands are genetically isolated and that migration (gene flow) is restricted among them. Eleven sampling locations throughout Southern California were selected based on representation of subspecies, availability of specimens, and feasibility of collection. Results indicate that pygmy nuthatch populations fragmented in the disjunct mountain ranges of Southern California exhibit low but significant levels of genetic differentiation.

The purpose of this study is to investigate the level and distribution of genetic variation in Astragalus jaegerianus by using molecular markers. The objectives of the study are: to estimate levels of genetic variation within and among populations of Astragalus jaegerianus; to test the hypothesis that levels and patterns of genetic variation in species of restricted ranges and few individuals is low and partitioned at the population level; and to discern whether, or how well, genetic partitioning of Lane Mountain milk vetch correlates with its geographic partitioning in the field.

The Diadectomorpha is a central taxon in understanding the origin and early evolution of amniotes. It is considered a sister taxon to Amniota and is so similar to amniotes that some researchers have placed it within the Amniota itself. This group is composed of three families: the Limnoscelidae, the Tseajaiidae, and the Diadectidae. Being the most basal member of this group, the family Limnoscelidae is especially important in these studies.

The thermal environment exerts a strong influence on many aspectsof squamate life history. Environmental temperatures constrain daily activity time, fecundity, growth rate, and also influences hatchling phenotype. Two populations of the Big Red Skimmer Dragonfly, Belonia saturata (Odonata: Amisoptera), were studied to determine the thoracic temperatures maintained during flight, the presence of polymorphic enzyme systems, and to determine the differences in activity of glycerol-3-phosphate dehydrogenase between a desert and non-desert population.
