The purpose of this study was to determine the associations between isokinetic leg muscular strength and cardiovascular disease (CVD) risk factor characterizations in Americans aged 50 and older. Using a publicly available dataset from the National Health and Nutrition Examination Survey (NHANES), a secondary analysis was conducted on participants (males ≥50 yrs; females ≥55 yrs; N=10,858) pooled from 1999 to 2002. CVD risk factors were determined using the American College of Sports Medicine (ACSM) cutoff values, with all nine ACSM risk factors analyzed. Mycobacteriosis is a fatal disease in fishes caused by acid-fast bacteria in the genus Mycobacterium. This disease impacts aquaculture, aquariums, and wild fishes. Unfortunately, there are currently no non-lethal diagnostic tests for mycobacterial infection in fishes. Type IV delayed type hypersensitivity (DTH) responses in vivo and in vitro are currently used for the non-lethal detection of mycobacterial infections in humans and in animals; however, there is little information available on DTH responses in fishes. Malaria is among the most devastating human diseases, and a majority of lethal cases are caused by the protozoan parasite, Plasmodiumfalciparum. The emergence of multi-drug resistant P.falciparum is a major obstacle to malaria control and is highlighted by the abandonment of chloroquine (CQ) as a first-line treatment of P.falciparum infections worldwide. Chloroquine resistance (CQR) is associated primarily with mutations in the transmembrane digestive vacuole protein, PfCRT. However, CQR P.falciparum parasites harboring the same mutant pfcrt allele vary in their CQ response, suggesting the CQ response is multigenic in nature. Cartilage deformities within the human chest wall, specifically pectus excavatum (PE) and pectus carinatum (PC) are common (1/400-1/1000) and yet, despite their clinical significance, are some of the least studied disorders pertaining to cartilage [1]. The costal cartilage connecting "false ribs" 8-10 to the sternum is often abnormally grown and can lead to formation of a severely sunken "funnel" chest (PE) or push outwards to form a "pigeon" chest (PC). Costal cartilage is a type of hyaline cartilage that forms rod-like structures that connect the ribs to the sternum. Deformation of costal cartilage is observed in the chest wall deformities, pectus excavatum and pectus carinatum. Pectus excavatum involves a sternal displacement causing a depression of the chest while pectus carinatum causes a protrusion of the chest. As costal cartilage is not a widely studied tissue, this leaves little knowledge into possible factors involved in the pathogenesis of pectus deformities. Human endogenous retroviruses make up approximately 8-9% of the human genome. A number of expressed HERVs, those that are actively transcribing, have been associated with various cancers. Suppression mechanisms that control HERV expression often fail or become more permissive in tissues where expression should be restricted. Previous studies have identified HERV expression in breast cancer tissues, whereas normal tissue HERV expression remained suppressed. In addition, studies of DNA hypermethylation have correlated with the ability to contribute to cancer development. Hypertension is one of the common complications of obesity. Using a rat model of diet induced obesity and hypertension we investigated some of the mechanisms that are involved in regulation of blood pressure in obesity. The first aim of this study was to determine the role of proximal tubule transporters on the renal sodium handling in obese hypertensive (OP) and lean normotensive (OR) rats. Research has also shown that there are many misconceptions among youth sports coaches regarding concussions. The Durham Collection and the Cambridge and Oxford University Museums provided the materials to investigate the possibility that the extinction of indigenous rats of Christmas Island was a result of disease introduced by infected ship rats (R. rattus) in 1899. The collections of H.E. Durham in 1901-1902 reveal that R. macleari was present on Christmas Island up to then and includes specimens of R. rattus together with specimens that exhibit characteristics of both R. rattus and R. macleari. Vibrio cholerae serotypes 01 and 0139 are responsible for world-wide epidemics of cholera. These pandemic causing strains must possess genes that encode for the cholera toxin (CTX) and toxin co-regulated pili (TCP) in order to infect their hosts. In this study, 284 isolates of Vibrio cholerae from ballast and coastal waters were serotyped, with 11 % testing positive for serotype 01 and 21 % testing positive for serotype 0139. PCR assays were used to detect the presence of ctxA and tcpA genes in all positive isolates, none of which contained ctxA while 2% of the isolates contained tcpA. The 01 …

Diabetes mellitus is a group of conditions characterized by hyperglycemia due to an inability to produce or properly utilize insulin. The majority of cases fall into two categories, Type I and Type 2. Type I results from the autoimmune destruction of pancreatic β-cells of the islets. The beta cells are the exclusive source of insulin and the patient becomes entirely dependent on exogenous insulin to survive. Exercise-induced asthma (EIA) is a common disorder that occurs in individuals who compete in recreational activities. The purpose of this study was to determine the knowledge of EIA for high school athletic coaches. A 22 question demographic instrument was used for background information, and an 18 question multiple-choice assessment was used to assess knowledge of prevention, recognition and management of EIA. The purpose of this study was to examine the short-term effects of Grades 1 and 2 posteroanterior joint mobilizations (JM) at the pathological lumbar spine on subject pain, range of motion (ROM), and muscle force. All subjects were determined to have sustained mechanical dysfunction of the lumbar spine of less than 48 hours as assessed through a standardized evaluation. Subjects were randomly assigned to a group; the control group received ice and stretching while the experimental group received JM in addition to ice and stretching. In 1999, I discovered the first virus known to be pathogenic to any species of lobster. HLV-PA is a pathogenic herpes-like virus that infects juvenile Caribbean spiny lobster, Panulirus argus, in the waters off south Florida (USA), and it alters the behavior and ecology of this species in fundamental ways. Gross signs of HLV-PA infection are lethargy, morbidity, cessation of molting, and discolored, “milky” hemolymph that does not clot. HLV-PA infects the hemocytes of host lobsters, specifically the hyalinocytes and semi-granulocytes, but not the granulocytes. Human T-cell leukemia virus type 1 (HTL V-1) is dependent upon the enzymatic activity of its protease for maturation. Maturation of the protease is facilitated by cleavage of specific amino acid residues, followed by dimerization. The effects of the amino acid sequence located N-terminally to the cleavage site on the ability of the protease to become active were the focus of the current study. These amino acid sequences were contributed by the plasmid vector into which the protease gene was inserted.

The most prevalent life-threatening disease worldwide, secondary to coronary artery thrombosis is Acute Myocardial Infarction (AMI). Increases in three clinical markers have been explored as an early means of AMI detection: Cardiac myoglobin, and the two specific cardiac isoforms of Creatine Kinase, CK-MM and CK-MB. However, myoglobin levels have shown to give a more accurate correlation (R = 0.89) than does Creatine Kinase evaluation. Therefore, we have developed a two-site Enzyme-Linked Immunosorbent Assay (ELISA) using monoclonal antibodies against myoglobin isolated from human heart tissue. Recently, it has been reported that human type I iodothyronine deiodinase, an enzyme important in the conversion of T4 to T3 , is a selenoenzyme. Several studies have reported alterations in plasma selenium level in hypo- and hyperthyroid patients.

Using polarized Zeeman-effect atomic absorption spectroscopy, we measured selenium, zinc, copper and manganese concentrations in the plasma and/or red blood cells in children with or without thyroid disease being seen at the outpatient clinic at Children's Hospital of the King's Daughters. Primary gout is uric acid overproduction, caused by excessive synthesis of the purine precursors. The human immunodeficiency virus (HIV) reverse transcriptase (RT) (EC 2.7.7.49) is the central replication enzyme for HIV. In general, the kinetic mechanism for this and all other polymerases involves the ordered binding of two substrates: a primer-template (PT) followed by a deoxyribonucleoside triphosphate (dNTP). Previous investigations prompted this research when it was discovered that the substrate dNTP, in absence of PT, could protect a recombinant c-terminal mutant HIV-1 RT from inhibition by pyridoxal-5'-monophosphate (PLP), an active-site dNTP inhibitor. Drugs such as adrenoceptor antagonists and certain centrally acting agents are known to lower blood pressure and thus serve as clinically useful antihypertensive agents. Norepinephrine 4, a natural ∝-adrenoceptor agonist, is the major agent associated with increased blood pressure in the periphery. Based on a careful structure-activity comparison, a series of substituted oxazolidines, 20, 21, 22 and 23, have been designed as possible antihypertensive candidates. Specifically, 22 and 23 have been synthesized to serve as potential norepinephrine antagonists.

Human serum high density lipoprotein subclasses, HDL2 and HDL3 , were isolated by preparative salt density gradient ultracentrifugation and further analyzed by electrophoresis on a 4 to 15% concentration gradient polyacrylamide gel.

The separation of the major classes of serum lipoproteins was achieved after a single ultracentrifugation for 272,000 g at 15°c in a swinging bucket rotor. Duck plague virus causes a hemorrhagic disease affecting the family Anatidae, resulting in 60-80% mortality in captive flocks. Plaque and neutralization assays for Anatid herpesvirus (AHV) require 5-10 days for completion and are restricted by the seasonal availability of fertile duck eggs. The enzyme linked innnunosorbent assay (ELISA) is rapid and specific and avoids the use of living cells. The Holland strain of AHV was grown on duck embryo fibroblasts and purified by banding on CsCl gradients. The resulting virus was used to raise antisera in prebled New Zealand white male rabbits. Duck plague is a hemorrhagic, necrotic disease found exclusively in the order Anseriformes, family Anatidae (ducks, geese, swans). The etiologic agent has been reported to be a herpes-like virus. The Holland strain of duck plague was found to induce cytopathic effects in cell culture (perinuclear vacuolation followed by lysis). A simple reproducible plaque assay for quantitating the virus was developed. Under conditions of the plaque assay, one-hit kinetics were observed. The host range of duck plague virus in cell culture was found to include the order Galliformes, family Phasianidae (pheasants, quails and partridges).
