The number of tuberculosis (TB) cases worldwide reached 1.5 million in 2018; thus, TB is considered a deadly disease. TB is caused by Mycobacterium tuberculosis and involves lipid synthesis. Considering the importance of lipid metabolism in bacteria, FabH may be an essential protein target for repressing lipid synthesis. Capsaicinoids from Capsicum annuum demonstrate potent antibacterial activity. This study predicted the ability of capsaicinoid compounds to inhibit FabH. In silico analysis was performed by retrieving the structure of FabH from PDB and those of selected capsaicinoid derivatives from PubChem. DNA methylation is an epigenetic modification that transfers a methyl group onto the C-5 position of the cytosine to form 5-methylcytosine. DNA methylation regulates gene expression by recruiting proteins involved in gene repression or by inhibiting the binding of transcription factor(s) to DNA, especially in regulation of Allele Specific Expression (ASE). In this study, we used Oxford Nanopore long-read sequencing technology to profile methylome in the two inbred mouse strains, C57BL/6J (B6) and DBA/2J (D2). Compared with bisulfite conversion followed by Illumina Sequencing, long-read sequencing technology allows us to achieve much longer read length of 4,653.675 base pairs on average …

The shedding propensity of a person can assist data interpretation in casework when assessing the possibility of passive transfer for DNA analysis. Past studies on shedding propensity evaluated palmar skin (washed and unwashed) deposits. This study compared different skin locations with respect to shedding propensity, and explored the potential of tape-lifts as a skin surface collection method. Humpback whales are known for the plastic and variable nature of their acoustic communication, social interactions, and foraging behaviors. Distinct foraging strategies are used to capture different prey types at the surface and on the seafloor. Sounds produced during foraging are thought to facilitate intraspecific social communication. Previous studies have identified specific sound types produced by humpbacks associated exclusively with benthic foraging strategies, suggesting functions related to foraging behavior. Infertility in women is caused by a depletion of eggs below the threshold for functional ovulation. Unlike men where sperm production is maintained throughout their reproductive lifespan, a women's proliferation of eggs ceases prior to birth. Interestingly during development, a fetus has up to 7 million eggs and two-thirds of these eggs die via apoptosis before they are born, further signifying the importance of the quantity control of eggs to maintain fertility. Infection of the middle heart layer, or myocardium, is often caused by a viral agent. Introduction: Mutations in the KCNN2 gene have been linked to phenotypes of movement disorders and autism. The goal of this experiment was to use the CIUSPR-Cas9 system to create a deletion within the KCNN2 gene and to observe the effect on the expression of movement in zebra fish embryos.

The characterization of the most common photosynthetic biofilms in the Nerja Cave by the continuous monitoring of the in vivo chlorophyll a (Chl a) fluorescence and the incorporation of the irradiance as a new environmental variable related to previous studies in the cave, have allowed us to improve our knowledge about the photosynthetic pattern of the biofilms of the cave. It is typically assumed that dinitrogen (N2) fixation and denitrification are mutually exclusive processes in riverine ecosystems because N2 fixation is favored in high light, low nitrogen (N) environments but denitrification is favored under anoxic, high N conditions. Yet recent work in marine and lake ecosystems has demonstrated that N2 fixation can happen under high N conditions and in sediments, challenging this assumption. We conducted a cross-ecoregion study to test the hypothesis that N2 fixation and denitrification would co-occur in streams and rivers across a range of reactive N concentrations. Pancreatic cancer (PDAC) is a deadly disease, exacerbated by obesity, which lacks effective therapeutic interventions. Most PDAC has a limited response to immune- and chemotherapy. Treating PDAC is made additionally challenging by the rapid emergence of muscle wasting and cachexia, which predict poor response to several therapies. We have found that dietary supplementation with β-hydroxy-β-methylbutyrate promotes immunosurveillance in PDAC tumors and protects muscle. This dietary supplement has the potential to be an important adjuvant in PDAC therapy, opening the doors to immunotherapy response.

Streptomycetes are Gram-positive, soil-dwelling bacteria that possess a complex life cycle with the alternation of vegetative mycelium, aerial mycelium, and spores. Streptomyces. coelicolor spore maturation is a complex process that involves spore shape metamorphosis from cylindrical pre-spores into ellipsoid spores, but the details of this process have remained enigmatic. Previously, our lab identified a novel gene ssdA that might play a role in spore shape determination using a transposon-based insertion mutagenesis in S. coelicolor. Pancreatic cancer (PDAC) is an extremely lethal disease with an overall survival rate of 10%. Surgery remains the only potentially curative treatment option, but resections are complicated by infiltrative disease, proximity of critical vasculature, peritumoral inflammation, and dense stroma. Surgeons are limited to tactile and visual cues to differentiate cancerous tissue from normal tissue. Furthermore, translating preoperative images to the intraoperative setting poses additional challenges for tumor detection, and can result in undetected and unresected lesions. Habitat fragmentation is a common result of urbanization and species living in these fragments are at risk of extirpation. We conducted a 12-y (2008–2019) capture-mark-recapture study on snakes living in a 593-m section of Rock Castle Creek flowing through an urban area in central Virginia, USA. Our study site was occupied primarily by Queensnakes (Regina septemvittata). We used data from 168 individual Queensnakes to examine several aspects of their population ecology including survival rates (0.52), reproductive effort (21.3% juvenile to adult ratio), growth rates (68% and 30.6% increase for 1 to 2 y-old and 2 to 3+ y-old snakes, respectively), …

Landfills contain a trove of valuable materials, such as critical, precious, and rare earth metals, that are integral to the United State’s economy and national security. The leachate that filters through landfills picks up these materials, which allows for the possibility of recovery. For this research, samples will be analyzed from landfills throughout the Midwestern United States to provide a baseline on water quality constituents, elements present, and microbial activity. Mixed-conifer forests play an important role in the Southwestern USA’s regional carbon cycle by sequestering and storing large amounts of atmospheric carbon onto the landscape. Despite this, little is known about how large, stand-replacing fire alters ecosystem carbon sink-source dynamics. In this study, we used eddy covariance to quantify how stand-replacing fire altered forest sink strength, and the sensitivity of carbon uptake to soil and atmospheric drought in a mixed-conifer forest located in The Valles Caldera National Preserve, NM. Polyhydroxybutyrate (PHB) is a biodegradable, plastic-like polymer produced by some bacteria and degraded by others. The filamentous bacterium Streptomyces sp. SFB5A degrades PHB for growth using an extracellular PHB depolymerase, encoded by the phaZ gene. A morphological mutant of this bacterium, bld4, cannot form aerial filaments and cannot produce PHB depolymerase, despite having the phaZ gene. The inability to synthesize PHB depolymerase may be due to a mutation in a gene encoding one of its transcriptional regulators. A gene (lrp) coding for a potential transcriptional regulator is located 2,700 base pairs upstream from phaZ. Our goal …

Low acceptance of evolution remains an obstacle to quality biology instruction. We develop and utilize a novel assessment which measures emotional experience in light of real-world evolution education scenarios. We presented 296 science teachers 4 pro-evolution and 8 anti-evolution scenarios and asked them to rate their levels of joy, anger, sadness, fear, disgust, shame, and guilt elicited by that scenario on an ordinal 5-point scale. Dyslipidemia and lipotoxicity are pathologic signatures of the metabolic syndrome and type 2 diabetes. Excess lipid causes cell dysfunction and induces cell death through pleiotropic mechanisms that link to oxidative stress. However, pathways that regulate the response to metabolic stress are not fully understood. To identify novel genes involved in metabolic stress, our group performed an unbiased forward genetic screen for lipotoxicity resistance. My studies focused on characterizing one of the mutant cell lines isolated from this screen, in which promoter trap mutagenesis disrupted one allele of the small nucleolar RNA hosting gene 3 (Snhg3).

SSV1, spindle-shaped virus 1, is an archaeal virus with a unique shape that belongs to the family of fuselloviruses and infects hyperthermophilic archaea that thrive at 80°C and pH 3. SSV1 has two capsid proteins, VP1 and VP3, that presumably produce the lemon-shaped virion structure characteristic of fuselloviruses. The VP1 amino acid sequence is highly conserved among SSVs. Here, we investigate the importance of the proteolytic cleavage site in position E66 in VP1 by changing its glutamic acid to the other 19 amino acids. Herbicide use within conventional agriculture has contributed to greatly increased crop yields since its widespread adoption, but environmental concerns regarding overuse and reliance on selective herbicides continue to mount. Using five fungal species and two crop residues in a factorial design, I created a novel slurry to control weeds through inhibition by the mycelial mat formed after application to soil. I monitored weed stem counts and the strength of the mycelial mat under the treatments. The electron transport system complexes form supercomplexes (SCs) within mitochondrial membranes, perhaps increasing respiratory capacity or reducing reactive oxygen species production. My project aimed to determine the abundance, composition, and stability of SCs in a hibernator. Hibernators have dynamic metabolisms that change greatly during the winter. I isolated mitochondria from rats and thirteen-lined ground squirrels (TLGS) in different hibernation states and measured mitochondrial respiration. I extracted mitochondrial proteins using two detergents of different strengths, and quantified SC abundance using 2-dimensional gel electrophoresis and immunoblotting. Rats had fewer SCs than TLGS.
