Viruses are the most abundant biological entity on Earth and their interactions with microbial communities are recognized to influence microbial ecology and impact biogeochemical cycling in various ecosystems. While the factors that control the distribution of viruses in surface aquatic environments are well-characterized, the abundance and distribution of continental subsurface viruses with respect to microbial abundance and biogeochemical parameters have not yet been established. Isolation And Comparative Genomic Analysis Of Final Third Of Satis Genome, Kelly Hartigan, Nicole Curnutt, Matthew Mcdermut

A highly novel Streptomyces phage, Satis, was isolated from a direct environmental sample collected from outside Danforth House on the Washington University campus. Satis infects bacterial species Streptomyces lividans producing pinpoint, cloudy plaques less than 1mm in diameter. Electron microscope data shows rare atypical physical features. Rather than the common octahedral capsid shape, Satis has a prolate head with visible cross-linked hexagonal protein structure and average measurements of 285 nm by 47 nm with a long, flexible tail measuring 268 nm. Obesity has become a serious health issue in the world (Ogden 2012), and its correlation to the human gut microbiota has been studied extensively since 2007 (Cani 2007). In 2013, researchers identified Enterobacter cloacae B29 from an obese patient’s gut and demonstrated its causal relationship to the host’s obese symptoms( Fei & Zhao 2013). However, there have not been many methods developed to alleviate obesity symptoms through eliminating pathogenic endotoxin-producing bacteria (such as E. cloacae B29), from the host’s gut flora. Background: Most tailed bacteriophages (phages) feature linear dsDNA genomes. Characterizing novel phages requires an understanding of complete genome sequences, including the definition of genome physical ends.

Result: We sequenced 48 Bacillus cereus phage isolates and analyzed Next-generation sequencing (NGS) data to resolve the genome configuration of these novel phages. Most assembled contigs featured reads that mapped to both contig ends and formed circularized contigs. Independent assemblies of 31 nearly identical I48-like Bacillus phage isolates allowed us to observe that the assembly programs tended to produce random cleavage on circularized contigs. Biofilms can be found in any environment within proximity to water and are problematic in an assortment of industries. Numerous efforts have been employed to dislodge biofilms including bacteriophages (viruses that infect bacteria). Phage remediation is a promising solution for combatting biofilms that form on catheters in long term use patients. These biofilms often result in catheter-associated urinary tract infections (CAUTIs) and are the most common type of healthcare related infection reported.
