Epibrassinolide Activates Akt To Trigger Autophagy With Polyamine Metabolism In Sw480 And Dld-1 Colon Cancer Cell Lines, Kaan Adacan, Pinar Obakan Yerli̇kaya

Epibrassinolide (EBR), a plant-derived polyhydroxylated derivative of 5?-cholestane, structurally shows similarities to animal steroid hormones. According to the present study, EBR treatment triggered a significant stress response via activating ER stress, autophagy, and apoptosis in cancer cells. EBR could also increase Akt phosphorylation in vitro. While the activation of Akt resulted in cellular metabolic activation in normal cells to proceed with cell survival, a rapid stress response was induced in cancer cells to reduce survival. Leaf rolling (LR), particularly in crops, is a dehydration avoidance mechanism that results from water loss in plants under drought conditions. Two maize cultivars (Zea mays L.) differing in their tolerance to drought (cultivar Side was selected as stress-tolerant and cultivar Karaçay was selected as stress-sensitive) under PEG-induced drought stress were used in this study to understand the effects of polyamines (PAs) on LR response and the role of the antioxidant system in the delayed LR process. Fission yeast Schizosaccharomyces pombe has a variety of stress-signaling proteins that protect cells against environmental or intracellular stress. These proteins help the cells to respond to stress conditions and regulate intracellular functions such as cell division or gene expression. Polyamines (spermidine, spermine, and putrescine) are known to be important in the regulation of stress response and cell division. In this study, we tried to experimentally characterize novel S. pombe genes that are involved in the polyamine pathway and understand their potential roles. The cyclin-dependent kinase (CDK) inhibitors purvalanol and roscovitine are therapeutic agents that control cell proliferation through regulating cell-cycle machinery. They also affect polyamine (PA) metabolism, which is activated in malignant tissues. Therefore, PA catabolism became a remarkable target in cancer therapies. Induction of the PA catabolic enzyme spermidine/spermine N1-acetyltransferase (SSAT) is under the control of transcription factors such as NF?B and PPARγ. Roscovitine and purvalanol are specific cyclin-dependent kinase (CDK) inhibitors, which induce apoptosis by triggering cell cycle arrest in various cancer cells such as colon, prostate, and breast cancer cells. Although the apoptotic action of roscovitine was clarified at the molecular level, the exact mechanism of purvalanol-induced apoptosis is still under investigation. The mitogen-activated protein kinase (MAPK) signaling cascade is activated by different inducers related to growth, proliferation, differentiation processes, or environmental stress factors. The effects of various concentrations (0.01, 1, 2mM) of spermine (Spm) on germination and some early growth parameters of sunflower ( Helianthus annuus L. cv. Santafe) seeds were investigated in media containing different concentrations of NaCl (50, 100, 200mM). Regardless of its concentrations, application of Spm under non-saline conditions remained ineffective on germination percentage as well as on the early growth parameters studied (length of radicle, fresh and dry weights). The salt, depending on its concentration, inhibited or delayed the germination to a great extent. The growth parameters studied in the early growth period were also adversely affected by salt …

Effects of salt (NaCl) and putrescine (Put.) and various combinations of these (salt+Put.) on germination and some early growth parameters of sunflower ( Helianthus annuus L. var. Santafe) plants, grown under the controlled conditions, were investigated. Salt was used in 3 different con-centrations (50, 100, 200mM) and also, Put. was applied in 3 levels (0.01, 1, 2 mM). The salt treat-ment alone inhibited or delayed the seed germination, depending on concentration.
