Excessive microvascular permeability is a serious complication involved in traumatic injury and inflammatory diseases. Alcohol intoxication can exacerbate the physiological derangements produced by microvascular endothelial barrier dysfunction in such disease conditions. Sphingosine-1-phosphate (S1P) has known endothelial barrier-protective properties, and has been shown to ameliorate microvascular leakage in a model of combined alcohol intoxication and hemorrhagic shock and resuscitation (HSR). However, whether the barrier-protective properties of S1P extend to endothelial cells of the blood-brain barrier (BBB) is unclear. Amphibians are currently the most threatened vertebra taxa on the planet. Hundreds of species are thought to have gone extinct while thousands more have been listed as threatened or endangered over the past few decades. Habitat loss, invasive species, climate change, and disease are all thought to have partially contributed to these declines. Two pathogens in particular, infectious viruses in the genus Ranavirus (simply referred to as ranavirus) and the fungus Batrachochytrium dendrobatidis (Bd), have been associated with global mass mortality events of amphibians. As technologies advance the study of ocean dynamics, new approaches to vexing problems of scale and process are becoming more widely available. Originally conceived as a tool primarily for indexing the abundance of near-bottom fishes, the Camera-based Assessment and Survey System (C-BASS) may also be an effective tool for monitoring benthic invertebrate resources vulnerable to natural and anthropogenic perturbations, and for characterizing the composition of benthic communities to inform spatial management. Fecal pollution of surface water can lead to human health issues because waterborne transmission of pathogens is a common cause of gastroenteritis. Genetic markers developed for human-associated Escherichia coli (H8, H12, H14, and H24) are promising tools for microbial source tracking (MST) of sewage in environmental waterbodies but are untested in the US. H gene marker performance characteristics (specificity, sensitivity, and prevalence) were assessed, with the goal of developing a quantitative PCR assay for the markers with the best performance. Ovarian cancer is a leading cause of death among women diagnosed with cancer. Mortality rate is high because an overwhelming majority of new cases are diagnosed with late-stage disease when the survival statistics are very poor with an overall 5-year survival rate of less than 40%. Despite the large burden of disease, the etiology of ovarian cancer is not well understood. Survival and establishment of mangrove propagules at higher tidal elevations beyond the landward margin of their distribution is a requirement for the continued existence of mangrove populations in response to rising sea-level. Despite the growing body of literature that discusses mangrove recruitment patterns, few studies have empirically examined establishment and post-establishment growth success of propagules at the higher intertidal positions into witch mangrove populations are migrating. Cephalopods are an important component of many marine ecosystems and support large fisheries. Their active lifestyles and complex behaviors are thought to be driven in large part by competition with fishes. Although cephalopods appear to compete successfully with fishes, a number of their important physiological traits are arguably inferior, such as an inefficient mode of locomotion via jet propulsion and a phylogenetically limited means of blood-borne gas transport. In active shallow-water cephalopods, these traits result in an interesting combination of very high oxygen demand and limited oxygen supply. Altered signaling pathways, which are mediated by post-translational modifications and changes in protein expression levels, are key regulators of cancer initiation, progression, and therapeutic escape. Many aspects of cancer progression, including early carcinogenesis and immediate response to drug treatment, are beyond the scope of genetic profiling and non-invasive monitoring techniques. Global protein profiling of cancer cell line models, tumor tissues, and biofluids (e.g. serum or urine) using mass spectrometry-based proteomics produces novel biological insights, which support improved patient outcomes. Plants inherently produce polyphenols (i.e., antioxidants) as a response to reduce oxidative stress caused by abusive environmental pre- and postharvest conditions. These antioxidants, as well as vitamin C, are present in considerable levels in strawberries; however, excessive oxidative stress brought on by improper postharvest handling conditions can reduce the levels of antioxidants in the fruit and shorten the shelf-life of strawberries. Nevertheless, it may be possible to utilize strawberry’s naturally occurring polyphenols to reduce postharvest stress and extend their shelf life. The goal of this study was to identify novel drug repurposing opportunities in cancer by utilizing the off-target profiles of clinically relevant kinase inhibitors. This was based on the observation that the global target profiles of compounds are largely ignored and that many compounds have activity that cannot be explained by their cognate target alone. Additionally, by utilizing clinically relevant compounds, any results would hold a high potential for eventual clinical implementation. Objectives: There are three aims for this thesis: the first is to develop a field and laboratory protocol for the storage and analysis of transforming growth factor–beta 2 (TGF-β2) in human breastmilk; second, to validate this protocol and the immunoassay used to assess this new method; and lastly, to explore the ramifications of biosimilar TGF-β2 across multiple milks on human health, growth, and immunity through the review of laboratory findings and previous literature.

Currently, in the United States, 17 million people suffer from alcoholism, of which 1.4 million people are receiving treatment [1, 2]. The link between ethanol metabolism, reactive oxygen species (ROS) and liver injury in ALD has been well characterized over the last couple decades [3-10]. The boletes are macrofungi which have undergone extensive taxonomic revisions since the advent of molecular tools. To further our understanding of the boletes in peninsular Florida, we sequenced two common Floridian boletes, and analyzed them with molecular phylogenetic tools. Boletus rubricitrinus, a common Florida bolete often found in lawns under Quercus, and likely has a distribution that extends to Texas. Based on ITS and LSU sequences and morphological studies, this species belongs in the genus Pulchroboletus. As the holotype is in poor condition, an epitype is established here. The giant freshwater shrimp, Macrobrachium rosenbergii is a large species of prawn grown extensively in aquaculture settings. A social hierarchy exists within the males of this species, representing three distinct male morphotypes. These male morphotypes differ in their behavior, physiology, and morphology and include the largest blue-clawed males (BC males), moderately- sized orange-clawed males (OC males), and the undifferentiated small-clawed males (SM males). All individuals of this species perform grooming behaviors to rid themselves of body fouling which can impede important functions such as movement, respiration, chemoreception, and reproduction. In normal myelopoiesis, the immature myeloid cells (IMCs) differentiate into macrophages, neutrophils or dendritic cells. However, under tumor burden, these IMCs differentiate into myeloid derived suppressor cells (MDSCs) result in an up-regulation of immune suppressive factors and pro-tumor effect. The studies presented in this dissertation establish the dynamics of Eleven nineteen Lysine-rich leukemia (ELL) family of elongation factors during B cell differentiation and provide a description of ELL3 function in B cells.

The transition from a mature naïve B cells into an activated B cell is dependent on a large increase in transcriptional output, which is followed by focused expression on secreted immunoglobulin upon terminal differentiation into plasma cell.
