In Vitro Evaluation Of The Effects Of Capsaicin On Normal And Cancerouscells Of Human Cartilage, Nagi̇han Helvaci, Sabahatti̇n Cömertpay

Turkish Journal of Biology

Chondrosarcoma is a common form of bone cancer which affects the fibrous connective tissue around a joint. It most commonly develops in legs, arms, shoulder blades, rib cage, and pelvis. Capsaicin is an active bitter compound found in red pepper, the fruit of the species Capsicum annuum, and it has been shown to have a lethal effect on different types of cancer. However, to date, investigation of its effect on human chondrosarcoma cells has remained limited. In the study presented here, we determined IC50 values of capsaicin for chondrosarcoma and chondrocyte cells in both fetal bovine serum (FBS)-containing and FBS-deprived …

Tumor Necrosis Factor-Alpha Induced Caspase-3 Activation-Related Inos Gene Expression In Adp-Activated Platelets, Özge Çevi̇k, Zelal Adigüzel, Ahmet Tarik Baykal, Azi̇ze Şener Jan 2017

Tumor Necrosis Factor-Alpha Induced Caspase-3 Activation-Related Inos Gene Expression In Adp-Activated Platelets, Özge Çevi̇k, Zelal Adigüzel, Ahmet Tarik Baykal, Azi̇ze Şener

Turkish Journal of Biology

Platelets are sensitive cells and are easily activated by different stimulants in the circulation system. It is known that tumor necrosis factor-alpha (TNF-α) is a proinflammatory cytokine and plays a role in inflammation. The role of TNF-α in the apoptotic process in blood platelets is unknown. In order to study the formation of apoptosis in platelets after incubation with TNF-α and/or ADP, several biomarkers were chosen: phosphatidylserine (PS) exposure and P-selectin binding; cGMP, Cyt-c, and Ca2+ levels and NOS activation; and gene and protein expression of caspase-3 and iNOS. Platelets were incubated with 100 pg/mL TNF-α and/or 50 mM iNOS …

The Sca1+ Mesenchymal Stromal Subpopulation Promotesdendritic Cell Commitment In The Niche, Na Li, Dandan Yin, Huijie Zhang, Jinmei Xu, Fan Wen, Zheng Liu, Yaozhen Chen, Ning An, Jiajia Xin, Yazhou Wang, Wen Yin, Xingbin Hu Jan 2017

The Sca1+ Mesenchymal Stromal Subpopulation Promotesdendritic Cell Commitment In The Niche, Na Li, Dandan Yin, Huijie Zhang, Jinmei Xu, Fan Wen, Zheng Liu, Yaozhen Chen, Ning An, Jiajia Xin, Yazhou Wang, Wen Yin, Xingbin Hu

Turkish Journal of Biology

The hematopoietic microenvironment regulates self-renewal and differentiation of hematopoietic stem cells. Mesenchymal stromal cells (MSCs) contribute to the niche and participate in supporting dendritic cell (DC) commitment in vitro and in vivo. However, due to MSCs being heterogenic, it is necessary to understand the function of the MSC subpopulation in modulating the DC commitment. The current study showed that one of the bone-related Sca1+ MSCs enhanced bone marrow cell differentiation into DCs by direct cell-to-cell contact. Furthermore, the expression of STAT3 and STAT5 genes decreased in supernatant cells after Sca1+ MSCs were cocultured with bone marrow cells. In contrast, the …

Melatonin Induces Antiproliferative Activity Through Modulation Of Apoptoticpathway In H-Ras Oncogene Transformed 5rp7 Cells, Ayşe Kaplan, Gülşen Akalin Çi̇ftçi̇, Hati̇ce Mehtap Kutlu Jan 2015

Melatonin Induces Antiproliferative Activity Through Modulation Of Apoptoticpathway In H-Ras Oncogene Transformed 5rp7 Cells, Ayşe Kaplan, Gülşen Akalin Çi̇ftçi̇, Hati̇ce Mehtap Kutlu

Turkish Journal of Biology

In the present study, cytotoxic and apoptotic effects of melatonin were investigated on H-ras oncogene transformed rat embryo fibroblast 5RP7 cells. Melatonin inhibited cell growth and induced apoptosis in 5RP7 cells. We showed inhibition of cell proliferation that was time- and dose-dependent in 5RP7 cells for 24 and 48 h. Melatonin was not toxic in NIH/3T3 primary mouse embryonic fibroblast cells at low doses for 24 and 48 h. The IC50 (380 μM) value of melatonin for 48 h was chosen for advanced assays. The percentages of early/late apoptotic cells to which melatonin (IC50: 380 μM) was administered were increased …

L-Arginine Protects Against Ethylene Glycol-Induced Gastric Mucosal Damage In Rats: Immunohistochemical And Electron Microscopic Study, Samah Kandeel, Thoryaa El_Deeb, Ehsan Salah, Nafisa El-Bakary, Naglaa Sarhan, Mohamed Balaha, Hiroki Sakai, Tokuma Yanai Jan 2013

L-Arginine Protects Against Ethylene Glycol-Induced Gastric Mucosal Damage In Rats: Immunohistochemical And Electron Microscopic Study, Samah Kandeel, Thoryaa El_Deeb, Ehsan Salah, Nafisa El-Bakary, Naglaa Sarhan, Mohamed Balaha, Hiroki Sakai, Tokuma Yanai

Turkish Journal of Biology

Ethylene glycol is an alcohol widely used in industry as an antifreeze. It affects the normal gastric mucosal defense mechanisms, inducing surface ulcerations. L-arginine is a precursor of nitric oxide that exhibits multiple biological properties; among them is the acceleration of wound healing, especially in the stomach. Caspase-3 immunohistochemistry, and semithin and ultrathin section examinations were used to investigate the possible protective effect of L-arginine on ethylene glycol-induced gastric mucosal damage in rats. Three animal groups were used: group A (control group), group B (received 3.3 mL/kg ethylene glycol for 2 weeks), and group C (received 200 mg/kg of L-arginine …
