The Flavoprotein Rcla Is A Hypothiocyanous Acid Reductase, Irina Chapman

Reactive chlorine species (RCS) - hypochlorous acid (HOCl), hypothiocyanous acid (HOSCN), and chloramines - are powerful antimicrobial oxidants generated by the innate immune system. Bacteria developed defense mechanisms against RCS, which are highly toxic for invading pathogens. One such defense system involves RclA, an enzyme from the flavin disulfide reductase family, which is massively upregulated upon exposure of Escherichia coli to RCS. Recent research suggests that RclA functions as a Cu(II) reductase to provide resistance against RCS. However, the exact mechanism used by RclA was subject to debate. Chronically high blood glucose levels lead to many problems, such as insulin resistance, the hallmark of Type II diabetes. Increased flux through the hexosamine biosynthesis pathway is one mechanism by which high glucose as well as glucosamine has been shown to induce insulin resistance. This study tests the effects of glucosamine induced insulin resistance on insulin regulation of the metabolic genes glucose-6-phosphate dehydrogenase (G6PDH) and fatty acid synthase (FAS) as well as insulin responsive proteins tribbles homolog (TRIB3) and sterol regulatory element binding protein (SERBP-1c) 1c.

Fatty acids are utilized for energy storage in the form of triglycerides or neutral fats. One way to regulate fatty acid biosynthesis is to regulate the activity of fatty acid synthase (FAS), the enzyme that catalyzes the reaction. Previous studies have shown that regulation of FAS activity and synthesis is regulated by the hormone triiodothyronine, T3, at the level of transcription. Thus, isolation and characterization of the promoter region o f the FAS gene will enable the identification of the thyroid hormone response elements (TREs). In chick embryo hepatocytes in culture, triiodothyronine (T3) stimulates enzyme activity, mRNA level and transcription rate for both fatty acid synthase (FAS) and malic enzyme (ME). Recent evidence has demonstrated the effects of selenate, molybdate and chromate on various physiological processes. Little information, however, is available on the effects of these metal ions on the T3- regulated gene expression for FAS and ME.
In chick embryo hepatocytes incubated in a chemically defined medium, addition of sodium selenate (20 μM) coincident with T3 almost completely inhibited the T3-induced stimulation of FAS and ME activity and accumulation of their respective mRNA's. Neither …

14C-leucine was incorporated into th e proteins of the mesophile Bacillus licheniformis and the thermophile Bacillus stearothermophilus. Aliquots of the cell suspension were treated with chloramine-T in order to decarboxylate the 14C-leucine that was released as the proteins degraded. The 14CO2 was trapped and counted. This allowed for the determination of the extent of intracellular protein degradation at various tem peratures and at various stages of the growth cun/e. For the mesophile, the extent of degradation increased with temperature from 30° C to 55° C. For the thermophile, no significant amount of degradation occurred until …
