Transcriptomics And Toxcast Data Identify Bioeffects In Zebrafish Embryos Exposed To Chemical Mixtures In An Effluent-Dominated Stream, Emma Meade

Theses and Dissertations

Wastewater treatment plant (WWTP) effluent-dominated streams provide critical habitat for aquatic organisms but also continually expose them to complex mixtures of pharmaceuticals and other contaminants of emerging concern (CECs) that can potentially impair growth, behavior, and reproduction. Limited toxicity data on the adverse biological impacts of in vivo exposure to these mixtures make it difficult to assess risk for aquatic organisms, particularly with respect to pharmaceuticals whose designed bioactivity often extends beyond conventionally monitored biological pathways. To address this knowledge gap, the goal of this thesis was to identify biomarkers of exposure to complex CEC mixtures relevant to specific chemicals …

Transcriptome Approach For Identifying Potential Biomarkers For Endocrine Disruption Due To Crude Oil Exposure Using Killifish (Fundulus Heteroclitus)., Frank J. Zadlock Iv Mar 2017

Transcriptome Approach For Identifying Potential Biomarkers For Endocrine Disruption Due To Crude Oil Exposure Using Killifish (Fundulus Heteroclitus)., Frank J. Zadlock Iv

Seton Hall University Dissertations and Theses (ETDs)

The killifish, Fundulus heteroclitus, is a common fish model in aquatic toxicology. However, little is known in this organism about how endocrine disrupting compounds, (EDCs) including polycyclic aromatic hydrocarbons (PAHs), impact the reproductive status from a molecular standpoint. The objective of this project is to apply high throughput sequencing to F. heteroclitus testes to examine the molecular mechanisms that impair reproduction when exposed to crude oil. First, a crude oil exposure experiment was conducted. Following exposure, semi-quantitative PCR was performed to detect changes in gene expression of the following genes known to respond to EDCs: gonadal aromatase (CYP19a), vitellogenin …

Hepatic Transcriptomic And Metabolic Responses Of Hybrid Striped Bass (Morone Saxatilis × Morone Chrysops) To Acute And Chronic Hypoxic Insult, Benjamin H. Beck, S. Adam Fuller, Chao Li, Bartholomew W. Green, Honggang Zhao, Steven D. Rawles, Carl D. Webster, Eric Peatman

United States Department of Agriculture-Agricultural Research Service / University of Nebraska-Lincoln: Faculty Publications

Striped bass (Morone saxatilis), white bass (Morone chrysops), and their hybrid are an important group of fish prized for recreational angling in the United States, and there and abroad as a high-value farmed fish. Regardless of habitat, it is not uncommon for fish of the genus Morone to encounter and cope with conditions of scarce oxygen availability. Previously, we determined that hybrid striped bass reared under conditions of chronic hypoxia exhibited reduced feed intake, lower lipid and nutrient retention, and poor growth. To better understand the molecular mechanisms governing these phenotypes, in the present study, we …
