A Genetic Evaluation Of The Impacts Of Dams On Sauger Movement Patterns In The Arkansas River, Emily R. Jonagan

Decreased habitat connectivity as a result of damming can lead to genetic isolation in fish communities, especially in highly migratory species. Sauger Sander canadensis is a migratory freshwater species native to the Arkansas River. Sauger are highly sought after by anglers during their annual spawning migration in late winter. In order to investigate the impacts Arkansas River dams on Sauger populations, fin clips were collected in the winters of 2019, 2020, and 2021 below eight dams in the McClellan-Kerr Arkansas River Navigational System (MKARNS). Fin clips were also collected from two reservoirs in Kansas to serve as distinct reference populations. …

Use of a genetic-based analogue of the traditional mark-recapture method (transgenerational genetic mark-recapture, tGMR) is rapidly expanding as a means to estimate total escapement of Pacific salmon. The tGMR approach is similar to the simple Lincoln-Peterson mark-recapture method. In tGMR, adults returning to fresh water to spawn are collected in the first sampling occasion and their juvenile offspring that are migrating out to sea are collected during the second sampling occasion. Recaptures are determined by the number of parent-offspring pairs identified through genetic parentage analysis of the adult and juvenile collections. Advances in molecular technologies have provided conservation biologist with the opportunity to quantify the genetic structure of a population and, in turn develop management guidelines and policies aimed at preserving the genetic diversity of fish stocks challenged by human activities. This thesis examines the status of genetics as applied to the management of freshwater fisheries by state natural resource agencies with a purpose of understanding the keys to a successful genetics program. Pax genes encode transcription factors that are highly evolutionarily conserved and are vital for animal development. Vertebrate Pax group III genes Pax3 and Pax7 are required for proper development of muscular and central nervous systems. In their roles in muscular systems, Pax3 and Pax7 specify myoblasts (muscle progenitor cells). Pax3 is predominantly involved in embryonic myogenesis and specifies embryonic myoblasts. In contrast, Pax7 specifies adult myoblasts (muscle satellite cells), and has been demonstrated to be vital for adult muscle regeneration. Recent evidence also implicates a role for Pax7 in the conversion of certain stem cells to the myogenic lineage. The …
