This work consists of studies on dwarfism in Miniature horses and a study on breakdowns in Thoroughbreds in relation to gene aggrecan. A correction of the description and designation of D3 dwarf variant in aggrecan (ACAN) from the master’s thesis Chondrodysplasia-like dwarfism in the Miniature horse (2013). Commercial sequencing showed previous sequencing reads presented an artifact and not a single base deletion. Analysis showed a single base missense mutation in exon 8 identified as D3* was the actual cause. Multiple alleles of ACAN associated with chondrodysplastic dwarfism in Miniature horses by Eberth et al. (2018), corrected the …

Prey remains have long been used as a mechanism to approach diet analyses. As understanding diet is key to comprehending ecosystem dynamics, prey remains identification requires a unique methodological approach to determine diversity within a sample. With the advancement of technology, molecular protocols designed for species-specific identification have improved to incredible accuracy and precision. Yet, the visual identification method has remained a predominant technique within diet studies. With entry-level observers, we matched visual identifications with molecular-based methods to quantify the accuracy of the visual identification method. To date, most genetic studies on the American alligator (Alligator mississippiensis) have focused on population genetics and multiple paternity. There have not been any studies looking at how relatedness affects alligator behavior and movement. This study focused on three main questions: 1) what is the genetic diversity among alligators in this location? 2) can parentage be accurately defined among parents and offspring? and 3) how does individual relatedness affect their spatial distribution? This dissertation focused on the efficacy and validity of whole genome resequencing (WGR) for fine mapping genetic determinants of particular traits in a given organism. Previously, our research group used WGR to identify haplotype blocks of single nucleotide polymorphisms associated with ascites resistance with some as strong candidates for use in marker-assisted selection (MAS). Chapter 2 discusses the completion of a MAS project through evaluation of ascites incidence as well as production traits of economic value to poultry producers. Decreased habitat connectivity as a result of damming can lead to genetic isolation in fish communities, especially in highly migratory species. Sauger Sander canadensis is a migratory freshwater species native to the Arkansas River. Sauger are highly sought after by anglers during their annual spawning migration in late winter. In order to investigate the impacts Arkansas River dams on Sauger populations, fin clips were collected in the winters of 2019, 2020, and 2021 below eight dams in the McClellan-Kerr Arkansas River Navigational System (MKARNS). Fin clips were also collected from two reservoirs in Kansas to serve as distinct reference populations. …

Water scarcity is a global reality and with the anticipated population growth, freshwater resources will be further strained to meet both human needs and agriculture applications. To ensure a water sustainable and food secure future, all aspects of agriculture must become more efficient. Two strategies were explored. The potential of improving water efficiency in broilers was examined. The first strategy was to develop a more efficient and accurate method for measuring water consumption/inputs in agriculture. A great number of studies have identified strong genetic differences between sympatric anadromous and resident populations of Salmonidae. However, Arctic char (Salvelinus alpinus) migratory phenotypes in the Nettilling Lake and Amadjuak River ecosystem in Nunavut, Canada have not been genetically characterized, and it remains unclear if distinct genotypes and phenotypes associated with migratory life history differences are maintained through reproductive isolation, and they have been assumed to be sympatric populations, or co-occurring populations. Co-occurring Arctic char (n=225) were sampled from eleven sites along the Amadjuak River in 2014 and 2015. Twelve microsatellite loci were used to quantify genetic variation among …

Sexually differentiated behavior has been shown to be affected by both genes and hormones. The discovery of the SRY gene, which codes for the development of testes, lead to the development of the Four Core Genotypes model of mice, and the separation of sex chromosomes and its resulting gonadal hormones. Associations Among Beef Cattle Genotypes, Neospora Caninum Infection, And Reproductive Performance, Ryan James Page

Reproductive performance is crucial for sustained financial success in the beef cattle industry. This dissertation includes a population study that quantified the incidence of Neospora caninum infections in the central region of the United States and tested its relationship with reproductive performance in beef cattle. Trial one of that study concluded that 6.9% of open, replacement heifers (n = 1306) tested seropositive. The second trial in that project found that 9.6% of the breeding age females (n = 500) tested were seropositive for Neospora caninum; and that state in which the cattle lived and age impacted (P < 0.05) infection rate. Breed composition, number of farm dogs on the ranch, and use of total mixed rations were not associated (P > 0.1) with seropositive …

Selection within and among breeds are strategies to mitigate the impact of parasitism given failing chemotherapeutics. While Texels have improved marketability compared to other parasite-resistant breeds, the mechanism by which Texels reduce fecal egg count (FEC) is unclear. The immune response to Haemonchus contortus (Hc) was compared in Texel, parasite-resistant St. Croix, and parasite-susceptible Suffolk sheep. Adult worms exposed to St. Croix- and Texel-derived peripheral blood mononuclear cells and serum in vitro had greater binding around the reproductive structures than Suffolk and worm egg release tended to be affected by breed (P = 0.09). Heat stress (HS) has a negative effect on poultry production sustainability due to its adverse consequence on bird welfare, health, growth, and mortality. Although modern broilers have greater gut mass and higher energy use efficiency than unselected birds, they are more vulnerable to HS that induces “leaky gut syndrome,” or increased intestinal permeability. Foot soundness and udder conformation are essential for longevity in beef cattle. The objective of this study was to investigate the effects of foot angle (FA), claw set (CS), teat size (TS) and udder suspension (US) scores on cowherd performance. Data were analyzed for 1,685 observations on Angus-based cows over a four-year period. At weaning FA, CS, TS, and US were evaluated. Scores are based on nine-point scales. A score of one indicates straight pasterns, divergent toes, enlarged bottle shaped teats, and absence of a median suspensory ligament; nine indicates weak pasterns, curled toes, small symmetrical teats, and a tight …

To prevent abnormal development caused by expressing both X chromosomes, female mammals inactivate one of their X chromosomes using an epigenetic process called dosage compensation. This literature review examines how X chromosome inactivation (XCI) occurs during the formation and development of an embryo. This condensation of DNA is marked by histone tail modifications, DNA methylation, and the arrival of structural proteins resulting in extraordinarily stable heterochromatin. X-inactivation is regulated in cis by the X-inactivation center (Xic) that contains the Xist gene and its antisense gene, Tsix. The effect of both genetic and environmental influences on broiler meat quality were evaluated over three experiments. Selection response was assessed for broiler lines selected for high (HBY4) and low (LBY4) four day percentage breast yield which were formed from a random bred control (RAN). A modern random bred population (MRB) created in 2015 was evaluated for differences in 4 day percentage breast yield and the resulting differences in the incidence and severity of the woody breast and white striping myopathies. Fescue toxicosis in grazing beef cattle is caused by consumption of Ergot alkaloids associated with tall fescue (toxic fescue) and is responsible for substantial economic loss to the U.S. beef industry. Cattle consuming toxic fescue suffer adverse physiological responses, such as: hyperthermia, increased respiration rate (RR), poor reproduction and growth performance. Other adverse responses to fescue toxins include the retention of a winter hair coat and vasoconstriction in the extremities, which can lead to tissue necrosis. In 2010, researchers observed an abrupt decline in nesting numbers of the Kemp’s ridley following 35 years of positive growth. In wake of this halt, and subsequent lowered nesting averages, evaluation of the genetic diversity of remaining individuals is relevant to investigate the plausibility of a genetic bottleneck, and determine the reproductive stability of the Kemp’s ridley.

The objective of this thesis was to identify polymorphisms in the glucocorticoid receptor (GR) and interleukin-8 receptor (CXCR2) genes and to associate genotypes between the above mentioned polymorphisms and production traits in crossbred cattle. The hypothesis was that polymorphisms will exist for GR and CXCR2 genes and will be linked to production traits. Glucocorticoid receptors have been positively associated with higher milk yields, lactose content, feed intake, and feed conversion rates. Mastitis, the inflammation of the mammary gland, dramatically decreases dairy industry revenues and milk quality, making improved control and prevention methods a goal of the industry. Prior research has observed variation in response to experimental challenge with regards to inflammation indicators, S. uberis concentrations, and the need for antibiotics. To determine possible causes of the observed variation following experimental S. uberis intramammary challenge, we performed genome association analyses (N = 34 – 36 Holstein dairy cows), a linkage analysis, and S. uberis milk inoculation (N = 21 Holstein dairy cows) trials. Calving rate is a qualitative trait regulated by several genes and is strongly affected by the environment. With the development of biotechnology and gene identification, scientists are able to determine which genes affect these productivity traits to improve accurate selection decisions. Prolactin (PRL) has been associated with reproductive traits, melatonin receptor 1a (MTNR1A) has been associated with meat quality traits, and the dopamine receptor D2 (DRD2) gene has been associated with hair coat score (HCS) in cattle. Use of a genetic-based analogue of the traditional mark-recapture method (transgenerational genetic mark-recapture, tGMR) is rapidly expanding as a means to estimate total escapement of Pacific salmon. The tGMR approach is similar to the simple Lincoln-Peterson mark-recapture method. In tGMR, adults returning to fresh water to spawn are collected in the first sampling occasion and their juvenile offspring that are migrating out to sea are collected during the second sampling occasion. Recaptures are determined by the number of parent-offspring pairs identified through genetic parentage analysis of the adult and juvenile collections. The cloning of domestic species has tremendous potential, as the technology can be used in selective breeding, conservation, and the production of transgenic animals. The technique of cloning involves the transplant of DNA from a cell to a recipient gamete. Following transfer to a surrogate, the cloned embryo may successfully complete development to a live offspring. Despite intensive research, the success rate of cloning remains prohibitively low, and the potential benefits of cloning have not yet been realized. Understanding gene flow, diversity, and dispersal patterns is important for predicting effects of natural events and anthropogenic activities on dolphin populations. With the very recent exceptions of false killer whales (Pseudorca crassidens), spinner dolphins (Stenella longirostris), and common bottlenose dolphins (Tursiops truncatus), Hawaiian odontocete species are managed as single stocks within the U.S. Hawaiian Exclusive Economic Zone. The San Luis Valley is a large valley formation in Colorado surrounded on either side by mountain ranges exceeding 4,267 m. Within the Valley, two of the 14 amphibian and reptile species are dwarfed: the short-horned lizard (Phrynosoma hernandesi) and the Great Plains toad (Anaxyrus cognatus). Since its initial reporting in 1968 and confirmation in 1981, no research further investigating this dwarfism has been conducted. I collected morphological measurements to determine the extent and patterns of dwarfism of both species. Pax genes encode transcription factors that are highly evolutionarily conserved and are vital for animal development. Vertebrate Pax group III genes Pax3 and Pax7 are required for proper development of muscular and central nervous systems. In their roles in muscular systems, Pax3 and Pax7 specify myoblasts (muscle progenitor cells). Pax3 is predominantly involved in embryonic myogenesis and specifies embryonic myoblasts. In contrast, Pax7 specifies adult myoblasts (muscle satellite cells), and has been demonstrated to be vital for adult muscle regeneration. It was the purpose of this study (1) to present a method of teaching principles of heredity at a level understandable to students of a high school biology class, (2) to present a list of resource materials for use in a heredity unit, and (3) to show how the use of live animals might intensify student interest in this subject as well as in other phases of bird life such as nest building, incubation of eggs, hatching of eggs, voice and communication, and effects of nutrition on chick development.
