Recent developments in the field of microfluidics have created a multitude of new useful techniques for practical particle and cellular assays. Among them is the use of dielectrophoretic forces in 'lab-on-a-chip' devices. This sub-domain of electrokinetic flow is particularly popular due to its advantages in simplicity and versatility. This thesis makes use of dielectrophoretic particle manipulations in three distinct spiral microchannels. In the first of these experiments, we demonstrate the utility of a novel single-spiral curved microchannel with a single inlet reservoir and a single outlet reservoir for the continuous focusing and filtration of particles. Dielectrophoresis has long been studied and utilized for the manipulation of microscale particles in solution. This phenomenon is due to the induced polarization of dielectric particles subjected to an electric field. When the field is also inhomogeneous in terms of the distribution of its strength through space, the polarized particles move and come to rest in certain areas due to the relationship between their and the solvent's relative permittivities. If the electric field is homogenous, such as within a parallel plate capacitor, the particles are polarized according to their permittivity and the field's frequency, but they will not move.

With the recent advancement in micro-fabrication technology, lab-on-a-chip devices have been developed in order to perform biological analysis through cell manipulation. Microchannels used in these lab-on-a-chip devices have been demonstrated to accurately perform many different cell manipulation techniques such as focusing, separation, trapping, and lysis. Although there are many methods available for these techniques, electrokinetics has been rapidly gaining popularity due to the simplicity of application and removal of the need for in channel micro-structures.
