Biomaterial platforms have been used to probe how cells respond to chemical, topographical or mechanical changes in their environments. However, these traditionally static biomaterial platforms have not enabled the study of how cells respond to dynamic changes in their environment. Tissue engineering has emerged as a promising strategy for the replacement of degenerating or damaged tissues in vivo. Also known as regenerative medicine, integral to this therapeutic strategy is biomimetic scaffolds and the biomaterial structural components used to form them. In this study, three different biomaterial scaffolds for tissue engineering applications were fabricated: three-dimensional reverse embedded collagen scaffolds, polymer fusion printed polycaprolactone (PCL) scaffolds, and electrospun gelatin scaffolds. Three-dimensional collagen and PCL scaffolds promoted human adipose-derived stem/stromal cell (ASC) spreading, proliferation, and fibronectin deposition in vitro. Proteins have intrinsic molecular properties that are highly useful for materials applications, especially for biomaterials. My research has focused on translating these molecular properties to materials surface behavior. In one approach, I developed a fluorous-based thermal treatment strategy to generate stable thin films from a variety of naturally abundant proteins. The different surface properties generated from the choice of protein were utilized to modulate cell-surface interactions, prevent bacterial adhesions, and control drug loading/release. Lower respiratory diseases are currently the third leading cause of death worldwide. For many end-stage patients with these diseases, there is no cure and a shortage of donor organs available for transplant. A promising solution is to design regenerative scaffolds or complete bioengineered lungs, using decellularized lung tissues as a template for regeneration. Recent advances in the field have made significant strides towards developing a transplantable lung.
