The decomposition of barley starch plays a fundamental role in the final quality of fermented beverages in the brewing industry. The process of mashing involves steeping milled barley in water to stimulate the enzymatic decomposition of starch into simple carbohydrates. The temperature of the water significantly impacts the activity of the enzymes and in turn the resulting concentrations of sugar products. A kinetic model accurately predicting the types and quantities of sugars formed as a function of mash temperature would serve as a tool to tailor sugar profiles and optimize mashing procedures for a desired product.

Opaque fluids have a limited irradiation transmission. Thus, their decontamination employing near UV irradiation poses significant technical challenges. In the present study, a thin UV-transparent/waterproof glue layer coated with a 1.5 wt% of TiO2 and a new PhotoReactor Cell were implemented. TiO2 irradiation in the PhotoReactor Cell was effected on the TiO2 particle side, not directly in contact with the fluid, allowing the postulation of an “h+” site mobility mechanism on photocatalysis.

The slow pyrolysis of different biomasses (maple wood, birch bark, switch grass, coffee pulp and corn stalk) was studied with the aim to produce a solid pyrolysis product (bio-coal) with promising properties and potential for use in traditional fossil-coal applications. Batch pyrolysis experiments were conducted in a Mechanically Fluidized Reactor (MFR) at temperatures ranging from 143 to 665 oC, which includes the torrefaction temperature range (200-300 oC). This thesis reports the viability of E. coli cell extracts prepared using equipment that is both common to biotechnology laboratories and able to process small volume samples and expression of proteins containing unnatural amino acids (UAAs) at higher level using PCR amplified linear DNA templates (LETs) in cell-free protein synthesis (CFPS) system. E. coli-based cell extracts are a vital component of inexpensive and high-yielding CFPS reactions. Biodiesel is produced from vegetable oils through transesterification. Triglyceride mixtures extracted from oilseed feedstocks are upgraded by reaction with an alcohol in the presence of a catalyst to produce fatty acid esters. This reaction produces a mixture of esters, glycerin, alcohol, and catalyst. Separation of the fatty acid esters (biodiesel) and glycerin can be accomplished through liquid-liquid extraction by water addition. Designing liquid-liquid extraction with water as the solvent requires ternary liquid-liquid equilibrium data for mixtures of water, glycerin, and fatty acid esters. The present work demonstrates the application of molecular simulations (MD) in two different areas: proton transport in bulk water and estimation of the dipole moment of polar organic compounds. In both areas, relatively few successful and robust methodologies exist. In the first part, a new polarizable water model is developed for MD simulations of the proton transport process. The model was parametrized from a combination of quantum chemical calculations and experimental water properties. The objective of this thesis was to determine appropriate conditions for cleavage of acetyl groups from northeast hardwood. Currently, acetyl groups end up in a waste product stream in most wood based pulp mills and thus are underutilized. Recently, a techno‐economic analysis was published that evaluated the suitability of the ‘Near‐Neutral Extraction’ process for recovery of acetic acid and ethanol in Kraft pulp mills. Recently, ensemble modeling was applied to metabolic networks for the sake of predicting the effects of genetic manipulations on the observed phenotype of the system. The ensemble of models is generated from experimental wild-type flux data and screened using phenotypic data from gene overexpression and knockout experiments, leaving predictive models. In this study, a novel continuous flow electroless (CF-EL) Au plating method is developed to fabricate uniform Au thin films in ambient condition. The detection of oligomers and aggregates formed by two amyloid proteins, insulin and amyloid-beta (AB), is of particular importance due to the role which these species play in Diabetes and Alzheimer's disease, respectively. However, existing techniques are limited in the ability to detect insulin and AB; oligomers due to the fact that these early aggregates are transient, present at low concentrations, and difficult to isolate. There is a growing interest in renewable, carbon-neutral biofuels such as ethanol and biodiesel. A life-cycle analysis is conducted in this study to determine the viability of using algae as a feedstock for biodiesel. The method involves assessing energy use, fossil fuel use, greenhouse gas emissions, and criteria pollutant emissions using a simulation developed by Argonne National Laboratory. The energy and emissions of algae-derived biodiesel are compared to those of soybean biodiesel, corn ethanol, conventional gasoline, and low-sulfur diesel. This dissertation explores the potential of using perfluorocarbon emulsion droplets to add an ultrasound-sensitive element to drug delivery systems. These emulsion droplets may be induced to vaporize with ultrasound; during the rarefactional phase of an ultrasound wave, the pressure around the droplets may fall below the vapor pressure of the liquid forming the emulsion, providing a thermodynamic potential for vaporization. For this work, ammonia (NH3) and benzene (C6H6) effects on cell growth, enzymatic activities of hydrogenase and alcohol dehydrogenase (ADH), and product formation were studied. NH3, after entering media, will be converted rapidly to NH4+, which will raise the total osmolarity of the media. NH3, as a common nutrient for the cell growth, is not the real culprit for cell growth inhibition. Bone defects are a prevalent problem in orthopedics and dentistry. Calcium phosphate-based coatings and nanocomposites offer unique solutions towards producing scaffolds with suitable physical, mechanical and biological properties for bone regeneration.

We developed a novel method to synthesize hydroxyapatite (HA) particles with high aspect ratio using sol-gel chemistry and hydrothermal treatment. We obtained tunable pure-phase carbonated-HA in the form of micro/nanorods and nanowires (diameters 25-800 nm). To mimic the structure of bone, HA nanowires were homogenously mixed within poly(ε-caprolactone) (PCL) to produce nanocomposites with improved mechanical properties as determined by uniaxial tensile testing.

Fluid CokingTM is a major refining process to upgrade heavy crude oil or bitumen from oil sands. In Fluid CokingTM bitumen is atomized with steam and sprayed inside a high temperature fluidized bed of coke particles where it thermally cracks into smaller molecules. Fluid CokingTM is a carbon rejection process as solid carbon residues are formed during fragmentation into lighter hydrocarbon vapors. New coke deposits over existing coke particles and, consequently, the particle size increases.
