Despite major advances in revascularization and drug therapy for post-myocardial infarction (MI) heart failure (HF), heart transplantation remains the only curative therapy to prevent progressive and debilitating disease. Researchers have long sought a replenishable and expandable source of cardiomyocytes (CMs), the primary functional cell in the heart muscle, for a broad array of applications related to the treatment of HF ranging from direct implantation to high throughput drug testing. Neuroendocrine tumors (NETs) are a heterogeneous group of neoplasms that originate in a diverse array of anatomical locations and have widely variable clinical presentations and responses to treatment. Patient-derived xenografts and cell lines are valuable models of tumor biology and for testing personalized therapeutics on tumors prior to their administration to patients. However, as PDX models and cell lines for neuroendocrine tumors are lacking in both number and validity, we have developed a 3D flow perfusion polydimethylsiloxane (PDMS) bioreactor model for the purpose of culturing patient derived NETs. Drug development is an expensive and time consuming process, requiring an average of over $1 billion and up to fifteen years of work for each newly released product. Drug-induced liver toxicity is the leading cause of post-market drug withdrawals despite the fact that extensive animal testing and clinical trials are required prior to approval of a novel drug or therapy. Hydroxyapatite (HA) makes up the mineral component of bone, and synthetic HA is commonly used to coat metal implants for use in dentistry and orthopedics. Additionally, allograft and synthetic HA are common alternatives to autograft bone, to circumvent the complications associated with a second surgery site. While both HA and allograft are known to be osteoconductive, osteoinductivity is limited. In this work, we developed methods for anchoring osteoinductive factors onto multiple types of HA biomaterials and human allograft bone, with the goal of enhancing their osteoregenerative potential.
