Levodopa is a dopamine replacement medication administered to patients with Parkinson’s disease (PD) to alleviate their motor symptoms. However, its long-term use can cause adverse side effects, including involuntary motor movements. We studied 16 PD patients before and after taking Levodopa based on resting-state electroencephalography (EEG) recordings to determine how Levodopa affects the functional connectivity of their brain networks. We used several metrics from graph theory, in particular the minimum spanning tree (MST) metric, and analyzed how they change after subjects take Levodopa. Non-motor Parkinson’s Disease (PD) symptoms are substantial factors of PD arising throughout disease stages, yet their diagnosis and monitoring remain a challenge. Sensory abnormalities in PD occur across sensory systems and disease stages, contributing to disease-related impairments. However, the extent of symptoms is unknown, with inadequate monitoring and treatment options furthering disease management difficulties. The current work studies movement-independent visual perceptions of time, displacement and velocity in PD patients across disease stages using levodopa, deep brain stimulation (DBS), or no PD therapy. An improved understanding of glenohumeral bone mechanics can be elucidated using computational models derived from computed tomography data. Although computational tools, such as finite element analysis, provide a powerful quantitative technique to evaluate and answer a variety of biomechanical and clinical questions, glenohumeral finite element models (FEMs) have not kept pace with improvements in modeling techniques or model validation methods seen in other anatomic locations. The present work describes the use of multi-level computational modeling to compare, develop and validate FEMs of the glenohumeral joint.

Hydrogels are 3-dimensional crosslinked polymer networks that can absorb significant amounts of water. The physical properties associated with hydrogels affords them resemblance to biological tissues making them good candidates for biomedical applications. Many pharmaceuticals, specifically non-steroidal anti-inflammatory drugs (NSAIDs), have poor aqueous solubility, which limits their bioavailability and efficacy. People suffering from chronic osteoarthritis (OA) are required to frequently take large doses to mitigate pain, which can lead to serious side effects. An in-vitro musculoskeletal loading simulator was developed to replicate the internal forces of mastication, and then employed in a comparison of clinically relevant facial fracture repairs. Muscle forces are simulated by pneumatic pistons via 3D printed mounts that are reverse-engineered from CT scan to match muscle attachment sites, which are adhered to bone in order to simulate native stress distributions. Bite force and bone strain pattern of the craniofacial structure under load were measured using a force sensor and strain gauges respectively. We studied the graph topological properties of brain networks derived from resting-state functional magnetic resonance imaging in a kainic acid induced model of temporal lobe epilepsy (TLE) in rats. Functional connectivity was determined by temporal correlation of the resting-state Blood Oxygen Level Dependent (BOLD) signals between two brain regions during 1.5% and 2% isoflurane, and analyzed as networks in epileptic and control rats. Graph theoretical analysis revealed a significant increase in functional connectivity between brain areas in epileptic than control rats, and the connected brain areas could be categorized as a limbic network and a default mode network (DMN). Patients diagnosed with Parkinson’s disease (PD) often associate with tremor. Among other symptoms of PD, tremor is the most aggressive symptom and it is difficult to control with traditional treatments. This thesis presents the assessment of Parkinsonian hand tremor in both the time domain and the frequency domain, the performance of a tremor estimator using different tremor models, and the development of a novel mechatronic transmission system for a wearable tremor suppression device. Many vascular and cardiac diseases are diagnosed and treated using a medical technique known as percutaneous transluminal catheter intervention (PTC). In PTC, the interventionalist inserts a catheter into the vasculature, and using the vessel as the guiding passageway, the catheter is navigated to desired anatomical targets where it would be used for various purposes such as catheter ablation for the treatment/management of cardiac arrhythmias. The catheterization procedure is conventionally guided with x-ray fluoroscopic imaging and more recently, but rarely, with Magnetic Resonance Imaging (MRI). The introduction of transcatheter aortic valve implantation (TAVI), an innovative stent-based technique for delivery of a bioprosthetic valve, has resulted in a paradigm shift in treatment options for elderly patients with aortic stenosis. While there have been major advancements in valve design and access routes, TAVI still relies largely on single-plane fluoroscopy for intraoperative navigation and guidance, which provides only gross imaging of anatomical structures. Inadequate imaging leading to suboptimal valve positioning contributes to many of the early complications experienced by TAVI patients, including valve embolism, coronary ostia obstruction, paravalvular leak, heart block, and secondary nephrotoxicity from contrast use.
