In general, biomaterials induce a non-specific host response when implanted in the body. This reaction has the potential to interfere with the function of the implanted materials. One method for controlling the host response is through local, controlled-release of anti-inflammatory agents. Herein, we investigate the spatial and temporal effects of an anti-inflammatory drug on the cellular dynamics of the innate immune response to subcutaneously implanted poly(lactic-co-glycolic) microparticles. Noninvasive fluorescence imaging was used to investigate the influence of dexamethasone drug loading and release kinetics on the local and systemic inhibition of inflammatory cellular activities. The non-specific host response to implanted biomaterials is often a key challenge of medical device design. To evaluate biocompatibility, measuring the release of reactive oxygen species (ROS) produced by inflammatory cells in response to biomaterial surfaces is a well-established method. However, the detection of ROS in response to materials implanted in vivo has not yet been demonstrated. Here, we develop a bioluminescence whole animal imaging approach to observe ROS released in response to subcutaneously-implanted materials in live animals. A computational vascular fluid–structure interaction framework for the simulation of patient-specific cerebral aneurysm configurations is presented. A new approach for the computation of the blood vessel tissue prestress is also described. Simulations of four patient-specific models are carried out, and quantities of hemodynamic interest such as wall shear stress and wall tension are studied to examine the relevance of fluid–structure interaction modeling when compared to the rigid arterial wall assumption. We demonstrate that flexible wall modeling plays an important role in accurate prediction of patient-specific hemodynamics. Discussion of the clinical relevance of our methods and results is provided.

To effectively postpone preterm birth, cervical ripening needs to be detected and delayed. As the cervix ripens, the spacing between the collagen fibers increases and fills with water, hyaluronan, decorin, and enzymes suggesting that the ultrasonic attenuation of the cervix should decrease. The decrease in ultrasonic attenuation may be detectable, leading to an effective means of detecting cervical ripening. Herein, the traditional attenuation slope-estimation algorithm based on measuring the downshift in center frequency of the ultrasonic backscattered signal with propagation depth was modified and applied to the cervix of rats. Magnetic resonance imaging is used to study changing microstructure and molecular motion during dissolution of poly(vinyl alcohol) (PVA) in water. One-dimensional water concentration profiles were measured as a function of distance from the polymer-solvent interface. Diffusion-weighted profiles were used to calculate the spatial variation of the self-diffusion coefficient of water. The results indicate that diffusion coefficient values decrease toward the glassy core of the polymer. Self-diffusion coefficient values within the dissolving polymer increase with increasing dissolution time, while those near the polymer-solvent interface remain fairly constant. A new postprocessing method of correcting for respiratory motion induced artifacts in MRI is presented. The motion of the chest during respiration is modeled as a combination of translation and dilation. Displacements of the chest wall are tracked via a thin, MR-sensitive plate placed on the patient's chest during a scan. Scanning with phase encoding left/right (L/R) and frequency encoding anterior/posterior (A/P) causes the motion artifacts to be repeated in the L/R direction, thus not overlapping on the plate. A prototype hypermedia decision support system for the selection and documentation of rural assistive technology (BNG DATA) was developed to aid professionals working with farmers, ranchers, and agricultural workers with physical disabilities. The hypermedia system (constructed using HyperCard, an environment that combines hypertext and database features) consists of a hypermedia database of rural assistive technology examples and an accompanying decision support system that helps users identify solution alternatives to meet the needs of their clientele. End user acceptance of BNG DATA was determined using an evaluation questionnaire. The end users evaluating the prototype considered BNG DATA to be easy to …
