In this work, an electrospinning device for fabricating three-dimensional scaffolds was constructed. The device can be used to produce tissue engineered scaffolds electrospun for directing the differentiation of human induced pluripotent stem (iPS) cells into glucose-responsive, insulin-secreting cells for diabetes treatment. Electrospinning utilizes high electrical charge to generate nanofibers from polymers dissolved in solution. A standard setup for such a device includes a high-voltage power source, a syringe pump, and a grounded collector. The process works by dissolving the polymer in a volatile solvent, and filling a syringe with the polymer/solvent mixture. Next, the syringe is locked into place in …

The overall goal of this study was to identify mineralized scaffolds which can serve as potential alternatives to bone graft substitutes intended for cleft palate repair. The aim of this preliminary study was to evaluate the role of fibrinogen (Fg) and nano-hydroxyapatite (nHA) in enhancing mineralization potential of polydioxanone (PDO) electrospun scaffolds. Scaffolds were fabricated by blending PDO:nHA:Fg in the following weight ratios: 100:0:0, 50:25:25, 50:50:0, 50:0:50, 0:0:100 and 0:50:50. Scaffolds were immersed in different simulated body fluids for 5 and 14 days to induce mineralization. This paper reports a new method of cross-linking electrospun zein fibers using citric acid as a non-toxic cross-linker to enhance the water stability and cytocompatibility of zein fibers for tissue engineering and other medical applications. The electrospun structure has many advantages over other types of structures and protein-based biomaterials possess unique properties preferred for tissue engineering and other medical applications. However, ultrafine fiber matrices developed from proteins have poor mechanical properties and morphological stability in the aqueous environments required for medical applications.
