Microdialysis (µD) sampling is a diffusion-limited sampling method that has been widely used in different biomedical fields for greater than 35 years. Device calibration for in vivo studies is difficult for current non-steady state analytes of interest correlated with both inflammatory response and microbial signaling molecules (QS); which exist in low ng/mL to pg/mL with molecular weights over a wide range of 170 Da to 70 kDa. The primary performance metric, relative recovery (RR), relating the collected sample to the extracellular space concentration varies from 10% to 60% per analyte even under controlled bench-top conditions. Bioluminescence resonance energy transfer (BRET) is a distance dependent, non-radiative energy transfer, which uses a bioluminescent protein to excite an acceptor through resonance energy transfer. In this thesis, BRET technology is incorporated into a sensor comprised of a recombinant protein and quantum dots. The recombinant protein, which includes the bioluminescent protein, Renilla luciferase (Rluc), is used as the donor molecule and cadmium tellurium quantum dots as the acceptor molecules. Separating the donor-acceptor pair is a recombinant protein, glucose binding protein, which changes conformation upon binding glucose and brings the pair closer together, thus allowing BRET to occur. Crude oil desalting operations produce an effluent stream which is challenging to treat due to its salt, heavy metal and hydrocarbon content. Consequently, desalter effluent (DE) is usually diluted into other effluent streams and sent to conventional wastewater treatment plants which may lead to upsets the plant operation.

In this study, a novel microbial approach was applied which investigated DE treatment using halotolerant yeast Debaryomyces hansenii (LAF-3 10 U) or the environmentally robust micro-algae Parachlorella kessleri strain CPCC 266. The aim of this study was to examine volatile fatty acid (VFA) production from a proteinaceous substrate, bovine serum albumin (BSA) for a pH range of 5 – 9, and to further assess its impact on hydrogen production in a co-fermentation process using starch and BSA at different ratios. The established optimum conditions for VFA production from BSA were an initial pH of 8, incubation time of 3 days and an operating temperature of 37 ℃. Volumetric muscle loss overwhelms skeletal muscle’s ordinarily capable regenerative machinery, resulting in fibrosis and severe functional deficits which have defied clinical repair strategies. My work spans the design and preclinical evaluation of implants intended to drive the cell community of injured muscle toward a regenerative state, as well as the development of an understanding of the molecular responses of this cell community to biomaterial interventions. Computational chemistry is a powerful tool to model different materials and predict their physical and chemical properties. The goal of this thesis was to apply different computational chemistry tools to model the properties of two classes of closely related organic compounds: formazans and tetrazolium salts for ionizing radiation dosimetry in water-based gels, whereby the absorbed dose is registered by optical detection of formazan formation from tetrazolium salts. In E. coli, fatty acid synthesis is catalyzed by the enzyme acetyl-CoA carboxylase (ACC), which converts acetyl-CoA into malonyl-CoA. Malonyl-CoA is a major building block for numerous of bioproducts. Multiple parameters regulate the homeostatic cellular concentration of malonyl-CoA, keeping it at a very low level. Understanding how these parameters affect the bacterial production of malonyl-CoA is fundamental to maximizing it and its bioproducts. Continued population growth and the decrease of existing energy platforms demands long-term solutions for development and implementation of scalable plasmonic metamaterials for energy and agricultural applications. Self-assembled nanoantenna into random and ordered arrangements are advanced herein for optical and thermal enhancements in scalable thin film. An analytical approach to estimating the thermal dynamics of random arrangements of nanoantenna resulted in estimates within 30% across a range of geometric parameters, nanoantenna-containing media, and thermal parameters. Cystic fibrosis (CF) is the most common genetic disease resulting in the morbidity and mortality of Caucasian children and adults worldwide. Due to a genetic mutation resulting in malfunction of the Cystic Fibrosis Transmembrane Conductance Regulator (CFTR) protein, CF patients produce highly viscous mucus in their respiratory tract. This leads to impairment of the mucociliary clearance of inhaled microbes. In addition to reduced microbial clearance, anoxic environmental conditions in the lungs promote biofilm-mode growth of the pathogenic bacterial species Pseudomonas aeruginosa. This environment-responsive phospholipid layer consisting of multiple dynamically balanced macromolecules, such as membrane proteins, carbohydrate and lipids, is regarded as a promising platform for various surface engineering strategies. Naturally occurring enzymatic pathways enable highly specific, rapid thiophenic sulfur cleavage occurring at ambient temperature and pressure, which may be harnessed for the desulfurization of petroleum-based fuel. One pathway found in bacteria is a four-step catabolic pathway (the 4S pathway) converting dibenzothiophene (DBT), a common crude oil contaminant, into 2-hydroxybiphenyl (HBP) without disrupting the carbon-carbon bonds. 2’-Hydroxybiphenyl-2-sulfinate desulfinase (DszB), the rate-limiting enzyme in the enzyme cascade, is capable of selectively cleaving carbon-sulfur bonds. Mesoporous silica nanoparticles (MSNPs) possess large surface areas and ample pore space that can be readily modified with specific functional groups for targeted binding of bioactive materials to be transported through cellular barriers. Engineered silica nanoparticles (ESNP) have been used extensively to deliver bio-active materials to target intracellular sites, including as non-viral vectors for nucleic acid (DNA/RNA) delivery such as for siRNA induced interference. Gamma-Secretase (γ-secretase) is a transmembrane protease of increasing interest, which has been shown to have significant connections to both cancer and Alzheimer’s disease. γ-secretase cleaves both Notch-1, a transmembrane signaling protein, and Amyloid precursor protein (APP), a transmembrane protein whose cleavage may result in the formation of β-amyloid plaques in the brain. Notch-1 and APP are widely studied proteins that have substantial impacts on the development and proliferation of cancer and Alzheimer’s disease, respectively. Due to the demand for in-situ electrolyte sodium sensors, prototype sensors have been produced to create a product for athletes and marathoners to monitor hydration levels. These senors address a desire in the marketplace for athletic monitoring for overall athlete wellness and performance. Sweat sensor samples were created using Multi-walled Carbon Nanotubes functionalized on a Nylon-6 fiber mat using cyclo-oligomeric Calixarene. This production process produces sensors capable of detecting sodium ion concentration present in human sweat in presence of an electric current. The tested hypothesis was that features on polydimethylsiloxane (PDMS) surfaces coated with a poly(N-isopropylacrylamide)/aminopropyltriethoxysilane or pNIPAAM/APTES thin film would accelerate cell detachment than the film coated on a unfeatured surface. This project tested samples with features generated by molds, wrinkling, and sandpaper roughened substrates. Surface feature generation methods were limited to mechanical means, and characterized by microscopy and strain rates. 50/50 mixtures of 1.5 wt.% pNIPAAM/ APTES were used to coat thin films (30-40 nm) on PDMS membranes by spin-coating, and the coated membranes were thermally annealed to chemically graft pNIPAAm/APTES on the membrane and their thermo-responsive property was assessed …

The main objective of this honor’s project is to encapsulate drugs with different molecular weights inside of chitosan-heparin based nanoparticles (Cs-Hep NPs). Three different drugs with low (NNC), medium (IFN-γ), and large (BSA) molecular weights were chosen in order to compare all types of drugs and how they react with the nanoparticles. It was hypothesized that through the use of Hofmeister Ions, the lower critical solution temperature (LCST) of poly(vinyl methyl ether) (PVME) and poly(ethylene glycol)- poly(N-isopropylacrylamide) (PEG-b-pNIPAAm) block-copolymer could be controlled. Through literature searches and small lab experiments, our team found that there may be a connection between Hofmeister effects and phase transition of a thermo-responsive polymer.
