Purpose: 1) To compare the effects of an acute bout of HIIT exercise (treadmill running) on autophagy to MICT exercise in human skeletal muscle 3 hours post exercise, and 2) to test an in vitro model of muscle contraction-induced autophagy using electrical pulse stimulation in C2C12 myotubes. Methods: Study 1: Ten recreationally active males and females completed a bout of high intensity interval training (HIIT) exercise and moderate intensity continuous exercise (MICT) exercise in a fasted state. Proteins play a critical role in nearly every biological activity. In consequence, organismal health and homeostasis often hinges on the ability of intracellular regulatory systems to sustain the quality and function of these diverse, structurally complex macromolecules. Correct protein function depends on correct form, and during periods of destabilizing cellular stress, protein quality is managed in part by the heat shock response, which acts to support, isolate, and reform new or damaged proteins, and in part by the autophagic recycling of abnormal proteins, cytotoxic protein aggregates, and terminally damaged organelles.
