Friedreich's Ataxia (FA) is an autosomal-recessive, neurodegenerative disease characterized by progressive lower extremity muscle weakness and sensory loss, balance deficits, limb and gait ataxia, and dysarthria. FA is considered a sensory ataxia because the dorsal root ganglia and spinal cord dorsal columns are involved early in the disease, whereas the cerebellum is affected later. Balance deficits and gait ataxia are often evaluated clinically and in research using clinical rating scales. Recently, quantitative tools such as the Biodex Balance System SD and the GAITRite Walkway System have become available to objectively assess balance and gait, respectively. Accurate fishery-independent methods for assessing the abundance of marine fish stocks are important tools for proper pelagic fisheries management. The review and improvement of standardized sampling methods used in fisheries-independent sampling, and the establishment of improved models of abundance and population dynamics utilizing novel statistical techniques for fisheries management will further improve our understanding of the way in which marine fish stocks vary spatially and temporally. One of the most important and longest running surveys of fisheries-independent data in the Southeastern United States is the spring ichthyoplankton survey conducted through the Southeast Area Mapping and Assessment Program (SEAMAP).

The interrogation of T cell signaling over the past fifty years has led to the discovery of amazingly intricate cascade networks and elaborate descriptions of individual proteins' domains and functions. A complex landscape has been rendered in which proteins relay messages from the extracellular ligation of the TCR by a cognate peptide loaded MHC via changes in sub-cellular location, phosphorylation, and binding affinities and partners to enact nuclear localization of three key transcription factors required for cellular effector function and proliferation: AP-1, NF-AT, and NF-κB. The biodiversity of two distinct marine environments was observed to describe the biogeocomplexity of these extreme ecological systems. A shallow-water hydrothermal vent in Papua New Guinea served as a study of a thermophilic ecosystem influenced by arsenic rich vent fluids while a 60 m deep offshore primarily anoxic karst sink served as a study of an anaerobic sulfur-influenced habitat. Both environments support unique biological communities that are influenced by the physical and chemical pressures imposed on them by the harsh conditions of these systems. Although iron occurs at extremely low concentrations in the world’s oceans, it is essential for all living organisms. It is the limiting nutrient in High Nutrient Low Chlorophyll (HNLC) areas of the ocean, and exerts critically important influences on levels of atmospheric CO2 and the global carbon cycle. Billfishes (marlins, spearfishes, sailfishes and swordfish) are one of the fastest and largest marine apex predators, and perhaps their most recognizable attribute is their bill or rostrum. The proposed function for this novel structure has ranged from hydrodynamic enhancement to defensive weaponry. However, the most supported hypothesis for its function has been linked to feeding. Billfishes have been observed to subdue their prey with their rostrum, either stunning or cutting them into pieces before ingestion. Soils are critical to ecosystem function as they provide essential nutrients for primary producers, habitat and organic energy for decomposers, and storage of organic matter. Irrigation with reclaimed water is an increasingly popular water conservation strategy; yet its high salinity and nutrient content potentially affect soil properties. In this study, set in a residential neighborhood of Tampa (U.S.). I tested whether there are distinct lawn system management strategies characterized by systematic differences in reclaimed water usage and irrigation and fertilization practices. The Deepwater Horizon blowout occurred on April 20th, 2010 and released nearly 5 million barrels of crude oil into the northern Gulf of Mexico causing pollution of the water and sediment inhabited by many fishes for at least 87 days while the wellhead went uncapped. The Deepwater Horizon blowout occurred on April 20th, 2010, releasing 4.9 million barrels of Louisiana crude oil into the Gulf of Mexico (GoM). Subsequent to the Deepwater Horizon blowout, sediment cores revealed oil on the northern GoM seafloor and abnormal skin lesions were seen in GoM fishes. Exposure to polycyclic aromatic hydrocarbons (PAHs), a component of crude oil, in fish has been associated with many sublethal effects, including cancer and population-level effects. Reef fish species tend to reside over high relief habitat which makes them difficult to sample with traditional gears such as nets and trawls. Therefore, implementing and understanding the strengths and weaknesses of new approaches which incorporate acoustic and optical methods has become a priority for reef fish stock assessment. Beginning in June of 2013, a towed camera system known as the Camera-Based Assessment Survey System (C-BASS) has been used to visualize over 500 kilometers of transect and record more than 80 hours of video over several habitats in the Gulf of Mexico. Cytokines are pleiotropic and redundant signaling molecules that govern the inflammatory response and immunity, a critical ecological parameter for organism success and population growth. Produced at the site of injury or pathogen intrusion by a variety of cell types, cytokines mediate cell-signaling in either an autocrine or paracrine manner. Over the past several decades, rapid decline in adult stony-coral (comprising the Orders Scleractinia and Anthomedusae, specifically Family Milleporidae) cover has occurred concurrent with an increase in adult octocoral (Octocorallia/gorgonian) cover along the Florida Reef Tract. In January 2010, the Florida Keys experienced extremely cold air and water temperatures, below the lethal threshold for many reef organisms including corals. Very high stony-coral mortality occurred on some patch reefs. The newly-available space created by this disturbance event provided the opportunity for recruitment and settlement of new coral larvae and other reef organisms.

Knowledge of species distribution and habitat associations are essential for conservation measures. Such information is lacking for many marine species due to their occupancy of broad and ephemeral habitats that are difficult to access for study. Sea turtles, specifically the surface-pelagic juvenile stage of some species, are a group for which significant knowledge gaps remain surrounding their distribution and habitat use. Recent research has confirmed the long-standing hypothesis that the surface-pelagic juvenile stage occurs within surface-pelagic drift communities (SPDC). Unfortunately, many patients either do not have any common mutations for which there are already targetable agents, or they eventually become resistant to these compounds. As such, there is a high demand for new, effective methods of treating this disease as well as predicting patient prognosis and potential benefit from chemotherapy. This dissertation is devoted to the study of the molecular biology of major tumor suppressors, defined as those that prevent the cellular processes identified as the hallmarks of cancer. Specifically, the major tumor suppressors pRb and STK11 are explored in the context of osteosarcoma and lung cancer, respectively.

RB1 was the first tumor suppressor gene discovered. Ovarian cancer is one of the most common causes of gynecological cancer related deaths in women. In 2014, the estimated number of deaths due to ovarian cancer is 14,270 with occurrence of over 22, 240 new cases (National Cancer Institute, http://seer.cancer.gov/statfacts/html/ovary.html). Unraveling the microbiological processes that occur as water travels from a river's mouth into the ocean is critical to understanding the role of river plumes in global biogeochemical cycles. Metranscriptomics, the gene expression of a whole community of organisms, was utilized to examine six stations along the Amazon River Plume (ARP) in 2010 to test thehypothesis that there were measurable differences in gene expression for key biogeochemical genes along the ARP. The overall goal of this dissertation is to define the hydrological, geochemical, and biological characteristics of a Karst Estuary. These types of estuaries represent a unique ecosystem created by freshwater inputs from direct flow through karst conduits and/or diffuse flow through a karst matrix. In order to determine the characteristics of a Karst Estuary we monitored short-term tidal fluctuations, long-term rainfall patterns, aquifer levels, spring discharge, multiple geochemical parameters, microbial communities in the water column and sediment, and macrofaunal communities in the sediment along a transect from a submarine spring through the Gulf of Mexico. Globally, seagrass ecosystems are located adjacent to coastal areas that are heavily impacted by human development and urbanization potentially altering the community structure within these ecosystem. Malaria is a critical and global public health problem, affecting over 200 million people every year, resulting in over 500,000 deaths. A vaccine is not currently available and only one drug, primaquine, is effective against the dormant stages of Plasmodium vivax. Very few cancer patients are cured through drug therapy alone, with the majority exhibiting acquired resistance. To date, most studies of therapeutic escape have focused upon tumor-intrinsic mechanisms of drug resistance with little attention paid to the role of normal host cells in preventing complete tumor eradication. In the present study we implicate co-operative bi-directional signaling between melanoma cells and fibroblasts in the generation of a pro-survival niche that mediates drug resistance. Although bite force is a commonly used metric of feeding performance, other factors such as bite pressure and strike speed are also likely to affect prey capture. Therefore, this study investigated static bite force, dynamic speeds, and predator and prey forces resulting from ram strikes, as well as bite pressure of the king mackerel, Scomberomorus cavalla, in order to examine their relative contributions to overall feeding performance. Such asymmetries may present themselves in hemiplegic patients, prosthetic users, lower limb injuries, limb height and weight discrepancies, or abnormal overground foot rolling. One such measure is to relocate tortoises to protected recipient sites on private lands. While the etiology of ovarian cancer (OC) is not completely understood, evidence suggests that chronic inflammation may promote malignant transformation. However, familial history remains the strongest risk factor for developing OC and is associated with germline BRCA1 mutations, such as the 185delAG mutation. Normal human ovarian surface epithelial cells expressing the 185delAG mutant, BRAT, exhibit molecular and pathological changes that may contribute to OC oncogenesis. In the current study, I sought to determine whether BRAT could promote an inflammatory phenotype by investigating BRAT's impact on the expression of the proinflammatory cytokine, Interleukin-1β (IL-1β).
