Electrical potentials were recorded from scalp of normal male subjects while they were viewing continuously moving sinusoidal gratings. Responses from the occipital cortex were amplified, digitized, stored and then transformed from the time domain to the frequency domain using a microcomputer based Fast Fourier Transform algorithm. High frequency components (Beta Activity) of the resulting power spectrum were analyzed for evidence on neural generators (electrical sources) in the striate cortex driven presumably by transient-cells (Y-cells or Magnocellular) responding to the continuously moving grating. Chronic Myelomonocytic Leukemia (CMML) is a rare myeloid malignancy with a dismal prognosis and no therapeutic options which are capable of altering the natural course of the disease. There remains a significant need for novel therapies that are able to meaningfully improve patient outcomes. In this study we explore the effectiveness of Bromodomain and Extra-Terminal domain protein inhibitor (BETi) combinations in CMML. Preclinical studies in myeloid neoplasms have demonstrated efficacy of BETi. However, BETi demonstrate poor single agent activity in clinical trials. Marine sediment-associated bacteria house many new and exciting novel secondary metabolites. These metabolites can be tested for bioactivity against various types of cancer and fungal, bacterial, and viral infections. In this thesis, we investigated the combination of biosynthetic gene cluster information with mass spectra to perform a chemical profiling of sediment- associated bacteria. Furthermore, we utilized a scoring technique to provide an identification and confidence score to each annotated compound. Mangroves are invading oyster reefs in areas no longer impacted by winter freezes and completely transforming them into mangrove islands. The mechanisms influencing the transition between these two habitat states remain unclear. Here, I test experimentally whether oyster reef (Crassostrea virginica) structure directly facilitates red mangrove (Rhizophora mangle) propagule capture via differences in propagule retention and successful mangrove establishment. In addition to oyster reef structure, I compared physical reef features of size, elevation, and wind-wave exposure to uncover whether these traits influenced mangrove retention. The intrinsically disordered transcription factor and tumor suppressor p53 binds to promoter response element DNA upon cellular stress and activates genes associated with cell cycle arrest, senescence, and apoptosis. Disruption of sequence specific binding to target gene promoters is heavily implicated in human health, where a majority of cancers contain mutations localized to the DNA binding domain (DBD) of p53. p53 DNA binding is regulated by posttranslational modifications, associations with cellular factors, and by an autoinhibitory intramolecular interaction. NRF2 is a redox-responsive transcription factor the directs the antioxidant program and several critical metabolic processes. Mutations in NRF2 or its negative regulator KEAP1 occur in up to one third of non-small cell lung cancers (NSCLCs) and are often associated with resistance to therapy and poor outcomes. In the present studies, murine alleles of the Keap1 and Nrf2 mutations found in human NSCLC were developed and I comprehensively investigated their impact on tumor initiation and progression. A Gait Extraction System (GES) was developed to investigate the accuracy of wearable sensors by producing gait parameters, to be compared against a gold standard motion capture system. Two inertial measurement units (IMUs) were placed in the lower limb region, specifically in the shank region of each leg. Lung cancer is the first cause of cancer-related deaths in both men and women with an overall five-year survival rate of 28%. Although immune checkpoint blockers (ICBs) are currently FDA-approved for the treatment of non-small cell lung cancer (NSCLC), only 17-20% of patients achieve durable responses by the induction of immunologic memory. The lack of response in most patients can be attributed to the tumor-intrinsic or tumor-extrinsic immune resistance mechanisms. Malaria, caused by the apicomplexan Plasmodium spp. , is a major public health issue that impacts over one-third of the world’s population ,with Plasmodium vivax accounting for over 130 million clinical cases annually. The circumsporozoite protein (CSP) is the most abundant molecule on the surface of Plasmodium sporozoites and is considered a leading pre-erythrocytic stage vaccine candidate. CSP is essential for sporozoite maturation, migration, and invasion. Anti-CSP antibodies interrupt sporozoite migration and infection of hepatocytes thus reducing liver-stage burden. Microplastic pollution is an increasingly alarming concern with widespread global distribution in aquatic environments. Filter feeding organisms such as bivalves, have a heightened risk of microplastic ingestion through free floating particulates in the water. This can lead to bioaccumulation moving up through trophic levels to apex predators and humans. Spatial and temporal differences in microplastic abundance were evaluated in the Eastern Oyster, Crassostrea virginica, at six sites within Tampa Bay. Oyster tissue was digested using 30% hydrogen peroxide (H2O2) and microplastics were quantified using Nile Red dye and fluorescent particle excitement. Institutional change initiatives involve executing an intentional strategy to influence teaching practices within a specific institution. Finite element analysis of the spine has become popular over the past several decades. However, the current studies often use minimal specimens for modeling. The purpose of this study was to validate and analyze multiple finite element models with data from in vivo experiments and other finite element studies before future investigations. Lymphatic valves play a critical role in maintaining unidirectional lymph flow and evidence for valve defects have been reported in lymphedema patients. This dissertation explores how multimodal walking impacts quantitative and qualitative aspects of gait for persons with multiple sclerosis (pwMS) experiencing foot-drop. Foot-drop can dramatically impede mobility and clinicians routinely prescribe ankle-foot-orthosis (AFO) and functional electrical stimulation (FES) devices to alleviate its impacts on daily life. The jumping and subsequent aerial behaviors of arboreal lungless salamanders (genus Aneides) had never been investigated or described. The aims of this research were to (1) compare jumping performance within the family Plethodontidae with a special focus on arboreal species from genus Aneides, (2) describe the biomechanics and kinematics of aerial behaviors after a jump or extended fall, and (3) model airflow over the morphology of the most aerially adept salamanders to investigate if they are capable of generating lift. This study demonstrates for the first time that HDAC8 function is critical for MCL survival, and abrogating its activity in human primary NK cells does not interfere with NK IgG antibody directed ADCC therapies ex vivo. Human NK cells, isolated from healthy donors, are highly resistant to HDAC8 inhibitor treatment with PCI-34051. Even at the highest concentration, 20uM, no toxicity was observed. Conversely, MCL cell lines representative of the aggressive MCL subtype, classical MCL, were especially sensitive to PCI-34051, an HDAC8 selective inhibitor, treatment. Neuroblastoma (NB) is a cancer that develops in the neuroblasts. It is the most common cancer in children under the age of 1 year, accounting for approximately 6% of all cancers. The prognosis of NB is linked to both age and degree of cell differentiation. This results in a range of survival rates for patients, with outcomes ranging from recurrence and mortality to high survival rates and tumor regression. Our previous work indicated that PKC-ι promotes cell proliferation in NB cells through the PKC-ι/Cdk7/Cdk2 cascade. Astronauts perform a multitude of tasks in space, both in the spacecraft and outside of the spacecraft. Extravehicular Activity (EVA) refers to any activity that is completed outside of the spacecraft, whether it is work on the actual spacecraft, spacewalks, or conducting sample collections on other planets and moons. To do this safely, a pressurized Extravehicular Mobility Unit (EMU) or commonly known as the spacesuit is used.
