Somites are important mesoderm structures that form in regularly timed intervals from the anterior end of the paraxial mesoderm at the end of gastrulation during embryogenesis. Cells of the ventromedial portion of the somite, the sclerotome, undergo epithelial to mesenchymal transition, surround the notochord, and give rise to connective tissues of the axial skeleton including the vertebrae, tendons, and ligaments. We are interested in understanding how the sclerotome is compartmentalized into the region that makes tendons. Genes expressed in the sclerotome, twist1 and twist2, are likely involved in the downstream differentiation of sclerotome to tendon progenitors. Previous studies have shown …

Rhythm is ubiquitous to human communication, coordination, and experience of music. In this dissertation, I address three empirical questions through three different methodologies, all of which contribute to the growing body of literature on human auditory rhythm processing. In Chapter 2, I present a registered report detailing the results of independent conceptual replications of Nozaradan, Peretz, Missal, & Mouraux (2011), all using the same vetted protocol. Listeners performed the same tasks as in Nozaradan et al. (2011), with the addition of behavioral measures of perception. Antibiotic resistance is a pressing concern that demands further action to reduce and develop new countermeasures. Antibiotic resistance impacts all fields of health and medicine, including dentistry. Many of the preventive and restorative dental procedures are due to a bacterial cause. Knowledge of oral biofilm development mechanisms and the interactions and relationships between oral microbiota is necessary to combat the spread of antibiotic resistance and develop more effective dental treatments. The intertwinement of mother-daughter psyches throughout the early developmental process bonds maternal and filial parties up unto differentiation, at which point the child comes to understand her status as an individual and her mother’s status as a separate entity. However, when trauma is introduced midway through the differentiation process, this psychological phenomenon may be hindered, stunting the advanced personal development of the daughter. Abandoned by loss, she may subconsciously fall victim to repressive defenses, insufficient socialization, and destructive behaviors.

Ovarian follicular development is closely regulated by a multitude of genes and specific non-coding RNAs (ncRNAs), which are typically altered during the oestrous cycle. Identification of the microRNAs (miRNA) expression profiles in sheep ovaries during the oestrous cycle is a prerequisite to further understanding their functional status. This study presents a genome-wide analysis of miRNAs from three groups of Qira black sheep: oestrus (day 0), dioestrus (day 10) and pro-oestrus (day 14). Tendons are fibrous connective tissues that connect skeletal muscle to bone. Functioning primarily to facilitate locomotion, these tissues are rich with collagen and other minor collagens and proteoglycans. Nevertheless, with over 4000 tendons in the human body, the molecular pathways that define different tendons remain largely unknown. Fetal Alcohol Spectrum Disorder (FASD) describes a lifelong neurodevelopmental disability caused by prenatal alcohol exposure that has a devastating impact on individuals, families and communities. The prevalence of FASD is high in some Indigenous communities around the World and the only active case ascertainment prevalence study conducted in Australia found a rate of 19.44 per 100 children in the remote Fitzroy Valley region of Western Australia. Following this study community led FASD prevention activities were implemented under the Marulu (“Worth Nurturing”) Strategy in the Fitzroy Valley.

Progress towards achieving the Sustainable Development Goals (SDGs) has been uneven across and within countries - particularly in Africa, least developed countries, and low-income countries. The COVID-19 pandemic has threatened to reverse much of the progress made towards achieving the SDGs, especially SDG 3, which aims to “ensure healthy lives and promote well-being for all at all ages”.

Background: Occupational therapists focus on caregiver-infant reciprocity, which is influenced by a host of biopsychosocial factors and is predictive of developmental outcomes across domains. It is important for early intervention professions to understand how different forms of reciprocity may predict infant development in salient domains (i.e., language, mobility, and co-occupation). Therefore, the purpose of this study was to investigate associations among related measures of development in and across age, while also exploring how reciprocity influences the acquisition of developmental milestones.

Parents with children who have neurodevelopmental disorders, such as autism spectrum disorder (ASD) and attention deficit hyperactivity disorder (ADHD) experience high levels of parenting stress. The prevalence of ASD is on the rise in the United States. As a result, there has been an increase in the use of treatment modalities, in effort to better understand which modalities have the most positive impact on children and their parents. Human consumption of cannabinoid-containing products during early life or pregnancy is rising. However, information about the molecular mechanisms involved in early life stage Δ9-tetrahydrocannabinol (THC) and cannabidiol (CBD) toxicities is critically lacking. Here, larval zebrafish (Danio rerio) were used to measure THC- and CBD-mediated changes on transcriptome and the roles of cannabinoid receptors (Cnr) 1 and 2 and peroxisome proliferator activator receptor γ(PPARγ) in developmental toxicities. Transcriptomic profiling of 96-h postfertilization (hpf) cnr+/+ embryos exposed (6 - 96 hpf) to 4 μM THC or 0.5 μM CBD showed differential expression of 904 and 1095 genes for THC and CBD, respectively, …

In this dissertation we explore the morphological and neural plasticity underlying vision at different scales—within and between species of Drosophila—to elucidate the role of eye development in the evolution of vision. In chapter 2, we offer a tool to accelerate large-scale research into compound eye morphology, and validate it on the eyes of several insect orders and image media. Then, in chapter 3 we demonstrate the developmental plasticity of eye morphology and neural summation in fruit flies, finding an interesting interplay between the two systems. This study contributes to the body of knowledge in leader development by examining how higher education programs in a female dominated profession assist learners in developing person-related characteristics that support leader development. A ten-part online survey was sent to directors of health information management (HIM) programs accredited by the Commission on Accreditation for Health Informatics and Information Management (CAHIIM). Results suggest an opportunity for improvement in the curriculum for development of person-related characteristics critical for leadership positions. In the interest of the progression of women these results should be taken into consideration.

The granule cells are the most abundant neuronal type in the human brain. Rapid proliferation of granule cell progenitors results in dramatic expansion and folding of the cerebellar cortex during postnatal development. Mis-regulation of this proliferation process causes medulloblastoma, the most prevalent childhood brain tumor. In the developing cerebellum, granule cells are derived from Atoh1-expressing cells, which arise from the upper rhombic lip (the interface between the roof plate and neuroepithelium). In addition to granule cells, the Atoh1 lineage also gives rise to different types of neurons including cerebellar nuclei neurons. In the current study, I have investigated the …

Little research has been conducted on the consequences of maternal use and developmental exposure to ∆9-tetrahydrocannabinol (THC) and cannabidiol (CBD. The purpose of this capstone experience was to gain advanced clinical experience and explore occupational therapy’s role in the NICU and post-NICU settings. Further focus of this capstone was on the feeding concerns and sensory development of infants following a NICU stay. This capstone project was completed at a Level IV NICU and an outpatient speech and occupational therapy clinic in a midwestern city. Background: Questions remain regarding whether genetic influences on early life psychopathology overlap with cognition and show developmental variation.
Methods: Using data from 9,421 individuals aged 8-21 from the Philadelphia Neurodevelopmental Cohort, factors of psychopathology were generated using a bifactor model of item-level data from a psychiatric interview. Five orthogonal factors were generated: anxious-misery (mood and anxiety), externalizing (attention deficit hyperactivity and conduct disorder), fear (phobias), psychosis-spectrum, and a general factor. Genetic analyses were conducted on a subsample of 4,662 individuals of European American ancestry. Organismal development is a process that requires a fine-tuned control of cell fate and identity, through timely regulation of lineage-specific genes. These processes are mediated by the concerted action of transcription factors and protein complexes that orchestrate the interaction between cis-regulatory elements (enhancers, promoters) and RNA Polymerase II to elicit transcription. A proper understanding of these dynamics is essential to elucidate the mechanisms underlying developmental diseases. Cognitive-behavioral therapy (CBT), a first-line treatment for pediatric anxiety disorders, is based on principles of threat learning and extinction. However, CBT does not work sufficiently for up to 40% of clinically anxious youth. The neural and behavioral correlates of conditioned inhibition might provide promising targets for attempts to improve CBT response. During conditioned inhibition, threat and safety cues appear together, forming a safety compound. Here, we test whether this safety compound elicits a reduced fear response compared to pairing the threat cue with a novel cue (novel compound). Autism spectrum disorder (ASD) is a neurodevelopmental disorder that affects one in 66 children in Canada. The contributions of changes in the cortex and cerebellum to autism have been studied for decades. However, our understanding of brainstem contributions has only started to emerge more recently. Disruptions of sensory processing, startle response, sensory filtering, sensorimotor gating, multisensory integration and sleep are all features of ASD and are processes in which the brainstem is involved. In addition, preliminary research into brainstem contribution emphasizes the importance of the developmental timeline rather than just the mature brainstem.
