A Molecular Basis For Erythromycin Sensitivity And Resistance In Escherichia Coli, Harold S. Chittum Dec 1993

A Molecular Basis For Erythromycin Sensitivity And Resistance In Escherichia Coli, Harold S. Chittum

Electronic Theses and Dissertations

The effect of erythromycin on the 50S ribosomal subunit during cell growth has been extensively investigated. Sucrose density gradient analysis of ribosomes formed in the presence and absence of the drug revealed a 50S specific assembly defect is partially responsible for erythromycin's inhibitory effects on wild type cells. Examination of two erythromycin-resistant mutants of E. coli (N281 and N282) revealed that mutant N281 (L22 mutant) but not N282 (L4 mutant) was assembly defective in the presence of the drug, although only at much higher drug concentrations (300 ug/ml vs. 75 ug/ml for wild type cells). The altered genes from each …

A Characterization Of Extractable, Hydroxylated Fatty Acid Bearing Components In Legionella Pneumophila, Jonathan R. Lane Dec 1993

A Characterization Of Extractable, Hydroxylated Fatty Acid Bearing Components In Legionella Pneumophila, Jonathan R. Lane

Electronic Theses and Dissertations

Extraction of the lipids of Legionella pneumophila yields phases unlike those produced from other Gram-negative bacteria. A viscous interface forms between the aqueous (wash) and organic phases. More than half of the hydroxylated fatty acids were found distributed between the aqueous phase and the interfacial material, fractions in which such constituents have not been reported in other Gram-negative species. It was further observed that after the material from the aqueous/interfacial phase was dissolved in methanol or chloroform/methanol (2:1 (V/V)), the addition of acetone would create a white, flocculent precipitate. Analyses showed that the supernatant contained fatty acids that were nonhydroxylated …

A Test Of Self-Directed Practice And Feedback In The Reduction Of A Disturbance In Body Image, Carla Carolyn Adams

Electronic Theses and Dissertations

A disturbance in body image has long been implicated in the development and maintenance of a variety of eating disorders such as anorexia nervosa and bulimia nervosa. Recently, the role of feedback in the reduction of a disturbance in body image has received increased attention with the suggestion that a combination of experimenter-provided accuracy feedback plus self-directed practice may be a powerful element in the reduction of such a disturbance.

This study provides an examination of the role of self-directed practice and feedback in altering a disturbance in body image. Forty-eight women suffering from body image dysphoria participated in a …

Intuition In Decision-Making, Joanne Horton Dec 1993

Intuition In Decision-Making, Joanne Horton

Electronic Theses and Dissertations

This was a two-phase study designed to identify intuitive principals and then to determine if there were common elements of their decision making. The Myers-Briggs Type Indicator was used to identify intuitive principals, and then a sampling of these principals were interviewed. The results showed that the principals with the highest preference scores for intuition differed from the principals with lower preference scores in that they came from supportive environments, were more conscious of using and developing their intuition, drew from experience but combined that with a future orientation, and were more comfortable with risk-taking and change. Recommendations included the …

Development Of Stereoscopic Particle Tracking Velocimetry For Turbulent Flow Field Diagnostics, Veera P. Rajendran Dec 1993

Development Of Stereoscopic Particle Tracking Velocimetry For Turbulent Flow Field Diagnostics, Veera P. Rajendran

Electronic Theses and Dissertations

Stereoscopic particle tracking velocimetry (SPTV) was used to analyze a turbulent submerged jet of Reynolds number (based on the diameter of the nozzle) 8000. SPTV involves tracking the motion of tracer particles over time in a flow in three dimensions. The first part of this work consisted of developing a stereoscopic particle tracking technique using two high-speed video cameras. This included calibration of the cameras with "known" points, and developing an algorithm to automatically match and track particles. The second part was to apply the same to a submerged water jet in order to measure velocities. The velocities were subsequently …

Unusual Structure Of A Human Middle Repetitive Dna, Duminda D. Ratnasinghe Dec 1993

Unusual Structure Of A Human Middle Repetitive Dna, Duminda D. Ratnasinghe

Electronic Theses and Dissertations

The L2Hs sequences are a polymorphic, interspersed, middle repetitive DNA family unique to human genomes. Genomic fingerprinting indicates that these DNAs vary from one individual to another and between tissues of the same individual. Sequence analysis reveals that they are AT-rich (76%) and contain many unusual sequence arrangements (palindromes, inverted and direct repeats). These sequence properties confer on the L2Hs elements the potential to fold into non-B-form structures, a characteristic of recombination hot spots. To test this hypothesis carbodiimide, osmium tetroxide and S$\sb1$ nuclease were used as single-strand specific probes to study a recombinant plasmid, pN6.4.39, containing a single L2Hs …

The Problem of this study was to determine if the cognitive style of elementary school principals affects the principal's evaluation of a teacher when there is a match or mismatch between the principal's and teacher's cognitive style. Using the Productivity Environmental Preference Survey, the dimensions of right brain/left brain hemispheric dimensions of cognitive style were measured for the sample population. The sample population included 40 elementary school principals and 120 elementary school teachers. The 120 teachers were made up of teachers selected by each of the 40 principals as the most effective teachers in the school. The statistical analysis of …

Nitric Oxide Production: A Mechanism For Inhibition Of Chlamydia Trachomatis Replication, Bojun Chen Dec 1993

Nitric Oxide Production: A Mechanism For Inhibition Of Chlamydia Trachomatis Replication, Bojun Chen

Electronic Theses and Dissertations

Chlamydia trachomatis (CT) replicates in macrophages, but is inhibited by IFN-$\gamma$ or LPS. IFN-$\gamma$ and/or LPS induced nitrite production in mouse peritoneal macrophages, macrophage cell lines (RAW264.7 and J774A.1) and McCoy cells. Kinetic studies indicated that peak production occurred 48 hours post-treatment. CT infection itself was insufficient to induce nitrite production, but resulted in enhancement of nitrite production in IFN-$\gamma$-treated cells. Treatment with IFN-$\gamma$ or LPS resulted in significant inhibition of CT replication in these cells. Strong correlation between nitrite production and inhibition of CT replication was observed in RAW264.7 and J774A.1 cells (correlation coefficients: $-$0.93 and $-$0.94, p $<$ 0.001). N$\sp{\rm g}$- monomethyl-L-arginine (L-NMMA) specifically inhibited nitrite production and partially reversed inhibition of CT replication in macrophage cell lines. NOS mRNA was measured in RAW264.7 cells by Northern blot and Dot blot hybridization. Strong correlation between NOS mRNA expression and inhibition of CT replication (correlation coefficient: $-$0.97, p $<$ 0.05) was observed. Anti-TNF-$\alpha$ antibody completely neutralized the biological activity of TNF-$\alpha$ secreted by LPS-treated RAW264.7 cells, yet the antibody neither reduced nitrite production nor restored CT replication. Combination of the antibody and L-NMMA significantly enhanced restoration of CT replication. In peritoneal macrophages, inhibition of CT replication induced by IFN-$\gamma$ was partially restored by L-NMMA or anti-TNF-$\alpha$ antibody. In McCoy cells, inhibition of CT replication induced by IFN-$\gamma$ and LPS was not significantly restored by L-NMMA. Great restoration of CT replication by 1 mM L-NMMA was observed in LPS-treated J774A.1 cells (31%), but not in IFN-$\gamma$-treated cells (5%). Our data indicate that (1) NO production is one of the mechanisms for inhibition of CT replication in IFN-$\gamma$-activated peritoneal macrophages and RAW264.7 cells; (2) NO plays a significant role in CT inhibition in LPS-treated macrophage cell lines, but not peritoneal macrophages; (3) TNF-$\alpha$ may be associated with inhibition, but the mechanism(s) may not involve NO production; (4) NO production may not be the mechanism for CT inhibition in McCoy cells treated with IFN-$\gamma$ and LPS.

A Study Of The Atomic Number Dependence Of Charmed D Meson Hadroproduction, Eric Matthew Aitala

Electronic Theses and Dissertations

We have investigated the nuclear A dependence for the production of D0 and D+ charmed mesons in 500 GeV/c πN interactions. The data were taken by the E791 collaboration at Fermilab's Tagged Photon spectrometer and partially analyzed on a large UNIX/RISC based computer farm located at the University of Mississippi. The measurement of the A dependence for charmed meson production reveals information about the quark distribution within nucleons, specifically the quark distribution function and the. nuclear cross sections. The study also reveals information on the state of the E791 reconstruction and Monte Carlo simulation.

Called To Serve : American Nurses Go To War, 1914-1918., Katherine Burger Johnson

Electronic Theses and Dissertations

World War I resulted in the deaths of over 8,500,000 military personnel and in addition, millions of civilians. There were not enough doctors to provide the necessary medical care for the masses of seriously sick and wounded, and other than in Great Britain, there was little in the way of an organized nursing profession in Europe. Over ten thousand American women served overseas as nurses and nurses' aides from 1914 to 1918, caring for soldiers and civilians of diverse nationalities and proving to be a mainstay in the medical treatment provided for victims of "The Great War." Little has been …

Abilene city fathers perceived the onset of World War II as a potential answer to both economic and population growth goals. Citizens cooperated to secure an Army post, and in February 1941, troops commenced training at Camp Barkeley. With more than 50,000 men eventually serving at the camp, soldiers spent both leisure hours and dollars in the city. Abilenians, eager to protect their virtuous culture, recognized the need for wholesome entertainment for hundreds of unattached young men. Abilene leaders sponsored a women's volunteer association, the Bluebonnet Brigade. Bluebonnets provided companionship for the soldiers while helping the community maintain its moral …

Mechanisms Of T Cell-Mediated Macrophage Activation: Role Of Antigen Specific And Antigen Nonspecific Cognate Interactions, Xiang Tao Jun 1993

Mechanisms Of T Cell-Mediated Macrophage Activation: Role Of Antigen Specific And Antigen Nonspecific Cognate Interactions, Xiang Tao

Electronic Theses and Dissertations

Macrophages play an important role in host antimicrobial immunity and in non-septic inflammatory reactions. Most studies on macrophage activation have focused on the roles of the T cell-produced cytokine, interferon-$\gamma$ (IFN$\gamma)$ and bacterial product, lipopolysaccharide (LPS). T cell-macrophage interaction is a critical step in initiating both specific and nonspecific immune responses to antigenic stimulation. The current study examines the role of cognate T cell-macrophage interaction in activation of macrophage effector functions and induction of macrophage early activation gene expression. Viable resting T$\sb{\rm H}$2 clone cells can activate IFN$\gamma$-primed macrophages to produce reactive nitrogen intermediates (RNI) or express cytostatic activity. The …

Human Lung Mast Cell Tryptase Isozymes: Separation And Examination Of Structural And Functional Differences, Susan S. Little May 1993

Human Lung Mast Cell Tryptase Isozymes: Separation And Examination Of Structural And Functional Differences, Susan S. Little

Electronic Theses and Dissertations

Tryptases are trypsin-like enzymes found in mast cell granules. Although in vivo substrates have not been positively identified, tryptases cleave a limited number of potential physiological substrates in vitro, including high molecular weight kininogen (HMWK) and vasoactive intestinal peptide (VIP). Purified human lung mast cell tryptase (HLT) apparently exists as a tetramer with an M$\sb{\rm r}$ of 135-144 kDa by gel filtration, whereas SDS-PAGE yielded two bands of M$\sb{\rm r}$ 29 Kda and 33 Kda. Tryptases are resistant to inhibition by most natural trypsin inhibitors and display some affinity for heparin. The existence of tryptase isozymes has been implied from …

A total of 52 family members from 22 family units participated in 46 separate, qualitative interviews. The information collected from the interviews was inductively analyzed. Building upon Schlossberg's theory for human adaptation to transitions, seven factors or coping resources emerged from the data that affected the adaptation of …

A Theologically-Rooted Order Of Worship For The Pipeline Road Church Of Christ, Jim Mcdoniel May 1993

A Theologically-Rooted Order Of Worship For The Pipeline Road Church Of Christ, Jim Mcdoniel

Electronic Theses and Dissertations

When a church does not plan its worship, that worship will typically demonstrate a lack of progression, theme, and appropriate movement. As a result, the assembly will neither adequately edify the saints nor praise God. Yet, many churches do little planning for worship. The Pipeline Road Church of Christ in Hurst, Texas, was one of those churches. Little was done to coordinate the activities of the assemblies before this study.

Old Testament Poems For Vow Fulfillment: A Form-Critical And Theological Study, R. Christopher Heard Mar 1993

Old Testament Poems For Vow Fulfillment: A Form-Critical And Theological Study, R. Christopher Heard

Electronic Theses and Dissertations

The most pervasive and enduring contribution of Hermann Gunkel's form-critical program for psalms studies is the insight that certain types of poems are particularly well suited to certain types of cultic occasions. One such occasion is the fulfillment of a vow, mentioned in eight psalms, one poem outside the book of Psalms and several other contexts. To date, however, a separate category of poems for vow fulfillment has not been widely recognized. The form-critical insight mentioned above raises the question whether there exists among Hebrew poems a genre of poems for vow fulfillment. The recurrence in the psalms of phrases …

Visionaries Of The American West : Mari Sandoz And Her Four Plains Protagonists, Lisa R. Lindell Jan 1993

Visionaries Of The American West : Mari Sandoz And Her Four Plains Protagonists, Lisa R. Lindell

Electronic Theses and Dissertations

The authorial reputation of Mari Sandoz has long rested in the shadow of other writers of her era. First of all, Sandoz wrote from and about a relatively remote region of the United States. In addition, she firmly refused to produce popular works at the expense of sacrificing the truth she perceived and wished to express. Consequently, Sandoz has often been classified as a regional writer and her works have been overlooked by many readers and critics. Her status as a woman, her unconventional writing style, point of view, and subject matter, and the blending of historical and fictional elements …
