A Temperature-Sensitive Mutant Of Escherichia Coli Affected In The Alpha Subunit Of Rna Polymerase, Majid Mehrpouyan

A temperature-sensitive mutant of Escherichia coli affected in the alpha subunit of RNA polymerase has been investigated. Gene mapping and complementation experiments placed the mutation to temperature-sensitivity within the alpha operon at 72 min on the bacterial chromosome. The rate of RNA synthesis in vivo and the accumulation of ribosomal RNA were significantly reduced in the mutant at 44$\sp\circ$C. The thermostability at 44$\sp\circ$C of the purified holoenzyme from mutant cells was about 20% of that of the normal enzyme. Assays with T7 DNA as a template showed that the fraction of active enzyme competent for transcription was reduced as a …

The purpose of this study was to determine the differences between industrial and municipal executives' practices and perceptions toward the administrative requirements of a water pollution control system as set forth in a National Pollutant Discharge Elimination System permit. This was a descriptive study that utilized a two-part attitudinal survey designed to obtain the perceptions of the executives (N = 171) of all major industries and all major municipalities in the State of Tennessee who had direct responsibility for the administrative requirements of an NPDES permitted water pollution control system. The electrophysiologic technique (Reuss, L., Proc. Natl. Acad. Sci. USA 82:6014, 1985) was modified to measure changes in steady-state hepatocyte volume during osmotic stress. Hepatocytes in mouse liver slices were loaded with tetramethylammonium ion (TMA$\sp{+}$) during transient exposure of cells to nystatin. Intracellular TMA$\sp{+}$ activity (a$\sp{\rm i}\sb{\rm TMA}$) was measured with TMA$\sp{+}$-sensitive, double-barreled microelectrodes. Loading hepatocytes with TMA$\sp{+}$ did not change their membrane potential (V$\sb{\rm m}$), and under steady-state conditions a$\sp{\rm i}\sb{\rm TMA}$ remained constant over 4 min in single impalements. The purpose of this study was to determine whether the continuous instability of policy decisions relating to the effectiveness of Civilian Conservation Centers and Contract Centers was a result of measurable differences in performance between the two models. Six hypotheses tested differences between types of centers on six quantitative variables which were: average length of stay, job placement, hourly starting wages, educational gains in reading, math, and General Educational Development (GED). To investigate the problem, 30 Civilian Conservation Centers and 17 comparably sized Contract Centers were selected. Although the United States has had a department, bureau, or office of education since 1867, the newly established Department of Education gave education a seat at the Cabinet table. Since that time, four individuals have served as secretaries: Shirley M. Hufstedler, Terrel H. Bell, William J. Bennett, and Lauro F. Cavazos. The purpose of this study was to identify personality types among dental hygiene school administrators and faculty and to determine if correlations existed between leader style, ideologies of leader style, perceived effectiveness, and personality type. Selected demographic variables were also examined. The dimensions of personality investigated were derived from the Myers-Briggs Type Indicator: extroversion, introversion, sensing, intuition, thinking, feeling, judging, and perceiving. The leadership behavior dimensions were the two dimensions of the real and ideal Leadership Behavior Description Questionnaires: consideration and initiating structure. Dopamine (DA) agonist-induced behavioral supersensitivity in the adult rat has served as the standard model for certain of the motor and behavioral side effects associated with long-term exposure to DA agonists in humans. The mechanisms relating receptor events with behavior mediation, however, remain unclear. The striatum of rats progresses through a prolonged and varied postnatal developmental period. Throughout the eighteenth century, interest in the East continued to flourish and incentives to travel there increased. As colonial expansion grew, the middle class began touring abroad with their entire families. Returning home full of excitement from their adventures in exotic places, they felt compelled to publish travel books and to share what they had seen. The materials and interpretations of the East that appeared in the travel books captured the imagination of numerous writers in England and created a large body of literature in which the Orient was the background.

Benzodiazepines are a class of drugs that have several actions including hypnotic features, anti-convulsant features, and anxiolytic features. Midazolam (MZ), a short acting benzodiazepine, is of special interest because of its probable high abuse potential and its efficacy as a preanesthetic.

Tolerance to drug effect has been found for both the hypnotic and anti-convulsant activity of benzodiazepines. Tolerance to the anxiolytic activity of benzodiazepines, however, is less clear. Enoyl-(acyl-carrier-protein) reductase was purified from the phytoflagellate Euglena gracilis. Its purification employed DEAE-Sephacel chromatography, Matrex Orange chromatography, and affinity chromatography using acyl carrier protein (ACP) covalently bound to Sepharose as the affinity ligand. Matrex Orange chromatography resolved two different enoyl-ACP reductases having different characteristics. Euglena gracilis appears to resemble higher plants in the possession of two isoforms of this enzyme. Antibodies specific for the cofactor binding site of NADP (H)-requiring dehydrogenases were obtained. They were isolated from a polyclonal population of antibodies directed against yeast glucose-6-phosphate dehydrogenase by affinity chromatography using chicken liver malic enzyme as the affinity ligand. The purpose of this study was to compare teachers' perceptions of principal effectiveness in secondary schools nominated for the National Secondary School Recognition Program and a randomly selected sample of schools not nominated for the National Secondary School Recognition Program in Tennessee. Evaluation of principal effectiveness was based upon teachers' perceptions as measured by the Audit of Principal Effectiveness, (APE). The data were gathered by mail survey. This research explores violent crimes by women utilizing an interactionist and feminist framework. Detailed interviews were conducted with women serving time for offenses of violence at Huron Valley Women's Facility; a maximum security penitentiary located in Ypsilanti, Michigan. In accordance with other interactionist research on violence, emphasis is on situational definitions and the impact of self concept on the development of violent interactions. This is a history of the medical care insurance system in Ontario. I undertook this study in order to help explain a part of Canadian history that has been severely neglected. This is an historical account of the events that led to Ontario's acceptance of the national medical care insurance plan, and the consequent introduction of the Ontario Health Services Insurance Plan (OHSIP). After introducing OHSIP (1969) Ontario soon amended the program to the current OHIP (1972). A history of these programs would certainly be a logical extension of this study. Latticed triangular-base steel towers have been used as communication structures for a long time. Since these triangular-base towers are economical, they are also being increasingly used as electrical transmission line towers. The legs of these towers generally consist of 60$\sp\circ$ equal leg angles (either 90$\sp\circ$ rolled angles bent to 60$\sp\circ$ or rolled 60$\sp\circ$ angles) which are primarily compression members. The issue was cast in the context of the historical conflicts between the federal government and aboriginal people in Canada over issues of power, culture, language, economics, spirituality, land claims and the environment. Vocational bias can be defined as an implicit assumption that certain careers are more appropriate for one gender than another. Study 1 examined vocational bias from a student's perspective. More specifically, this study addressed the careers to which Canadian undergraduates currently aspire, individuals who influence these choices, students' career confidence, and principally, recent experiences with high school counsellors. In particular, the occupational choices of female students were focused on, especially in the context of previous experience with counsellors. The computer model presented here utilizes a finite difference approximation of the groundwater flow continuity equation in an aquifer to predict water table elevations and the Todd (1980) equation to calculate velocity dependent capture zones for pumping wells. The model presented in this thesis has a series of wells, with variable pumping rates, advance through designated areas of a finite difference grid system discretizing the aquifer. Wells advance through the grid to new nodal configurations and an optimum well configuration solution is found when prescribed conditions are fulfilled. Transmission electron microscopy, scanning electron microscopy, energy dispersive x-ray spectroscopy, and x-ray diffraction have been used to study the zirconium alloys, Zr-1wt%Nb. Zr-2.5wt%Nb, Zircaloy-2, Zircaloy-4, and bulk stoichiometric $\rm Zr(Fe\sb{1-x}Cr\sb{x})\sb2$ alloys, where x = 0, 0.25, 0.45, and 0.75. Both Zr-2.5Nb alloy pressure tubing and Zr-1Nb fuel sheathing contain two main phases, $\alpha$-Zr and $\beta$-Zr. Zr(FeCr)$\sb2$ Laves phase precipitates and Zr$\sb3$Fe intermetallic phase were found in cold-worked Zr-1Nb alloy. Only Zr$\sb2$Fe precipitates were found, however, in Zr-2.5Nb pressure tubing, which contains more Fe and Cr than Zr-1Nb. Calcineurin (CaN), a calmodulin stimulated protein phosphatase is a heterodimer composed of a 61kD catalytic subunit and a 19kD regulatory subunit. The larger subunit has three binding sites for transition metal ions and also contains the intrinsic metal ions, Zn$\sp{2+}$ and Fe$\sp{3+}$. The smaller subunit contains four "EF-hand" Ca$\sp{2+}$ binding sites. All ten of calcineurin's cysteine residues are found on the catalytic subunit. This study explores the metal ion dependence of CaN using Ca$\sp{2+}$, Mg$\sp{2+}$, Mn$\sp{2+}$ and Ni$\sp{2+}$ as well as its thiol chemistry using Ellman's reagent, iodoacetate, iodoacetamide, glutathione and a novel thiol reagent introduced by this study, 1-p-chlorophenyl-4,4-dimethyl-5-diethylamino-1-penten-3-one …

Surface enhanced Raman (SERS), surface enhanced Resonance Raman (SERRS) of mono and multilayer films of three N,N$\sp\prime$-unsymmetrical dialkyl-3,4:9,10-perylenebis (dicarboximides)s, namely N-octyl-3,4:9,10-perylenetetracarboxylic monoanhydride(Oc-PTCDO), N-octyl-3,4:9,10-perylenetetracarboxylic monoimide(Oc-PTCDNH) and N-octyl, N$\sp\prime$-isobutyl-3,4:9,10-perylene-bis (dicarboximide) (PBDC) have been studied and vibrational analyses have been attempted. SERS and SERRS of evaporated thin films of perylene, 3,4:9,10 perylene tetracarboxylic dianhydride (PTCDA) and 3,4:9,10 perylene tetracarboxylic N,N$\sp\prime$ bis (methyl) diimide (PTCDIMe) on tin sphere, Ag island and Au island films are used to characterize the vibrations of the three unsymmetrical perylene derivatives. The purpose of the present study was to fully characterize the force-interval relationship of the right ventricular papillary muscle from chronic diabetic rats, and, to more closely examine the alterations in the beta phase reported previously. The condition of chronic diabetes resulted in several alterations in the force-interval relationship, primarily in the beta phase of force development. Alterations in the percent contribution of beta to the total amplitude and the rate and time constants of the beta phase were identified and suggest alterations in the capacity of the diabetic tissue to transport calcium through the exchange compartment of the sarcolemma …

The study area lies within the western section of the Elzevir terrane, in the Central Metasedimentary Belt. A supracrustal sequence of carbonates and clastic metasediments, interdigitating with metavolcanics occur in this area, intruded by a series of dykes. The majority of minor intrusions are mafic in composition; a minor number of felsic samples have been identified. The dykes have been metamorphosed to middle and upper amphibolite facies of low-pressure type, transitional into moderate pressures. This study examined the effects of language impairment and age on the pattern of performance on the scales of the Kaufman Assessment Battery for Children (K-ABC). Four groups of preschool children were examined: 14 four-year-old children and 10 five-year-old children with a clinical diagnosis of expressive and receptive language impairments; 14 four-year-old children and 19 five-year-old children referred for reason other than language impairment. Subjects were matched on age, sex, SES, and presence of behavior problems. The results provided some support for Telzrow's (1984) predicted pattern of performance. The present study examined epistemic style predispositions associated with individual differences in responsiveness to style-consistent and style-inconsistent persuasive communications. Two hundred and fifty-seven undergraduate students participated. In Session 1, subjects were given (a) a questionnaire measuring their attitudes toward two relevant issues, student access to course evaluations and equal automobile insurance rates for men and women in Ontario, (b) the Psycho-Epistemological Profile (PEP) to assess their characteristic epistemic styles (rational, empirical, or metaphorical), and (c) a brief demographic survey. Juries have been a long standing institution within the Canadian criminal justice system. However, there is to be found very little research in the area. This exploratory study seeks to examine the selection of juries from the lawyer's perspective, both Crown and defence, and the factors that the lawyers find as being significant to them. The research is conducted from a symbolic interactionist perspective of the study of the definition of the situation, of selecting a Jury for a criminal trial. This study investigated the relationship between preservice teachers' mathematical and language arts learning histories and beliefs about their (1) attribution of students' achievement, (2) self-efficacy as future teachers, and (3) other subject-/topic-related perceptions. The belief most closely linked to the learning history was the self-efficacy as future teachers. The group with a less favourable mathematical learning history and a more favourable language arts learning history believed they were significantly less able to influence students' effort, interest, and achievement in mathematics compared to language arts.
