Investigation Of Histone Lysine Methylation In Stem Cell Differentiation Using Inhibitor Peptide, Wendy Zhu

In an effort to expand the histone code, we examine a novel site of methylation on lysine 43 of histone H2B. In mouse embryonic stem cells (ESCs), KDM5b acts as the lysine demethylase for H2BK43me2, diminishing this histone mark as cells differentiate. We utilize a synthetic peptide mimetic corresponding to amino acids 37-49 of histone H2B in order to sterically inactivate KDM5b enzyme. The addition of inhibitor peptide into culture enhanced stem cell differentiation, upregulating cell cycle and neural-specific markers while downregulating the expression of pluripotency genes. Global gene analysis patterns of peptide-treated ESCs were representative of differentiated cell populations. …

The preimplantation period of development represents the highest interval of embryonic loss throughout pregnancy. It is therefore imperative that we elucidate the mechanisms involved in regulating preimplantation embryonic responses to stress and that govern development. The MAPK pathways are involved in both responding to environmental stress and regulation of development throughout embryogenesis, and are therefore good candidates to study the mechanisms involved in preimplantation embryonic adaptation to stress and development. The preimplantation embryo culminates in the development of a fluid filled structure called the blastocyst. Effects of a low protein (LP) diet during gestation on the metabolism of rat offspring have been well characterized and leads to glucose intolerance in adulthood. It is unknown how LP impacts endocrine pancreas development in the mouse, or whether this affects future β-cell plasticity. Streptozotocin (STZ) – induced β-cell injury has been demonstrated to be followed by β-cell regeneration in young animals, but the mechanism(s) of regeneration are not clear. Insects migrate in order to escape deteriorating habitats and colonize new ones. However, long distance flight may utilise resources that would otherwise be allocated to reproduction and it has been postulated that migrants have adaptations to reduce the cost of migration to future reproduction. I determined the suitability of the rearing method used extensively for previous studies on Pseudaletia unipuncta adult moth eco- physiology that is primarily based on artificial diets and environmental conditions to mimic cues of habitat quality. An emerging class of theories concerning the functional structure of the brain takes the reuse of neural circuitry for various cognitive purposes to be a central organizational principle. According to these theories, it is quite common for neural circuits established for one purpose to be exapted (exploited, recycled, redeployed) during evolution or normal development, and be put to different uses, often without losing their original functions. In ―Star Trek‖, Scotty suggests that Transwarp beaming is ―like trying to hit a bullet with a smaller bullet, whilst wearing a blindfold, riding a horse‖ (Abrams, 2009). The study of social media faces similar challenges because new tools are developed at a rapid pace and existing tools are constantly being updated with new features, policies, and applications.
