Relationship Between Gene Frequencies And Indices Of Physiological Stress In The Estuarine Gastropod, Thais Haemastoma., David Wendell Garton

LSU Historical Dissertations and Theses

The relationship between individual energy budgets and genetic variability was examined in Thais haemastoma acclimated to 7.5, 20 and 35('o)/oo S at 21(DEGREES)C. Energy budgets, after the equations of Crisp (1971), were determined by measuring rates of caloric ingestion, absorption efficiencies, oxygen consumption and ammonia and primary amine excretion. Genotypes at six polymorphic loci coding for soluble enzymes, of 25 total loci assayed, were determined using starch gel electrophoresis. The polymorphic loci examined were those encoding for (alpha)-L-amino peptidase (LAP), phosphoglucomutase (PGM), glucose-6-phosophate dehydrogenase (G6PDH), a non-specific esterase (Est) and two dipeptidases (L-leucyl-L-proline and L-phenylalanyl-L-proline dipeptidases). The genetic variability observed …

Biological, Ecological, And Morphological Investigations Of The Rice Water Weevil, Lissorhoptrus Oryzophilus Kuschel, On Two Rice Genotypes., Gary Leonard Cave

LSU Historical Dissertations and Theses

The rice water weevil, Lissorhoptrus oryzophilus Kuschel, is the most important insect pest of rice, Oryza sativa (L.), in the United States. Although this insect has been associated with O. sativa since the introduction of this plant species into the United States in 1685, some aspects of its biology remain obscure. Larvae of L. oryzophilus have four instars, based on measurements of head-capsule widths of field-collected larvae in 1981 and 1982. The head-capsule widths of each instar were: I, 0.14 to 0.18 mm; II, 0.20 to 0.26 mm; III, 0.28 to 0.38 mm; IV, 0.40 to 0.60 mm. Further studies …

Inflammation And Leukocyte Responses Induced By Subcutaneous Turpentine Inoculation Of Young Alligator Mississippiensis., Margarita Rosa Mateo

LSU Historical Dissertations and Theses

Turpentine-induced skin reactions in young Alligator mississippiensis kept at 25(DEGREES)C were employed to study reptile inflammatory response. Skin lesions, harvested at intervals from 4 hours to 30 days post-inoculation exhibited no gross changes until day 24-26, when superficial skin necrosis and sloughing occurred. Early responses of congestion and dermal edema (4-8 hours) were seen by light microscopy, followed by necrosis and granulocyte migration (1-3 days). There was later predominance of monocytic cells, including vacuolated macrophages (7 days - 30 days). At 30 days post-inoculation, central dermal zones of necrotic debris were surrounded by palisades of vacuolated multinucleated giant cells and …

Evidence For An Outer Membrane - Association Precursor Of The Extracellular Protease Produced By Serratia Marcescens., Laura Robin Liss

LSU Historical Dissertations and Theses

Serratia marcescens LRL781, a gram-negative bacterium, produces an extracellular protease. Evidence is provided for an outer membrane-associated, larger molecular weight, enzymatically inactive precursor of the protease. Antiserum was prepared against the extracellular protease, which was purified from the supernatant fraction of S. marcescens LRL781 cultures. Column adsorption of the antiprotease serum was performed using a slurry of live cells and cellulose. The resulting effluent was serologically inactive against protease preparations and chromium chloride-coupled protease-rabbit erythrocytes. Whole washed cells agglutinated in the presence of the antiserum. Cells were uniformly labeled with ferritin conjugated goat anti-rabbit IgG, indicating that the protease precursor …

Water Budget, Benthological Characterization, And Simulation Of Aquatic Material Flows In A Louisiana Freshwater Swamp., Fred Hal Sklar Jan 1983

Water Budget, Benthological Characterization, And Simulation Of Aquatic Material Flows In A Louisiana Freshwater Swamp., Fred Hal Sklar

LSU Historical Dissertations and Theses

A two-year survey of the aquatic plants and invertebrates in the Des Allemands Swamp was designed to determine the ecological effects of altered hydrologic regimes. The aquatic invertebrate community in an impoundment was compared with a control site subject to "normal" overflow flooding and a crawfish farm where high rates of water exchange are maintained by a system of pumps and artificial levees. Swamp hydrology for the years 1914-1980 was modeled using Thornthwaite and Mather components. Time-series analysis found no evidence of any long-term cyclical behavior. High evapotranspiration and drainage during summer decrease water levels to zero in natural swamp …

The Beta-Fructofuranosidase Of Kluyveromyces Fragilis And Its Use In Inulin Hydrolysis., Wesley Earl Workman

LSU Historical Dissertations and Theses

Inulin, a polyfructan, is found as the reserve carbohydrate in the roots and tubers of various plants. The (beta)-fructofuranosidase (inulase) from the yeast Kluyveromyces fragilis is of interest because of its industrial potential in fructose syrup and alcohol production from inulin containing plants. The enzyme was purified and characterized with respect to its physical and kinetic parameters. The relationship between the enzyme and the yeast cell wall was also investigated. It was found that the inulase of Kluyveromyces fragilis could be immobilized in the yeast cells by glutaraldehyde treatment. Although the exact nature of the immobilization is not fully understood …

Systematics, Redescription, And Geographic Variation Of The Blackside Darter, Percina Maculata (Pisces; Percidae)., Eugene Clifton Beckham Iii

LSU Historical Dissertations and Theses

A systematic study of the North American percid fish Percina (Alvordius) maculata (Girard) was undertaken to determine the extent and pattern of geographic variation, subspeciation, and sexual dimorphism. A brief nomenclatorial history is presented, the species is redescribed, and variation, distribution, habitat, and subgeneric relationships are discussed. Other than dorsal fin pigmentation, absence of modified midventral scales in females, and limited intrapopulational sexual dimorphism in a few morphometric characters, no variation in most of the thirty-nine characters examined is present between males and females. Morphometrics exhibited no pattern of geographic variation; however, meristic characters revealed a major trend in geographic …

Some Relationships Between Crop Damage Inflicted By The Sugarcane Borer, Diatraea Saccharalis (F.), And Selected Plant Diseases (Louisiana)., Emmanuel Oludele Ogunwolu

LSU Historical Dissertations and Theses

A 3-fold difference (P < 0.05) was found in a 5-replication split plot comparison of % yield loss due to ratoon stunting disease (RSD) alone in contrast to that due to the disease in combination with sugarcane borer (SCB), Diatraea saccharalis (F.), damage in 1st stubble sugarcane. The research plan utilized differential levels of SCB damage as whole plots and a factorial combination of 2 levels of RSD (0 vs 63%) and 2 types of seed cane ( 35% SCB bored internodes) as subplots. Based on the plant cane data, a highly significant negative correlation r = -0.82, was found between SCB damage and cane weight. Loss in sugar production due to 55% SCB bored internodes was 24.4%. The association of simulated or actual SCB injury with stalk rots, particularly Colletotrichum falcatum Went, caused from 9.2-34.1% reduction in germination, and as much as 2.5 metric tons/ha loss in sugar production. Excepting SCB control, a survey spanning 2 years of sugarcane management practices among producers in 3 geographic areas of Louisiana showed substantial variability in the efficacy of control tactics applied against weeds and RSD, with effective suppression of weeds in sugarcane being the most inconsistent and difficult to achieve. These results indicate the need for an integrated approach to managing several components of pest complexes, disease, weeds, and insects in Louisiana sugarcane.

Bacteriological And Toxigenic Studies On Vibrio Species Isolated From The Blue Crab (Callinectes Sapidus) And The Purification Of Toxic Factors Produced By Vibrio Fluvialis (Louisiana)., Susan Lynne Payne

LSU Historical Dissertations and Theses

Eleven cases of cholera occurred in Louisiana in 1978 and except for an isolated case in Texas in 1973, this was the first cholera outbreak in the United States since 1911. The source of the infectious agent was traced to the consumption of crabs, a finding which had adverse effects on the crab industry in Louisiana. During 1979 and 1980 blue crabs were examined bacteriologically for the presence of Vibrio cholerae, the etiological agent of Asiatic cholera, and other pathogenic Vibrio species. Four hundred-eighteen V. cholerae isolates were recovered from crab feces and 144 V. cholerae were recovered from hemolymph …

Of the remaining 8 species, Cyclocoelum brasilianum, Plagiorchis maculosus, Stephanoprora denticulata, and Eurytrema lubens …

The Effects Of Acute Dehydration On The Anaerobic Threshold During Incremental Cycle Ergometer Exercise., Penelope England Jan 1983

The Effects Of Acute Dehydration On The Anaerobic Threshold During Incremental Cycle Ergometer Exercise., Penelope England

LSU Historical Dissertations and Theses

Much of the previous research concerning dehydration and performance has centered around prolonged submaximal work while seemingly little attention has been focused upon short term incremental exercise. Numerous investigators have suggested that dehydration may alter carbohydrate metabolism in muscle. Theoretically, altered glycolytic flux in exercising muscle could result in a change in the rate of blood lactate (La) accumulation, and thus shift the anaerobic threshold. Anaerobic threshold (AT(,La)) has been formally defined as the oxygen uptake (V(,O(,2))) at which there is a systematic increase in blood La. Additionally, the AT(,La) has been estimated from a nonlinear increase in expired ventilation …

In oyster homogenates, the D values in minutes were 4.7 at 49(DEGREES)C, < 0.35 at 54(DEGREES)C, < 0.35 at 60(DEGREES)C, < 0.33 at 66(DEGREES)C, < 0.30 at 71(DEGREES)C, < 0.29 at 77(DEGREES)C, and < 0.24 at 82(DEGREES)C. In shrimp homogenates, the D values in minutes were 9.17 at 49(DEGREES)C, < 0.43 at 54(DEGREES)C, < 0.39 at 60(DEGREES)C, < 0.32 at 66(DEGREES)C, < 0.31 at 71(DEGREES)C, < 0.30 at 77(DEGREES), and < 0.28 at 82(DEGREES)C. Whole oysters, inoculated with 10('6) V. cholerae/gram, were cooked in boiling water (100(DEGREES)C) or in hot oil (191(DEGREES)C) for one minute. Whole shrimp, inoculated with 10('6) V. cholerae were cooked in boiling water (100(DEGREES)C) or in steam (100(DEGREES)C) for 10 minutes. Vibrio cholerae was not isolated from the cooked oysters or shrimp. Sterile and unsterile shrimp, oyster, crab, and crayfish meat homogenates were inoculated with 10('7) V. cholerae/g. The homogenates were treated with 0, 25, 50, or 100 krad and stored at 4(DEGREES), 0(DEGREES), or -8(DEGREES)C for 21 days. No V. cholerae were isolated from any of the homogenates treated with 100 krad. Vibrio cholerae was able to survive longer in the sterile oyster, crab and crayfish homogenates stored at 4(DEGREES) and 0(DEGREES)C than in the unsterile homogenates stored at the same temperatures. The organism was able to survive longer in the unsterile shrimp homogenates stored at 4(DEGREES) and 0(DEGREES)C than in the sterile homogenates stored at the same temperature. At -8(DEGREES)C, survival time was usually longer in the unsterile seafood homogenates than in the sterile seafood homogenates stored at -8(DEGREES)C. Due to the retardation of competitive inhibition the organism was usually able to survive longer in the unsterile homogenates stored at -8(DEGREES)C than in the unsterile homogenates stored at 4(DEGREES) or 0(DEGREES)C.

Sodium Ion Transport Regulation By A Serotonin Stimulated Adenylate Cyclase System In Freshwater Mussels., John Iver Scheide Jan 1983

Sodium Ion Transport Regulation By A Serotonin Stimulated Adenylate Cyclase System In Freshwater Mussels., John Iver Scheide

LSU Historical Dissertations and Theses

Freshwater bivalves increase Na influx when the mussels were injected with dibutyryl cAMP, serotonin (5-HT), or 5-HT (10('-4) M/l) was added to the bath; a response similar to a sodium depleted mussel. Serotonin stimulated Na influx kinetics (K(,m) = 0.09 mM/l and J(,max) = 4.33 (mu)eq (g dry tissue (.) hr)('-1) were similar to Na depleted mussels. Stimulated Na influx (by dibutyryl cAMP or 5-HT) was inhibited by amiloride (5 x 10('-4) M/l) and the amiloride inhibition was reversible. Serotonin also changes water balance; the 5-HT stimulated mussels gain 0.36 ml H(,2)O (g dry tissue (.) hr)('-1) above controls. Serotonin …
