Genetically Modified Mosquitoes: A New Strategy To Control Mosquito Borne Diseases, Padet Siriyasatien, Usavadee Thavara

The Thai Journal of Veterinary Medicine

Mosquito borne diseases remain a major human and animal health concern in many areas of the world. Chemical control strategy faces a number of problems including the development of insecticide resistance in mosquitoes, the impact of insecticides on human and animal health and the environment and the constant inability to control mosquitoes effectively. Advances in the knowledge about mosquito-pathogen relationships and the molecular biology of mosquitoes make it now possible to produce mosquito strains that are unable to transmit various parasites or viruses. Strategies of using the genetically modified mosquitoes to control mosquito borne diseases are discussed here.

Although cattle develop humoral immune responses to Shiga-toxigenic (Stx+) Escherichia coli O157:H7, infections often result in long-term shedding of these human pathogenic bacteria. The objective of this study was to compare humoral and cellular immune responses to Stx+ and Stx- E. coli O157:H7. Three groups of calves were inoculated intrarumenally, twice in a 3-week interval, with different strains of E. coli: a Stx2-producing E. coli O157:H7 strain (Stx2+ O157), a Shiga toxin-negative E. coli O157:H7 strain (Stx- O157), or a nonpathogenic E. coli strain (control). BACKGROUND: National blood donation screening for West Nile virus (WNV) started in June 2003, after the documentation of WNV transfusion-associated transmission (TAT) in 2002.

STUDY DESIGN AND METHODS: Blood donations were screened with investigational nucleic acid amplification assays in minipool formats. Blood collection agencies (BCAs) reported screening results to state and local public health authorities. Donor test results and demographic information were forwarded to CDC via ArboNET, the national electronic arbovirus surveillance system. State health departments and BCAs also reported suspect WNV TATs to CDC, which investigated these reports to confirm WNV infection in blood transfusion recipients in the absence …

At least, 4 mosquito species, Culex tritaeniorhynchus, Culex gelidus, Anopheles vagus and Mansonia uniformis were identified on a pig farm in Nakorn Pathom, Thailand between March 2004 to February 2005. C. tritaeniorhynchus was found to be the predominant species (88.387.63%). To determine whether C. tritaeniorhynchus could serve as a mechanical vector for PRRSV transmission, 2 experiments were conducted on: the duration of PRRSV within the mosquitoes and PRRSV transmission from the PRRSV-infected pigs to naive pigs by the infected mosquitoes. The study developed a thoracoscopic-guided lung biopsy technique that was able to be performed on the right or left lung lobes of dogs. Ten experimental dogs weighing 6-20 kilograms were anesthetized, ventilated and positioned in a lateral recumbency. Either the left or right hemithorax was approached. Rapid Isolation Of Dna From The Mucus Of Asian Arowana (Scleropages Formosus, Osteoglossidae), Nantarika Chansue

Owing to arowana being listed in CITES, DNA diagnostics should not rely on conventional methods of obtaining tissue samples. A non-invasive method of obtaining material containing DNA has been developed. Mucus samples for DNA isolation can be obtained by cotton swabbing the arowana body surface with subsequent purification of DNA fragments by using phenol/chloroform. DNA sample quality is assessed by UV absorbancy at wave lengths of 260 nm and 280 nm, as well as by calculating the absorbancy ratio involving 260 nm/280 nm results in order to determine DNA sample quality and quantity. Morphological Changes In The Oviduct Of Culling Replacement Gilts, Paisan Tienthai, Kriengyot Sajjarengpong, Padet Talummuk

The Thai Journal of Veterinary Medicine

The aim of this study was to investigate the morphological changes of the oviduct from 30 culling replacement gilts and 6 control sows. Blood samples and female genital organs were taken immediately after slaughter. Post-mortem examinations were made on the genital organs and oviductal samples, i.e. utero-tubal junction (UTJ), isthmus and ampulla, were collected and fixed for histological analysis. Observations indicated that the reasons for culling the gilts were 'no estrus' (68%) and 'repeat breeding' (32%). The greatest pathological changes (67%) were found in repeat breeding gilts, whereas 52% of no estrous gilts were at prepubertal phase. The female hormonal …

Enterotoxigenic Escherichia coli (ETEC) strains that produce K88 (F4) + fimbria are important causes of diarrhea and post-diarrheal septicemia in swine. ETEC O8:K87, a serotype represented by a number of these strains, is typically serum resistant. Strain-specific antibodies are known to activate alternative C pathway-mediated killing of other serum-resistant E. coli [Hill, A.W., Shears, A.L., Hibbitt, K.G., 1978. Background: Oral rabies vaccination programs have been implemented to control the spread of wildlife rabies in the United States. However, current surveillance systems are inadequate for the efficient management and evaluation of these large scale vaccine baiting programs. With this in mind, a GIS-based rabies surveillance database and Internet mapping application was created. This surveillance system, RabID, provides a new resource for the rapid mapping and dissemination of data on animal rabies cases in relation to unaffected, enzootic, and baited areas where current interventions are underway.

BACKGROUND: The Silver coat color, also called Silver dapple, in the horse is characterized by dilution of the black pigment in the hair. This phenotype shows an autosomal dominant inheritance. The effect of the mutation is most visible in the long hairs of the mane and tail, which are diluted to a mixture of white and gray hairs. Herein we describe the identification of the responsible gene and a missense mutation associated with the Silver phenotype.

Panulirus argus Virus 1 (PaV1) is the first virus known to be pathogenic to a wild lobster. It infects the Caribbean spiny lobster P. argus from the Florida Keys, and has a predilection for juveniles. The monitoring of the virus in wild populations and study of its behavior in the laboratory require the development of reliable diagnostic tools. A sensitive and specific fluorescence in situ hybridization (FISH) assay was developed for detection of PaV1. The structures of the canine, rabbit, bovine and equine EIF2AK2 genes were determined. Each of these genes has a 5' non-coding exon as well as 15 coding exons. All of the canine, bovine and equine EIF2AK2 introns have consensus donor and acceptor splice sites. In the equine EIF2AK2 gene, a unique single nucleotide polymorphism that encoded a Tyr329Cys substitution was detected. Regulatory elements predicted in the promoter region were conserved in ungulates, primates, rodents, Afrotheria (elephant) and Insectifora (shrew). The leading genetic cause of infant mortality is spinal muscular atrophy (SMA), a clinically and genetically heterogeneous group of disorders. Previously we described a domestic cat model of autosomal recessive, juvenile-onset SMA similar to human SMA type III. Here we report results of a whole-genome scan for linkage in the feline SMA pedigree using recently developed species-specific and comparative mapping resources. We identified a novel SMA gene candidate, LIX1, in an ~140-kb deletion on feline chromosome A1q in a region of conserved synteny to human chromosome 5q15. Twenty cadaver canine forelimbs, 9 right and 11 left, were used to evaluate the most effective radiographic position that can enhance the cranial aspect of the medial coronoid process (MCP) of the ulna. A lead pellet (1 mm) was buried to represent the area representing the cartilegenous fragment in the canine patient in the fragmented medial coronoid process (FMCP). Classical swine fever virus (CSFV), serum neutralizing antibody titers of 854 sows were determined using a neutralizing peroxidase linked assay (NPLA). Blood samples were collected one-week post-partum from 1st- 6th parity sows, from 30 swine fever-free farms. The farms were divided according to their vaccination programs, 10 farms for each program. Program A sows had been vaccinated 3 weeks pre-partum (288 samples). Program B sows had been vaccinated 3 weeks post-partum (276 samples). Nuclear transfer from adult somatic cells is an advanced reproductive biotechnology. This technique can be applied in animal production for producing a high genetically identical copy for endangered species conservation and for therapeutic cloning for human medicine. Our three-year preliminary study indicates the possibility of somatic cell cloning by using the rabbit as model of technical development. Later, interspecies cloning in wild cats and domestic cats was performed successfully in terms of cloned embryo production. Meanwhile, goat cloning served as the model for farm animals.

Porcine small intestinal submucosa biomaterial was used to repair corneal staphyloma in 10 dogs. There was no evidence of leakage of aqueous humor and graft rejection following PSIS grafting. Anterior synechia was presented in 25-50% of the corneal area. Granulation tissue vanished within 50 days of the operation. Even though scar formation and anterior synechia were observed at the end of the study, all dogs had vision.

Progesterone Receptors And Proliferation In The Bitch Uterus During Different Stages Of The Oestrous Cycle, Sayamon Srisuwatanasagul, Kongkiat Srisuwatanasagul, Sudson Sirivaidyapong

Immunopresence of the progesterone receptors (PR) and proliferative activity in the bitch uterus were studied during different stages of the oestrous cycle. Uterine samples were collected at 4 different stages of the oestrous cycle; prooestrus, oestrus, dioestrus and anoestrus. Immunohistochemistry was used to evaluate the localization and to semiquantitate PR and uterine proliferation by using monoclonal antibodies to the PR and the proliferative marker, Ki-67 respectively. The results showed that PR and Ki-67 levels were highest during prooestrus and oestrus while they were low at other stages. However, a significant difference was observed only in the surface epithelium for Ki-67 …

Observations On The Structures Of Red Blood Cells, White Blood Cells And Platelets In Some Felis Spp. And Panthera Spp., Wimon Pothiwong, Kamol Sakulwira, Pakorn Prachammuang, Siripen Vethchagarun

The Thai Journal of Veterinary Medicine

Blood cells and platelets from twenty-seven animals belonging to two genera of the Family Felidae, were studied using both light and transmission electron microscopes (TEM). Generally, blood cells can be categorized into red blood cells, white blood cells and platelets. The red blood cells or erythrocytes from these species were round and had neither a nucleus nor organelles. They showed a central pale area, Heinz bodies and rouleaux formation. The white blood cells could be divided into two groups; granulocytes and agranulocytes.
