Anthropogenic nutrient pollution is one of the most serious threats facing aquatic systems in modern times. Pollution with compounds containing nitrogen and phosphorous encourages rapid development of autotrophic biomass, or blooms, that severely degrade the quality of an aquatic system both as a habitat for organisms and as a water resource for human use. Microalgae can be grown on municipal wastewater media to both treat the wastewater and produce feedstock for algae biofuel production. However the reliability of treatment must be demonstrated, as well as high areal algae productivity on recycled wastewater media and efficient sedimentation harvesting. This processes was studied at pilot scale in the present research.

In river systems, there are many factors that impede or facilitate algal standing stock and therefore impact primary production in these environments. I am particularly interested in the influence of edges in riverine systems and how these geomorphic features affect the available surface area for algal production. This study investigates the middle Rio Grande system near Albuquerque, New Mexico, USA. The middle Rio Grande is a turbid, partially braided lotic ecosystem. Algal biofuels have the potential to provide a scalable source of renewable fuels in the near future. The high nutrient use in algae cultivation and its recovery and recycling is one of the challenges that may limit the scalability and sustainability of algal biofuels. The present study evaluates the use of Hydrolysate obtained after Flash Hydrolysis (FH) of Scenedesmus at 280 C as a nutrient source for microalgae cultivation. FH Hydrolysate nutrient recycling was compared with low temperature batch Hydrothermal Liquefaction (HTL) nutrient recycling. The advent of photosynthesis facilitated the evolution of aerobic life on Earth. However, species such as Prototheca wickerhamii and Plasmodium falciparum, among many others, have lost photosynthesis and opted for a free-living/parasitic lifestyle. Despite this loss, these species have retained the plastid for its metabolic pathways, without which they would die. Polytomella is a nonphotosynthetic free-living alga, closely related to the photosynthetic model organism Chlamydomonas reinhardtii, and has been shown to lack a plastid genome. I set out to determine Polytomella plastid metabolic pathways using bioinformatics to look for mRNA and DNA homologous sequences matching pathway enzymes in model organisms. …

While treating domestic wastewater, algae can be grown and harvested for biofuel production. Water is a vital resource and it is imperative to conserve and reuse as much as possible. Several pilot and lab scale experiments were conducted to further research into a full scale wastewater treatment and biofuel production facility. This thesis will include these topics: nitrogen and phosphorus concentrations of clarified wastewater pond feed, nitrogen and phosphorus transformations by algae, and the potential of algae cell disruption technologies to increase nutrient solubilization.

The flow of energy through an ecosystem is largely dependent how productivity moves through organisms of various sizes. The use of algae as a feedstock for hydrous pyrolysis has a high potential for producing biofuels. The non-food nature of algae in addition to the various advantages associated with the hydrous pyrolysis process makes this combination for the production of biofuels of high interest. However, current results from algae processing have alluded to some challenges: byproducts arising from the thermal transformation of carbohydrates and proteins, which become incorporated in the oil fraction, result in high oxygen and nitrogen contents of the oil. Beach fouling is a problem in the Great Lakes. We surveyed beaches along the southwestern shore of Saginaw Bay to determine the spatial and temporal distribution of beach fouling (muck), muck composition, and any relationship between muck accrual and weather conditions. Muck deposition was greater towards the end of the growing season. The Bay City Recreation Area (BCRA) had the highest frequency of muck presence. BCRA muck depth was much greater in 2010 and 2011 than in 2012 and 2013. No sites differed in muck depth in 2012, but in 2013 the two southernmost sites tended to have greater muck …

In the Arctic, brown algae (kelps) and seaweeds are ecologically important: providing habitat, protection, and nutrients for invertebrate and vertebrate species living in nearshore environments. Migrations of biota between the North Pacific and North Atlantic Oceans have occurred periodically during Earth’s history leading to colonization of the Arctic Ocean. Around 3.5 Mya the “Great Trans-Arctic Biotic Interchange” occurred and the Laminariales order of kelp, thought to be of North Pacific origin, underwent a massive radiation and speciation event around the Arctic Ocean. The objective of the first study was to quantify the effects of feeding Schizochytrium sp. microalgae (SP-1, Alltech, Inc., Nicholasville, KY) on milk fat and DHA content. Eight cows were fed: 0, 100, 300, or 600 g of algae per day. Fat percentage was greater (P < 0.05) for cows on treatments 0 g and 100 g than for cows on treatments 300 g and 600 g (P < 0.05). Docosahexaenoic acid in milk was greater for cows on treatment 300 and 600 than for cows on treatment 0 and 100 (P < 0.05).

However, thousands of colonias residents (settlements in the Southwest US that lack access to basic infrastructure) still lack access to clean drinking water. Many different algal species can provide an acceptable protein ingredient, with good digestibility, for shrimp feeds. Compared to fish meal, similar protein, carbohydrate, and lipid levels can be found in select algal species. Traditional shrimp diets in aquaculture rely on fish meal and fish oil from pelagic fish fisheries. A reduction or elimination of these ingredients would reduce the dependency of shrimp aquaculture on offshore fisheries and increase economic competiveness. A novel procedure, ASAPALLE, for lipid extraction from wet algae, Nannochloropsis Salina, was developed and reported. This method does not require pre-dehydration of algal biomass and the total extraction time needed is under one minute. The ASAPALLE extraction was compared with hexane Soxhlet extraction to evaluate the lipid extraction efficiency and the lipid profile. Algal lipid yield of the ASAPALLE extraction was 14.3% of the total dry algal biomass weight, while the hexane Soxhlet extraction after 18 hours yielded 7.9% of algal lipid. The total lipid content was approximately 14.6%, determined by a modified Bligh-Dyer method. The quality of the …

Cyanobacteria show much promise in reducing biodegradable thermoplastic production costs; however, most currently characterized strains are ill-equipped to do so. The result of Objective I produced a high-throughput assay designed to discover existing cyanobacterial strains and rapidly characterize them as PHA-producers or potential PHA-producers. This assay will play an instrumental role in the attainment of a novel cyanobacteria environmental isolate capable of accumulating high levels of PHA naturally. The tidal-fresh James River experiences recurring blooms of toxin-producing cyanobacteria, including Microcystis aeruginosa. However, cyanobacteria cell densities in the James are relatively low. Our purpose was to identify key factors suppressing cyanobacteria growth in the face of eutrophication. A mesocosm experiment was designed to test the effects of light, mixing and grazing on cyanobacteria abundance, with nutrients held constant. We predicted that toxic cyanobacteria would be most abundant under stagnant conditions, with enhanced light, with no bivalve grazers present. Abundances of indicator gene copies and phytoplankton counts supported this hypothesis. However, chlorophyll-a, phycocyanin, and the toxin microcystin were all …

Installations similar to Louisiana sugar mills have been proposed as potential biorefineries for sugarcane, energy cane, and sweet sorghum. The nutrient-rich wastewater generated from the milling process can be used as an algal growth medium. Simultaneously, algae can serve as a water treatment strategy to deplete these effluents of excess nitrogen, phosphorus, and organic carbon.
