Construction Of An Instrument For Doing Fast Time-Resolved Infrared Spectroscopy, Susan R. Richter

We have designed and assembled a system for doing flash photochemical studies of organotransition metal carbonyl complexes with fast time-resolved infrared detection. Photolysis is with conventional xenon flashlamps (400J/flash) and time resolved infrared absorption spectra are measured with a diode laser (Laser Photonics, Inc.) and indium antimonide detector operating in the range 1700 - 2100 cm -1. Signals are collected and averaged on a digital oscilloscope (LeCroy 9410) for subsequent computer storage and analysis. The waveform is downloaded to the computer where calculations of percent transmittance and absorbance vs time are calculated. The information is then transferred to a spreadsheet …

Evaluation Of The Pti Ls-100 Lifetime Instrument And The Binding Of 2-Acetylnaphthalene To Γ-Cyclodextrins And Modified Β-Cyclodextrins, Timothy R. Cregan

The first part of this thesis is to determine the procedures for optimizing the data obtained from the PTI LS-100 lifetime instrument. The molecules 9-cyanoanthracene, pyrenebutyric acid, anthracene and N-acetyltrytophanamide have lifetimes which vary over the range 1 to 115 nsec and all follcw a s:ngle exponential decay. Therefore, we can evaluate the instrument by comparing these data using fluorescence lifetime reference standard values for the aforementioned molecules. The second part of this thesis is to investigate the binding of 2-acetylnaphthalere (2-AN) to various cyclodextrin molecules.

Tyrosine Flourescence To Monitor The Denaturation Of A Bacterial Protein, Kristin L. Trudeau

Tyrosine fluorescence was used to monitor the denaturation of a protein. The protein used as our model system was the regulatory subunit (RSU) of aspartate transcarbamoylase from E. coli. RSU is a dimer of two identical chains with one zinc ion bound per chain. It is also a typtophan-deficient protein so tyrosine flourescence was used as a probe to monitor changes during denaturation.

Two separate but overlapping projects were studied. First, the tryptophan corrected fluorescence emission spectra of ATCase and CSU were characterized under a variety of denaturant conditions at excitation 295 nm. Because CSU is a portion of ATCase, the environments of the same two tryptophan residues were being probed in both systems.

This paper will analyze the export-led model for development by looking at the intended versus the actual results of the Caribbean Basin Initiative (CBI) as it relates to the Caribbean country of Jamaica. The CBI was passed into law by the U.S. Congress under the Caribbean Basin Economic Recovery Act (CBERA) in 1983. An exploration is made of the circumstances leading to the CBI, the main precepts and legislation of the CBI, its implementation and its effects. Current research indicates that "...the teaching of problem solving is regarded by many as the most important and fundamental goal of school mathematics instruction" (Silver & Thompson, 1984). The fact that problem solving is considered a goal of mathematics instruction leads this writer to believe that the methodology involved should also be examined. In order to have successful programs both the teachers and the students must feel comfortable working with the problems. Past experience has made this writer aware that often this is not the case.

Isolation Of Protein Synthesis Initiation Factors Using Volvox Carteri: A Clue To The Translational Control Of Cytodifferentiation, Claudia Hackethal

Volvox carteri, a multicellular green algae, possesses two distinct cell types--the gonidia and somatic cells. This simple germ-soma dichotomy is ideal for the study of the control of cytodifferentiation. The control of cytodifferentiation in this organism has been identified as a posttranscriptional mechanism, which suggests translation. Initiation factors have been shown to play an important role in controlling translation in many other developmental systems. Therefore, study of the initiation factors in Volvox may shed light on a translational control mechanism. Edwin Arlington Robinson always felt that writing poetry was the only career that he could pursue in his life, but also always worried whether it was a worthy pursuit in the eyes of the culture in which he was raised. In a small town like Gardiner, Maine, where the 1690s' Puritan ethic was nearly as strong centuries later in the 1890s, the concept of a career in poetry or any artistic pursuit was absurd but not nearly as absurd as being successful at it. The victorious Sandinistas and their governing junta stood on the threshold of a Nicaragua eager to be transformed form the exploited, dependent, and terribly poor nation-state it had been virturally since its independence from Spain into a truly new society based on popular will and welfare. Research is increasingly suggesting that therapist and client ratings of greater beneficialI outcomes from therapy are strongly related to client and therapist perceptions of increasing convergence and generally positive expectations before counseling even begins. Using an analogue format, this study attempts to fill a void in the counseling literature by investigating Christian and non-Chiistian subjects' expectations for counseling with a Christian counselor, an agnostic counselor, or a counselor with no specified theistic orientation. After reviewing an analogue counselor description, subjects responded to the modified Brief Form of Tinsley's (1982) Expectations About Counseling questionnaire. Using my past experience in historic costume preservation, written resources, and the knowledge of those interested in this area of clothing & textiles, museum studies, and costume research, I hope to transform the state of the historic costume facility in Quigley Hall to one that's organized, utilizes proper storage techniques, and is available for use by both faculty and students. As a result of this reorganization, I also hope to have the opportunity to share this learned knowledge with the students and faculty of the Clothing & Textiles Department through some formal means.
