A Ph-Sensitive Delivery System For The Prevention Of Dental Caries Using Salivary Proteins, Yi Zhu May 2019

A Ph-Sensitive Delivery System For The Prevention Of Dental Caries Using Salivary Proteins, Yi Zhu

Electronic Thesis and Dissertation Repository

Dental caries remains one of the most common chronic diseases worldwide. Salivary proteins such as histatins have demonstrated biological functions directly related to tooth homeostasis and prevention of dental caries. However, histatins are susceptible to the high proteolytic activities in the oral environment. Therefore, pH-sensitive chitosan nanoparticles (CNs) have been proposed as potential carriers to target major oral diseases that occur under acidic conditions (e.g. dental caries and dental erosion). Four different types of chitosan polymers were investigated and the optimized CNs successfully loaded histatin 3 and released it selectively under acidic conditions. Through loading the survival time of histatin …

Study On The Interactions Between Nanomaterials And Proteins, Yueqi Bi Nov 2012

Study On The Interactions Between Nanomaterials And Proteins, Yueqi Bi

Electronic Thesis and Dissertation Repository

Incorporating proteins with nanomaterials is an effective way to enhance the stability and function of proteins. The protein-nanomaterials hybrid systems have been extensively applied in drug delivery and biocatalysis. This thesis focuses on the different interactions between proteins and nanomaterials. Three sub-projects have been studied as follows;

1) Chemical interaction: Gold nanoparticles with a particle size of 15nm were applied to label bovine serum albumin (BSA), a globular protein, for realizing a colorimetric protein assay. The results of FTIR and Raman spectra indicate that gold nanoparticles bond to BSA via the amine bonds. The surface plasma resonance (SPR) of …

Encapsulation And Controlled Release Of Rhu-Erythropoietin From Chitosan Biopolymer Nanoparticles, Cody Bulmer Apr 2012

Encapsulation And Controlled Release Of Rhu-Erythropoietin From Chitosan Biopolymer Nanoparticles, Cody Bulmer

Electronic Thesis and Dissertation Repository

The objective of this research project was to develop a drug delivery system for recombinant human erythropoietin (rHu-EPO), a glycoprotein hormone used in the treatment of renal anaemia and chemotherapy induced anaemia, using the biopolymer chitosan as the base component. Two types of chitosan nanoparticles were produced through ionotropic gelation using flush mixing with either tripolyphosphate (TPP) or carrageenan polymer. Chitosan-TPP and chitosan-carrageenan nanoparticles were generated under a variety of conditions to evaluate the effects of chitosan concentration, chitosan to anion mass ratio and solution pH on the nanoparticle characteristics of particle diameter, surface charge and particle size distribution. A …

Synthesis And Characterization Of Ph Sensitive Hydrogel Based Nanocarriers For The Controlled Release Of Protein Drugs, Kazi Farida Akhter Jan 2010

Synthesis And Characterization Of Ph Sensitive Hydrogel Based Nanocarriers For The Controlled Release Of Protein Drugs, Kazi Farida Akhter

Digitized Theses

Controlled drug delivery technology represents one of the frontier areas of science, which involves a multidisciplinary scientific approach and contributes to human health care. Hydrogel based nanoparticles (NPs) have gained considerable attention in recent years as one of the most promising nanoparticulate drug delivery systems, owing to their unique potentials by combining the characteristics of a hydrogel system, i.e. hydrophilicity and extremely high water content, with a nanoparticle i.e. very small size. In this research, hydrogel-based nanoparticles, i.e. gelatin NPs, were used as a carrier to encapsulate protein drug for controlled release in a prolonged manner. Bovine serum albumin (BSA) …

Encapsulation And Controlled Release Of Rh-Erythropoietin From Biopolymer Nanoparticles, Mehrdad Bokharaei Jan 2009

Encapsulation And Controlled Release Of Rh-Erythropoietin From Biopolymer Nanoparticles, Mehrdad Bokharaei

Digitized Theses

Recombinant human erythropoietin (rhEPO) is a glycoprotein, which is produced commercially from Chinese hamster ovary (CHO) cells. It is used for chronic therapy of renal anemia and chemotherapy-induced anemia in cancer patients. Recent evidence suggests that rh-EPO exerts tissue protective effects via multiple mechanisms which include inhibition of apoptosis, promotion of angiogenesis and decreased inflammation. After Intra Venus (I.V.) injection, the blood concentration of rh-EPO rapidly decreases due to proteolysis with a short half-life of 8.5 h, which makes treatment expensive. It is desirable to develop an encapsulation method which will provide a controlled release of rh-EPO and maintain the …
