Streptococcus pyogenes chromosomal island M1 (SpyCIM1) integrates by site-specific recombination into the 5’ end of DNA mismatch repair (MMR) gene mutL in strain SF370SmR, blocking transcription of it and the downstream operon genes. During exponential growth, SpyCIM1 excises from the chromosome and replicates as an episome, restoring mutL transcription. This process is reversed in stationary phase with SpyCIM1 re-integrating into mutL, returning the cells to a mutator phenotype. Here we show that elimination of SpyCIM1 relieves this mutator phenotype. Brazil has a unique mental health care system, characterized by universal coverage delivered by interdisciplinary teams both in the community and in specialized centros de atenção psicossocial (CAPS—psychosocial care centers). Provision of patient-centered mental health care is an important principle of Brazilian mental health care, but this topic has not been well-studied. We analyzed data from a cross-sectional survey of 151 community health workers (CHWs), nurses, and physicians in Santa Luzia, Minas Gerais State, Brazil. We demonstrate that topologically nontrivial states of light can be engineered in periodic photonic structures containing media with a Tellegen-type bianisotropic response. Whilst in such bianisotropic materials the time-reversal symmetry is broken, they are characterized by an intrinsic magnetic order which does not require macroscopic magnetization. Our design can therefore be considered as a direct analog of the solid state Chern insulator which exhibits a topological order in the absence of an external bias. Numerical simulations of such Tellegen photonic crystals reveal the existence of one way edge transport at domain walls and perfectly conducting boundaries not sensitive to structural …

The production of melanin significantly enhances the virulence of many important human pathogenic fungi. Despite fungal melanin’s importance in human disease, as well as melanin’s contribution to the ability of fungi to survive in diverse hostile environments, the structure of melanin remains unsolved. Nevertheless, ongoing research efforts have progressively revealed several notable structural characteristics of this enigmatic pigment, which will be the focus of this review. These compositional and organizational insights could further our ability to develop novel therapeutic approaches to combat fungal disease and enhance our understanding of how melanin is inserted into the cell wall.

Remarkable disparities in smoking rates in the United States contribute significantly to socioeconomic and minority health disparities. Access to treatment for tobacco use can help address these disparities, but quitlines, our most ubiquitous treatment resource, reach just 1%–2% of smokers. We used community-based participatory methods to develop a survey instrument to assess barriers to use of the quitline in the Arkansas Mississippi delta. Barriers were quitline specific and barriers to cessation more broadly. Over one-third (34.9%) of respondents (n = 799) did not have access to a telephone that they could use for the quitline. African American (AA) breast cancer patients often have triple negative breast cancer (TNBC) that contains mutations in the TP53 gene. The point mutations at amino acid residues R273 and R248 both result in oncogenic gain-of-function (GOF) phenotypes. Expression of mutant p53 (mtp53) R273H associates with increased cell elasticity, survival under serum deprivation conditions, and increased Poly (ADP ribose) polymerase 1 (PARP1) on the chromatin in the AA-derived TNBC breast cancer cell line MDA-MB-468. We hypothesized that GOF mtp53 R248Q expression could stimulate a similar phenotype in the AA-derived TNBC cell line HCC70. To test this hypothesis we depleted the R248Q …

Prostate cancer (PCa) is the most commonly diagnosed cancer as well as the greatest source of cancer-related mortality in males of African ancestry (MoAA). Interestingly, this has been shown to be associated with single nucleotide polymorphisms around regions 2 and 3 of the 8q24 human chromosomal region. The non-protein coding gene locus Plasmacytoma Variant Translocation 1 (PVT1) is located at 8q24 and is overexpressed in PCa and, therefore, is also a candidate biomarker to explain the well-known disparity in this group. This thesis traces Gego’s contestation of art historical modernism through examining her relationship to the modernist grid and to her modernist genealogy of Constructivism, Geometric Abstraction, and Kineticism. These two nonlinear developments subvert the teleological conception of progress presumed by Greenbergian modernism, and bring forth Gego’s own aesthetic conception of temporality.

This thesis explores the social and cultural climate surrounding the art commissions for Philip Johnson's New York State Pavilion at the 1964-65 World's Fair. The research presented herein examines how the economic and cultural climate of 1960's America affected the architectural landscape at the World's Fair and how Johnson's Pavilion was integrated into it. Finally, this thesis examines how artworks by John Chamberlain, Ellsworth Kelly, Roy Lichtenstein, Andy Warhol and others responded to the commercial premise of the fair itself. "Archival Enactment, Retelling 'The Big Book': Alison Knowles, Something Else Press and Fluxus," positions Knowles’ Big Book (1966) as a case study of historical methodology and interdisciplinary artistic practice in the post-war period. This comprehensive analysis of Big Book, a work of art no longer extant, contextualizes its publisher, Something Else Press through Dick Higgins’ concept of “intermedia,” and important lesser-known junctures relevant to Fluxus and the group’s leader George Maciunas are illuminated. Knowles' early and lesser-known silkscreen paintings are also examined.

Once-daily Truvada (Emtricitabine/Tenofovir) as a method of pre-exposure prophylaxis (PrEP) is one of the most promising biomedical interventions to eliminate new HIV infections; however, uptake among gay, bisexual, and other men who have sex with men has been slow amidst growing concern in popular/social media that PrEP use will result in reduced condom use (i.e., risk compensation). We investigated demographic, behavioral, and psychosocial differences in willingness to use PrEP as well as the perceived impact of PrEP on participants’ condom use in a sample of 206 highly sexually active HIV-negative gay and bisexual men. By laying out the tools of my practice the paper gives the reader a context on how I digest and synthesis works of art. Aim: To detect pathological brain conditions early is a core procedure for patients so as to have enough time for treatment. Traditional manual detection is either cumbersome, or expensive, or time-consuming. We aim to offer a system that can automatically identify pathological brain images in this paper.Method: We propose a novel image feature, viz., Fractional Fourier Entropy (FRFE), which is based on the combination of Fractional Fourier Transform(FRFT) and Shannon entropy.
