Thus, the purpose of this thesis was to characterize age and sex differences in the motivational/rewarding effects of nicotine (Aim 1) and withdrawal behavior (Aim 2). To more closely model human use patterns, the present study employed nicotine vapor methods involving passive exposure for 14 days in adolescent and adult female and male rats. The purpose of this study was to use semi-structured interviews to obtain a better understanding of ECIGs from the perspective of regular ECIG users. As such, the current literature has begun to explore factors associated with e-cigarette use in significantly affected populations. Anxiety sensitivity (AS), the fear of the sensations associated with the experience of anxiety, is a known transdiagnostic risk factor for tobacco use. Additionally, higher AS has been shown to be associated with higher levels of e-cigarette use, greater perceived benefits of use, greater positive outcome expectancies, greater perceived risks of use, more quit attempts, and more difficulty quitting. Vaping has become a common method to consume nicotine and cannabis on college campuses across the United States. Between 2017 and 2019 there was a significant increase in the prevalence of both nicotine (6% to 22%) and cannabis vaping (5% to 14%) among college students. As of 2019, there were over 2,000 vaping-associated lung injuries or deaths and recent data suggest nicotine vaping was associated with greater risk of contracting COVID-19. The goal of this study was to find out if inactivating the corticostriatal pathway through DREADDs (Designer Receptors Exclusively Activated by Designer Drugs) inhibited goal-tracking behavior in rats. In this experiment, rats underwent two surgeries. One was to inject DREADDs into the infralimbic cortex and the other in the dorsomedial caudate-putamen (dmCPu). Introduction: Aim 1 characterized sex differences and the role of ovarian hormones in physical signs and negative affective states produced by nicotine withdrawal in female, ovariectomized female (OVX), and male rats. We also compared nicotine withdrawal and corticosterone levels with estradiol (E2) and progesterone across the 4-day estrous cycle. Aim 2 validated the role of ovarian hormones in withdrawal in OVX females that received vehicle, E2, or E2+progesterone. Methods: Female rats received a sham surgery or an ovariectomy procedure. Electronic cigarette (ECIG) users can manipulate several device features including liquid nicotine concentration (mg/ml) and heating coil resistance (Ohms). One class of ECIG models, called “sub-Ohm” devices, use coils with a resistance of < 1 Ohms, lower than those observed in conventional ECIGs (e.g., ≥ 1.5 Ohms). Increasing voltage or decreasing coil resistance increases device power. Given that ECIG coil resistance and liquid nicotine concentration have not been manipulated systematically and simultaneously in clinical laboratory studies, the influence of these factors on ECIG acute effects remain unclear. The primary purpose of this study was to examine the influence of coil resistance and liquid nicotine concentration on nicotine delivery, heart rate (HR), subjective effects, puff topography, and liquid consumption.

Thirty-two experienced ECIG users completed four independent laboratory sessions that differed by coil resistance (0.5Ohm or 1.5Ohm ) and liquid nicotine concentration (3 or 8 mg/ml). In each session, participants used a 4.5 V “Kanger SUBOX” loaded with 3.5 ml ECIG liquid in a 10-puff directed and 60-minute ad libitum bout.
