Differences In Compulsive-Like Ethanol Consumption In Adolescent-Trained And Adult-Trained Rats Exposed To Acute Isolation Stressors, Maria J. Lopez

The Pavlovian Conditioned Approach (PCA) procedure is an experimental procedure used with animals to induce compulsive-like behaviors directed towards an object conditioned stimulus (CS) paired with a sucrose pellet unconditioned stimulus (US) presented in a nearby food tray (sign tracking). When the object CS is an alcohol-containing bottle, the rats demonstrate compulsive-like interactions with the bottle object (e.g., approach, contact, and licking) that also results in alcohol consumption (drinking) in most rats. Compulsive alcohol drinking in a sign-tracking procedure is a result of the combined effects of schedule-induced compulsive-like behavior and the intake of alcohol (Tomie & Sharma, 2013).

Adolescence is a critical developmental period. An important change that occurs in adolescence is the neurological maturation for adult-type cognitive abilities. Research has linked adequate sleep quantity to successful learning and memory capabilities. However, due to a shift in sleep timing drive in adolescence, in combination with early awakening for school, the adolescent population is experiencing chronic sleep restriction (CSR). Prior research has demonstrated that ketamine causes deficits in object recognition and location memory following both acute and chronic administration (Pitsikas & Boultadakis, 2009; Venãncio et al., 2011). Although it is well established that abuse of many different kinds of drugs during the critical developmental period of adolescence can lead to impairments in cognition later in adulthood (Gilpin et al., 2012; Wagner et al., 2010), no research has investigated the effects of chronic ketamine administration during adolescence and its effects on behavior in adulthood.
