Plants respond to environmental stress by altering their gene expression. Under stress conditions some genes are activated and some genes are repressed. Even though a lot of work has been done to understand mechanisms of gene activation under abiotic stress very little information is available on how stress responsive genes are kept repressed under normal growth conditions. Recent work has revealed that plants use transcriptional repression as common mechanism of gene repression. Transcriptional repression is achieved by recruitment co-repressor complexes to the target genes. Restoration Of A Wet Longleaf Pine (Pinus Palustris) Savanna In Southeast Louisiana: Burning Toward Reference Conditions, Alex K. Entrup

This study quantifies the changes in vegetation composition and structure of a fire-excluded Pinus palustris (longleaf pine) wetland savanna restoration site in southeastern in comparison to a proximate contemporary reference site. The restoration site was invaded by hardwood species and off-site pines, and never underwent extensive soil disturbance. The restoration treatments involved logging across portions of the site and the reintroduction of fire across the entire site. All species present in 10m2 quadrats were recorded prior to treatment and throughout the 17 year study at reference and treatment sites. The highly invasive grass species, giant reed (Arundo donax), has been a major contributor to riparian habitat degradation in California for over 50 years. Several modes of vegetative reproduction have allowed this alien species to take advantage of fluvial processes and rapidly spread within California watersheds. A. donax dramatically alters hydrologic regimes, displaces native vegetation, and removes food and habitat for native wildlife. Siliques are photosynthetically active seed capsules and their development is strongly influenced by embryo development. MicroRNA156 (miR156)-SQUAMOSA PROMOTER BINDING PROTEIN-LIKE (SPL) network is involved in regulating plant growth and development, but the downstream genes of this network are still not fully elucidated. Here, I show that the miR156/SPL2 pathway controls the development of floral organs, regulates pollen production, and thus affects male fertility in Arabidopsis thaliana. I present evidence that SPL2 binds to the 5’UTR of the ASYMMETRIC LEAVES 2 (AS2) gene in vivo, indicating that AS2 acts downstream of SPL2. Soybean [Glycine max (L.) Merrill] is the leading oilseed crop grown in the world. Yield, fatty acids, protein, and oil are commercially important soybean traits; thus evaluation of breeding strategies for improvement of these traits is merited. To accomplish this, a comparison of molecular and phenotypic breeding strategies from progeny row selections was performed. From this it was determined that molecular strategies consistently outperformed phenotypic selections (PS) in the progeny row stage for soybean yield, fatty acids, protein, and oil. variety of functions including antimicrobial, insecticidal, and nematicidal activities. This

research assessed the performance of the beet armyworm, Spodoptera exigua, following ad

libero feeding on Medicago truncatula accessions differing in specialized metabolite and

saponin profiles. Insect performance did not directly relate to the foliar saponin levels in the

accessions, which was measured using HPLC mass spectrometry. Accession ESP105 had the lowest foliar saponins overall, yet had similar levels of soyasaponins compared to the other accessions. Conversely, accessions A17, PRT178, and GRC43 had high foliar saponins with particularly high …

Edamame (Glycine max), also known as vegetable soybean, was introduced to the United States from Japan in 1890 and has been growing in popularity as a nutrient-rich, low-sugar snack in recent years. In 2012, the American Vegetable Soybean and Edamame, Inc. established the first domestic commercial processing plant in Mulberry, Arkansas and contracted local growers for production. Since the crop is harvested when seed are immature, management practices are different from those for traditional soybean. The diversity of isolates of M. oryzae from Africa and the U.S. were examined using vegetative compatibility and virulence phenotyping as well as determination of variation in the avirulence gene AVRPiz-t in isolates from Africa. Gray leaf spot, caused by Cercospora zeae-maydis, is one of the most destructive foliar diseases of maize worldwide. C. zeae-maydis orients hyphal growth towards stomata (stomatal tropism) and forms infectious structures (appressoria) that are necessary for successful infection. Although some genes involved in pathogenesis in C. zeae-maydis have been identified, the molecular mechanisms are not well understood. In fungi, heterotrimeric G-proteins consist of three subunits (α, β, and γ) and mediate responses to environmental stimuli. To produce fermentable sugars from lignocellulosic biomass feedstock, severe pretreatment conditions are needed (either high acid concentration, temperature, or retention times). High severities can produce toxic byproducts which inhibit enzymatic hydrolysis or fermentation. In order to reduce pretreatment severities (and thus increase enzyme and fermentation efficiency), the white-rot fungus Pleurotus ostreastus was seeded into square and round bales of Kanlow switchgrass (Panicum virgastum L.) and left in the field over a period of 9 month. The laccase producing fungus is believed to selectively degrade lignin, a common plant structural polymer, which can function as an enzymatic inhibitor. Samples were taken …

Lucinid clams and their sulfur-oxidizing endosymbionts comprise two compartments of a three-stage, biogeochemical relationship among the clams, seagrasses, and microbial communities in marine sediments. A population of the lucinid clam, Stewartia floridana, was sampled from a subtidal seagrass bed at Bokeelia Island Seaport in Florida to test the hypotheses: (1) S. floridana, like other lucinids, are more abundant in seagrass beds than bare sediments; (2) S. floridana gill microbiomes are dominated by one bacterial operational taxonomic unit (OTU) at a sequence similarity threshold level of 97% (a common cutoff for species level taxonomy) from 16S rRNA genes; …

An apparent reduction in the performance of neonicotinoid seed treatments in controlling thrips, especially in cotton, has been observed, which has coincided with the increased use of both pre- and post-emergence herbicides to control glyphosate-resistant weeds. The objective of this research was to evaluate the potential interactions of both pre- and post-emergence herbicides on the efficacy of insecticide and fungicide seed treatments in cotton

In plants, reactive oxygen species (ROS) are generated as a byproduct of normal metabolism, as well as in response to adverse conditions such as light stress, extreme temperatures, and exposure to pests and pathogens. Singlet oxygen (1O2) is a ROS that is formed during photosynthesis in photosystem II (PSII) of the chloroplasts. Levels of 1O2 and other ROS are tightly controlled in healthy plants, but some studies suggest that levels of fatty acid desaturase (FAD) activity in Arabidopsis thaliana can influence constitutive and/or stress-responsive ROS accumulation. An effective management program for stink bugs (SBs) in pecan groves requires knowledge of: stages of the pecans susceptible to SB damage; strata of the tree with SB damaged nuts; a practical SB monitoring method; and, effects of landscapes contributing SBs into pecan groves.

Stink bugs produced feeding punctures in pecan shucks at all phenological stages. Pecans punctured before the dough stage drop from the tree. Kernel damage occurs in the pecans’ dough stage, whereas mature pecans are not damaged.

Flooding can significantly reduce soybean growth, development, and yield. Therefore, screening and identification of flood-tolerant soybeans will enhance development of cultivars that are well suited for flood-prone areas. Sets of screening tests were conducted in Stuttgart, Arkansas using three separate but related experiments, with the purpose of establishing effective flood tolerance screening protocol that can identify tolerant and sensitive cultivars. Each experiment was completely randomized with three replications. The first experiment involved screening of 256 maturity group (IV and V) cultivars. The recent completion of the tomato genome sequence is a great milestone towards learning about the tomato genome. The vascular flora of Wayne County, Mississippi, was surveyed from 2013–2015, and the species of native and naturalized plants are listed. In total, 1,135 species were identified from field collections, previously collected herbarium specimens, and field observations. These belong to 556 genera and 165 families. Four of these species represent first recorded occurrences in the state of Mississippi: Blyxa aubertii Rich., Gomphrena serrata Pav. ex Moq., Isoetes valida (Engelm.) Clute, and Polypogon interruptus Kunth. GPS coordinates are given for important collection sites representing the diversity of plant communities in the county. New herbicide-resistant soybean traits will increase the number of herbicides that can be applied in soybean and change the technology currently used to make herbicide applications in soybean. Palmer amaranth (Amaranthus palmeri) remains one of the most difficult weeds to manage in soybean and thus is the primary focus of this research. Therefore, experiments were conducted to investigate Palmer amaranth temporal emergence, control of common Arkansas weeds with new soybean herbicide programs, and the effect of application technology on dicamba-containing tank mixtures. Piratebush (Buckleya distichophylla (Nutt.) Torr.) is a rare, hemiparasitic shrub with the only extant populations in western North Carolina, northeastern Tennessee, and southwestern Virginia. The preferred natural hosts of piratebush, Carolina and eastern hemlocks, have seen sharp declines over the last decade due to the invasive hemlock woolly adelgid. Virginia pine, another important host of piratebush, is also susceptible to disease, specifically Cronartium appalachianum, a rust fungus for which piratebush is the secondary host. Herbicide-resistant weeds in Arkansas cause problems for growers. Up-to-date information and new technologies can help plan mitigation strategies to slow resistant weeds. The objectives of this research were to provide a ‘snapshot’ of herbicide-resistant Echinochloa spp. in rice producing counties, determine how much resistance has spread across the state, and understand the effectiveness of 4-hydroxyphenylpyruvate dioxygenase (HPPD)-inhibiting herbicides for control of glyphosate-resistant (GR) Palmer amaranth relative to commercial standards currently labeled in soybean. Current Central American agricultural practices are environmentally and economically unsustainable, yet the ancient Maya who lived in the same region thrived for thousands of years. Archaeologists have attempted to understand the factors enabling the prolonged success and ultimate collapse of the Maya societies. Alternaria solani, cause of early blight, threatens potato yields. Fungicide resistance has made control of early blight difficult and there are concerns that in-season fungicide use results in resistance to boscalid, fluopyram, and chlorothalonil. Concern of high levels of resistance to boscalid a group 7 fungicide may confer cross-resistance to fungicides of the same group such as fluopyram. From 2014 to 2015, A. solani isolates were collected from field plots treated with boscalid, fluopyram, and chlorothalonil to test resistance levels. Isolates were determined resistant if EC50 values were higher than 5 µg ml-1. Boscalid and chlorothalonil mean EC50 values decreased …

Counter to what some people think, urban areas can be biodiversity hotspots. Maintaining this biodiversity can be challenging, since exotic shrubs and vines block sunlight and threaten native plant regeneration. Since 2007, the Louisville Olmsted Parks Conservancy (LOPC) has spent $2 million on invasive plant management in Cherokee Park. Before the project began, long-term transects were established by the LOPC to collect baseline presence/absence data on 11 invasive plant species. In 2014, I revisited these transects and documented presence/absence data on the entire plant community. A shift in public demand towards more organic and locally produced fruit and vegetables has been occurring across the United States in recent years. A common practice in organic fruit production is the application of organic ground covers to supply nutrients while enhancing other soil properties. A need for research exists in the southern region of the U.S. examining the effects of regionally applicable ground cover and nutrient management on nitrogen availability and the microbial community to provide information to organic farmers in the region.
