Large White BUTA hens were inseminated and euthanatized 24 to 48 h later. Oviducts were excised, UVJ tissue removed, and SST were procured by enzymatic digestion. Recovered SST were intact and contained motile sperm. The sperm were oriented with their acrosomes pointed towards the distal end of the SST, and their long axes in parallel with the long axis of the tubule’s lumen. This method for the isolation of intact SST can be readily applied …

Physiology And Reproduction: Gross Appearance Of The Turkey Blastoderm At Oviposition, M. R. Bakst, S. K. Gupta, W. Potts, V. Akuffo

United States Department of Agriculture-Agricultural Research Service / University of Nebraska-Lincoln: Faculty Publications

The blastoderm (fertilized ovum) and unfertilized germinal disc (UGD) of fresh laid eggs and eggs stored prior to incubation exhibit subtle but definable morphological variations. Such variations may lead to difficulty when attempting to determine true flock fertility based on the appearance of the blastoderm/UGD. The objectives of this study were to define and categorize such morphological variations and to determine whether sperm influence the frequency distribution of the different categories. Eleven categories of blastoderms were defined based on the relative density and appearance of the area alba, area pellucida, area opaca, and the periblast. The majority of the blastoderms …

Duration Of Fertility Of Turkeys Inseminated At Different Times After The Onset Of Photostimulation, M. R. Bakst

United States Department of Agriculture-Agricultural Research Service / University of Nebraska-Lincoln: Faculty Publications

To investigate the relationship between photostimulation, ovary and oviduct weights, and oviducal sperm-storage tubule (SST) development and functional capacity, 33-week-old turkey hens were placed in four groups of 15 hens each. Each group was inseminated three times in 30 min with 50 × 106 spermatozoa (a total of 150 × 106 spermatozoa) on the day of photostimulation, or on Days 7, 14, or 21 after the onset of photostimulation (Groups A, B, C and D, respectively). From weeks 4 to 8 after photostimulation, 5 additional hens were inseminated each week and then killed 24 h later to determine …

The onset of egg production (mean 18 · 3 days after the onset of photostimulation) and the rate of egg production (flock averaged 4 · 9 eggs per bird per week for the first 8 weeks of egg production) were not affected by 5 days of twice daily oviduct eversion ('venting') in the pre-laying period when compared to unvented controls. After the onset of photostimulation, pre-laying hens were inseminated twice daily on Days 12 to 16 with 3 μl semen containing 15 × 106 spermatozoa, and compared with groups of hens inseminated once daily on Days 15 and 16 with …

Historical Research Bulletins of the Nebraska Agricultural Experiment Station

This is the first report of results from a high protein-high lysine observation (HP-HL) nursery organized in 1974 by the Nebraska Agricultural Experiment Station and the Agricultural Research Service, U.S. Department of Agriculture, under a contract with the Agency for International Development, U.S. Department of State. Primary objectives of this nursery are to: (1) Systematically provide breeders and cooperators with superior genetic germplasm for elevated levels of protein and/or lysine. (2) Test the degree of expression of the high protein and high lysine traits in a diverse array of environments.
