The purpose of this paper is to present some understandings of the human problem-solving activity that we have gained in the Collaborative Agent Design Research Center (CADRC) over the past two decades. Since we feel strongly that the human decision-maker should be an integral component of any computer-based decision-support system, it follows that we would have endeavored to incorporate many of the elements that appear to be important to the user in the design of these systems. Glycosphingolipids (GSLs), composed of a polar saccharide head and a lipophilic ceramide tail, are ubiquitous components of the plasma membrane of eukaryotic cells. They serve in many regulatory capacities and have antigenic properties towards natural killer T (NKT) cells of the innate immune system. Critical to the recognition of glycosylceramides by NKT cells are antigen presenting cells (APC), such as dendritic cells, which are responsible for binding, processing, and delivery of ligands to these lymphocytes. This event is mediated by CD1d, a major histocompatibility complex-like protein expressed on the surface of APCs, which binds GSL antigens by the ceramide moiety …

The synthesis of zinc oxide through three routes was investigated; carboxylic acid chelation, polymer assisted synthesis in an alcoholic medium and slow thermolysis of precursor. In the case of carboxylic acid chelation, it was determined that the rate at which the precursor is formed has a significant effect on the quality of zinc oxide particles produced. Investigation of the effect of concentration, pH and species of chelating agent used on the purity and structure of product was undertaken. Aspernigrin A is a compound correlated with inhibiting the growth of cancer cells in the colon. A five-step synthesis was designed for aspernigrin A, projected to yield four intermediates and the final product. The intermediates were analyzed by high field 1HNMR and FT-IR spectroscopy before proceeding to the next step. A model pyridone ring system was successfully prepared from a commercially available pyrone precursor in two steps. These results provide a proof of concept that the synthetic scheme should successfully yield the natural product.

Knoevenagel condensation of 2-formyl-octamethylcalix[4]pyrrole with selected 1,3-indanedione derivatives yields calix[4]pyrrole anion sensors with push-pull chromophores displaying strong intramolecular charge transfer. The push-pull feature results in augmented signal output as well as in dramatic changes in anion selectivity exemplified by a 50-fold increase in acetate vs chloride selectivity compared to the parent calix[4]pyrrole.

Interaction-based and state-based modeling are two complementary approaches of behavior modeling. The former focuses on global interactions between system components. The latter concentrates on the internal states of individual components. Both approaches have been proven useful in practice. One challenging and important research objective is to combine the modeling power of both effectively and then use the combination as the basis for automatic design synthesis. We present a combination of interaction-based and state-based modeling, namely, Live Sequence Charts and Z, for system specification. The developed method of high-pressure synthesis (HPS) allows producing nanostructural highly dense material based on MGB2, which possesses the highest superconducting and mechanical characteristics among the known world analogues, in the form of blocks that are suitable for application in SC electromotors and pumps. Chiral α-hydroxy-aldehydes generated in situ by the ADH reaction of vinyl sulfones undergo a borono-Mannich reaction with β-styrenyl boronic acid and primary amines to give anti-1,2-amino alcohols in high enantiomeric purities (ee 83-95%). This new method allows much more rapid access to these valuable chiral building blocks that has been used in a short formal synthesis (10 synthetic steps from 4-penten-1-ol) of (-)-swainsonine.

Cell surface carbohydrates play an important role in a wide variety of biological processes such as inflammation, tumor metastasis, and viral and bacterial infection. The goal of our research has been two-fold. The first objective was the synthesis of antimycobacterial compounds. A mannose containing tetrasaccharide from the mannan core of lipoarabinomannan (LAM) of Mycobacterium tuberculosis has been synthesized using α-D-methylmannopyranoside as starting material and Koeings-Knorr reactions to couple saccharides. The synthesis was completed in nine steps and in 14% total yield. This compound should be useful in competitive inhibition studies with macrophages or as an immunological marker. Acetal of warfarin acid has been synthesized via a hemi acetal intermediate and structurally characterized by X-ray diffraction. The crystal structure shows that the title compound was formed by the hydrolysis of warfarin acid in methanol at pH 3, with space group P2_1/n and unit cell parameters a = 5.859 (17), b =16.745 (5), c = 16.402 (5) Å, \alpha = 90.00; \beta = 94.853 (4), and \gamma = 90.00°.

The Schiff base octaazamacrocyclic ligands derived from primary diamines and 3,6-dimethyl/diphenyl-4,5-diazaocta-3,5-diene-2,7-dione, and their binuclear complexes [M_2LCl_4]Cl_2 [M = Cr(III), Fe(III), Co(III), or Ru(III)] and [Mn_2L(AcO)_4](AcO)_2 were synthesized by template condensation reactions. Attempts to synthesize the corresponding metal-free macrocyclic ligands did not prove successful. The overall geometry and stereochemistry of these complexes were elucidated by elemental analyses, magnetic susceptibilities, electronic spectra, infrared spectra, molar conductance measurements, ^1H NMR, and thermogravimetric analysis. All the trivalent metal ion complexes appear to be 1:2 electrolytes. An octahedral geometry is proposed for all the complexes.

Six new 3-pyrrolin-2-one derivatives were synthesized via the condensation reaction of amino acid esters (2a-f) with 2,5-dimethoxy-2,5-dihydrofurane (1) in acidic medium. This simple one-pot reaction furnished the corresponding pyrrolinones (3a-f) in acceptable yields.

A series of 4-arylidenamino-4H-1,2,4-triazole derivatives (3-11) were synthesized from the treatment of 4-amino-4H-1,2,4-triazoles (2) with certain aldehydes. Compounds 3-11 were reduced with NaBH_4 to afford the corresponding 4-arylmethylenamino-4H-1,2,4-triazoles (12-20). Compounds 1-10 and 12-19 were characterized by elemental analyses and ^1H NMR, ^{13}C NMR, IR and UV spectral data. Compounds 11 and 20 were characterized by ^1H NMR, ^{13}C NMR, IR and mass spectral data. Compounds 14, 16, 17, and 18 were tested for anticancer activities. 4-(Benzeneazo)salicylaldehyde was synthesized with aniline, sodium nitrite and salicylaldehyde. Two tetradentate ligands, N,N'-bis[4-(benzeneazo) salicylaldehyde]-o-phenylenediamine (H_2L) and N,N'-bis[4-(benzeneazo) salicylaldehyde]ethylenediamine (H_2L') were formed by the 2:1 molar condensation of 4-(benzeneazo) salicylaldehyde with o-phenylenediamine or ethylenediamine. Their Cu(II), Ni(II), Zn(II), Co(II), Mn(II) and Cd(II) complexes were synthesized and characterized by elemental analysis, molar conductivity measurements, and infrared and electronic spectral data. Two ligands and their Ni(II) and Zn(II) complexes were further identified using ^1H NMR spectra. The results suggest that the metal is bonded to the ligand through the phenolic oxygen and the imino nitrogen.

We report the synthesis, structural characterization and thermal behavior of N'-(4-chlorobenzoyl)-N,N-di-n-butylthiourea (HL) and its nickel complex (NiL_2). Some physical properties of the prepared compounds were investigated using elemental analyses, IR, ^1H-NMR, and magnetic susceptibility. The molecular structures of HL and NiL_2 were determined by single crystal X-ray diffraction. HL is a monoclinic, space group P2_1/c with a = 10.7457(5) Å, b = 15.5909(8) Å, c = 10.4536(5) Å, \beta = 104.398(1)°, and V = 1696.34(14) Å^3 with Z = 4 for d_{calc} = 1.280 g/cm^3.
