Knowledge And Behavioural Interventions To Reduce Human Health Risk From Private Groundwater Systems: A Global Review And Pooled Analysis Based On Development Status, S. Mooney, C. P. Mcdowell, J. O'Dwyer, P. D. Hynds

Articles

Groundwater contamination constitutes a significant health risk for private well users residing in rural areas. As the responsibility to safeguard rural private domestic groundwater typically rests with non-expert homeowners, interventions promoting risk mitigation and awareness represent the most viable means of preventing supply contamination. However, no global review or pooled analyses of these interventions has been undertaken to date. The current study sought to identify and quantify the performance of private well interventions from 1990 to 2018 via a global systematised review and pooled analysis. The PICO (Population-Intervention-Comparison-Outcome) approach was employed for literature identification. Relevant studies were statistically analysed across …

Risk Communication Approaches For Preventing Private Groundwater Contamination In The Republic Of Ireland: A Mixed-Methods Study Of Multidisciplinary Expert Opinion, S. Mooney, J. O'Dwyer, Paul Hynds

Articles

The mechanisms of private-well groundwater contamination are uniquely complex, necessitating a multisector communicative approach to risk management, premised on behaviour promotion. In countries such as the Republic of Ireland (ROI), characterised by oftentimes high groundwater contamination risk and concurrently limited user awareness, incorporation of multidisciplinary, ‘expert-based’ knowledge may facilitate design of evidence-based, practical interventions. Expert interviews represent an efficient form of expert consultation, enabling ease of access to niche information and comparison of procedure, but remain under-utilised within the groundwater management literature. In response, the current study elicited opinion from 50 experts across four broad categories (communications, engineering/science, policy, and …

Approximately 12% of the Canadian population uses private wells for daily water consumption; however, well water testing rates are on the decline, resulting in an increased risk of waterborne acute gastrointestinal illness. To date, limited research has explored the determinants influencing well testing practices. Accordingly, the current study sought to investigate the drivers of “one-off” and repeat well water testing in southern Ontario during the 5-year period 2012–2016, using the worlds largest private groundwater testing data-frame. Data from >400,000 wells were geospatially integrated with all tests conducted by the provincial laboratory in southern Ontario. The Ontario Marginalization Index (ON-Marg) was …

Timely access to health services has become increasingly difficult due to demographic change and aging people growth. These create new heterogeneous challenges for society and healthcare systems. Congestion at acute hospitals has reached unprecedented levels due to the unavailability of acute beds. As a consequence, patients in need of treatment endure prolonged waiting times as a decision whether to admit, transfer, or send them home is made. These long waiting times often result in boarding patients in different places in the hospital. This threatens patient safety and diminishes the service quality while increasing treatment costs. It is argued in the …

A new chemical entity, which is a chiral indane dimer, PH46A, has been developed by our research group. As a clinical candidate. PH46A has recently completed Phase I clinical studies in man. Previously, during its pre-clinical development, in in vivo pre-clinical studies PH46A showed potent anti-inflammatory properties, which can be targeted at a range of diseases, including inflammatory bowel disease (IBD). To support the pre-clinical development of this drug candidate, we developed a LC MS/MS method for determining PH46 (the acid form of PH46A salt) in both dog and rat plasma using Compound 1 as internal standard (IS). Those species …

Improving the data descriptor calculation of crystal’s physical properties requires sophisticated imaging techniques and algorithms. It has been possible to construct 2D population balance models benefiting from characteristic measurements of both crystal’s length and width, compared to the single representative sizes used in 1D models. Our aim is to ameliorate the procedure of determining shape (and not only size) factors, in an automated fashion and directly from the process, for implementation in future models. Here, approaches suitable for real-time applications were employed including engineered imaging sensors and adaptive algorithms. We described the latter in detail for varying 2D image datasets. …

The presence of low amounts of specific proteins in urine can be an indicator of diagnosis and prognosis of several diseases including renal failure and cancer. Hence, there is an urgent need for Point-of-care (PoC) methods, which can quantify microproteinuria levels (30-300 ppm) and identify the major proteins associated with the microproteinuria. In this study, we coupled ultracentrifugation with attenuated total reflectance-Fourier transform infrared (ATR-FTIR) to identify and quantify proteins in urine at low parts per million levels. The process involves the preconcentration of proteins from 500 μL of urine using an ultrafiltration device. After several washings, the isolated proteins …

Urban agglomerations expose citizens to ever-increasing risks from heat, air pollution, noise stress, and reduced nature connectedness. Concurrently, accumulating evidence suggests various health benefits by exposure to urban natural spaces (World Health Organization, 2016a; Bratman et al., 2019). Existing research suggests an array of benefits of contact with nature which are linked to physical activity (e.g., green exercise), active travel, and residential proximity to greenspace. Psychological benefits appear to be related to mood, well-being, attention and pro-environmental behavior; physiological benefits have been described in terms of increased physical activity, improved cardiovascular parameters, reduced stress hormones, and enhanced immune resources (Bowler …

Articles

Chronic infections of Pseudomonas aeruginosa in the lungs of cystic fibrosis (CF) patients are problematic in Ireland where inherited CF is prevalent. The bacteria's capacity to form a biofilm in its pathogenesis is highly virulent and leads to decreased susceptibility to most antibiotic treatments. Herein, we present the activity profiles of the Cu(II), Mn(II) and Ag(I) tdda-phen chelate complexes {[Cu(3,6,9-tdda)(phen)2].3H2O.EtOH}n (Cu-tdda-phen), {[Mn(3,6,9-tdda)(phen)2].3H2O.EtOH}n (Mn-tdda-phen) and [Ag2(3,6,9-tdda)(phen)4].EtOH (Ag-tdda-phen) (tddaH2 = 3,6,9-trioxaundecanedioic acid; phen = 1,10-phenanthroline) towards clinical isolates of P. aeruginosa derived from Irish CF patients in comparison to two reference laboratory strains (ATCC 27853 and PAO1). The effects of the metal-tdda-phen …

Suicide is the second leading cause of death globally among young people representing a significant global health burden. Although the molecular correlates of suicide remains poorly understood, it has been hypothesised that epigenomic processes may play a role. The objective of this study was to identify suicide-associated DNA methylation changes in the human brain by utilising previously published and unpublished methylomic datasets. We analysed prefrontal cortex (PFC, n = 211) and cerebellum (CER, n = 114) DNA methylation profiles from suicide completers and non-psychiatric, sudden-death controls, meta-analysing data from independent cohorts for each brain region separately. We report evidence for …

Mast cells play a critical role in type 1 hypersensitivity reactions. Indeed, mast cell mediators are implicated in many different conditions including allergic rhinitis, conjunctivitis, asthma, psoriasis, mastocytosis and the progression of many different cancers. Thus, there is intense interest in the development of agents which prevent mast cell mediator release or which inhibit the actions of such mediators once released into the environment of the cell. Much progress into the design of new agents has been made since the initial discovery of the mast cell stabilising properties of khellin from Ammi visnaga and the clinical approval of cromolyn sodium. …

Casein, β-lactoglobulin and α-lactalbumin are major milk protein allergens. In the present study, the structural modifications and antigenic response of these bovine milk allergens as induced by non-thermal treatment by atmospheric cold plasma were investigated. Spark discharge (SD) and glow discharge (GD), as previously characterized cold plasma systems, were used for protein treatments. Casein, β-lactoglobulin and α-lactalbumin were analyzed before and after plasma treatment using SDS-PAGE, FTIR, UPLC-MS/MS and ELISA. SDS-PAGE results revealed a reduction in the casein and α-lactalbumin intensity bands after SD or GD treatments; however, the β-lactoglobulin intensity band remained unchanged. FTIR studies revealed alterations in protein …

Optimisation Of The Transmit Beam Parameters For Generation Of Subharmonic Signals In Native And Altered Populations Of A Commercial Microbubble Contrast Agent Sonovue®, Aoife M. Ivory, James F. Meaney, Andrew J. Fagan, Jacinta Browne

Articles

The aim of this work was to establish the optimum acoustic characterisation approach and insonation transmit beam parameters for subharmonic signal generation with ‘native’ and ‘altered’ populations of a commonly-used microbubble contrast agent. Dynamic contrast-enhanced (DCE) ultrasound is a non-invasive method of imaging the microvasculature, typically implemented using harmonic imaging. Subharmonic imaging, in which echoes at half the fundamental frequency are detected, detects signals which are generated by the ultrasound contrast agents (UCAs) but not by tissue. However, optimal transmission parameters and furthermore, the optimum acoustic characterisation method have not been established. The subharmonic response of ‘native’ and ‘altered’ UCA, …
