Huntington disease (HD) is a neurodegenerative disorder caused by a CAG trinucleotide repeat expansion in the huntingtin (HTT) gene. HD manifests as a triad of psychiatric, cognitive, and motor symptoms, the latter of which defines symptomatic onset of the disease. Psychiatric symptoms including aggression and depression often emerge decades prior to motor symptom onset. Psychiatric disorders are closely linked with dysregulated neural plasticity, the ability of the brain to form new synaptic connections. Neural plasticity is impaired in HD, potentially playing a role in psychiatric symptoms. However, the association between plasticity and psychopathology in HD has yet to be thoroughly …

Dispersal and connectivity within a population often promotes overall diversity and adaptive potential, therefore a disruption of existing patterns of connectivity by local extinction can have significant consequences for the affected species. However, the extent of these impacts on recovery is not always known. American flamingos (Phoenicopterus ruber) present an ideal opportunity to investigate how changes in gene flow influence within population diversity and genetic structure over the span of a century for a recovering population. Coarse outcomes of prescribed fires are well-understood, but fine scale impacts on many assemblages are still unknown. When fire approaches, animals escape to unburned areas, hide in place, or are killed by fire. Treefrogs are unique in their use of trees for most of their adult nonbreeding activities. This reliance on uplands and three-dimensional landscape composition makes them a great model to study prescribed fire impacts. We focused analyses on an upland flatwoods specialist comprising 99% of our captures, the pinewoods treefrog Dryophytes femoralis. This study engaged in a curricular redesign of an undergraduate biochemistry course undertaken by a collaborative action research group. The initial portion of this study investigated student conceptual understanding of foundational concepts via a rigorously tested concept inventory. the impact of the redesign on the student foundational conceptual understanding was determined by comparison with a baseline data set. Findings suggest that students enter and exit the biochemistry course with little understanding of key foundational concepts. Examination of student performance after the incorporation of creative exercises showed a significant impact of the curricular changes on two of the concepts of interest. …

This dissertation examines the representation of Alzheimer's Disease (AD) neuronarratives (NN), works that centralize the brain or consciousness in the plot, in multiple modalities to support that argument that neuronarratives should be expanded into a transmedia genre. These modalities include neurogames, neurocomics, and neuromemoirs. These three modalities were analyzed to determine how selfhood and social networks are represented by characters or authors with Alzheimer's Disease. As AD is often viewed as the loss of selfhood and is associated with isolation, these works combat these stigmas and complicate representations of the disease. The Y-chromosome and mitochondria (containing mtDNA) appear entirely dissimilar, the former being a sex chromosome found in the nucleus and the latter an organelle with thousands of copies in the cytoplasm. However, both experience uniparental inheritance, with Y-chromosomes being patrilineal and mitochondria matrilineal. This leads to various other similarities including haploidy, high mutation rates, and limited or lack of genetic recombination. Moreover, the low number of protein-coding genes contained on both led to the belief that neither contributes significantly to the adaptive evolution of complex traits. Alzheimer's disease (AD) involves progressive neurodegeneration leading to the loss of normal neuronal function. Extracellular accumulation of amyloid-beta (Aß), through the abnormal cleavage of the amyloid precursor protein (APP) by ß- and γ-secretases, is one of the hallmarks of AD. Current research focuses on finding potential candidates for biomarkers and techniques with improved sensitivity for early disease detection. Extracellular vesicles (EVs) found in body fluids are a source of biomarkers for AD diagnosis. Microplastic (MP) abundances in surface waters and tissues of the oyster Crassostrea virginica were examined for one year in the Indian River Lagoon (IRL), a 251-km long estuary on the east coast of central FL, USA to better understand spatial and temporal variability. A total of 3755 MP were found in 44% of lagoon-wide water samples. Mean density (± CI) was 1.47 ± 0.09 MP/L. Central IRL sites had the lowest MP abundance, while the south had the most, likely associated with proximity to urbanization, inlets (MP sinks) and tributaries (MP sources). What’s better than simply drinking wine in a vineyard? Maybe a beautiful destination where the whole family can enjoy a day together, tasting wine, enjoying a variety of activities, and watching the kids play. Dr. Robin Back at the UCF Rosen College of Hospitality Management and his collaborators have examined a case example that presents a new business model for wine tourism—micro-clustering with diverse forms of ownership on two adjacent wine farms. The team reveals how a single South African agribusiness achieved success, and raises the possibility of other similar businesses doing the same.

Mangrove forest sustainability hinges upon propagule recruitment and seedling retention. This study evaluates biophysical limitations to mangrove seedling persistence by measuring anchoring force of two mangrove species (Rhizophora mangle and Avicennia germinans). Anchoring force was measured in 362 seedlings via lateral pull-tests administered in mangrove forests of two subtropical estuaries and in laboratory-based experiments. Removal mechanism varied with seedling age: newly-established seedlings failed due to root pull-out while seedlings older than 3 months failed by root breakage. Apalachicola Bay, an estuary located in northwest Florida, is likely to experience an increase in climate change and human-induced stressors, such as sea level rise and changes in freshwater inflow, in the future. A coupled hydrodynamic and food web modeling approach was used to simulate future scenarios of low and high river flow and sea level rise in Apalachicola Bay from 2020 to 2049 and demonstrate the range of temporal and spatial changes in water temperature, salinity, fisheries species populations and the broader food web. Identifying bacterial strains is important not only for human health research but also for various environmental and industrial applications. The plethora of available shotgun metagenomic datasets provide an unprecedented opportunity to discover novel bacterial strains in different environmental niches. In this thesis, we evaluated the existing novel-strain-based tools and showed that their performance is still suboptimal. Due to the difficulty in distinguishing strains of similar abundance by available tools, the SMS (strains in multiple samples) tool was developed to de novo identify bacterial strains in multiple shotgun metagenomic samples. The high reactivity and electron-withdrawing ability of the nitro group makes it a powerful tool in synthesizing a diverse range of bioactive natural products. The potential of the nitro group was employed via two main methodologies: iron (III)-mediated oxidative dearomatization and oxime carbonate butyrolactone formation to reach three important heterocyclic scaffolds; spiro, tropone and γ-butyrolactone. Nitro precursors were enantioselective synthesized by key nitro-aldol (Henry) and nitro-Michael reactions. A library of polysubstituted butyrolactone oxime carbonates were formed under mild DMAP and Boc2O conditions from readily available δ-nitroalcohols in good to high yields with broad functional group tolerability. DNA-based diagnostics traditionally utilize hybridization probes: strands of DNA complimentary to a target sequence that, upon binding, generate a signal to indicate the presence of the target. The classic hybridization probes are the molecular beacon (MB) and TaqMan probes, both single DNA strands with a fluorophore and quencher at opposing ends. Colorectal cancer (CRC) is the third most common and third deadliest cancer worldwide with rising incidence rates attributed to environmental risk factors like diet. Investigating how these factors impact carcinogenesis requires an understanding of how transcriptional events evolve with respect to neoplastic progression. We employed meta-transcriptomics and latent trajectory modeling to establish a compendium of profiled healthy, adenoma, and CRC samples scored by their position along a pseudotemporal axis. By interpolating a continuous scale from cross-sectional data, dynamic processes occurring throughout disease progression can be analyzed more accurately. TxtE is a protein related to cytochrome P450 enzymes, which catalyze a number of reactions that typically involve oxygen and not nitrogen. It has been discovered that TxtE can nitrate tryptophan through an unusual reaction in which it uses nitric oxide (NO) as a nitrogen donor to install the nitro group despite NO typically being considered toxic to bacteria. This project will determine if all cytochromes P450 can catalyze nitration as long as they are given NO. This will have an impact on understanding drug delivery and metabolism for which nitration is important.

Muscular dystrophy is a genetically linked myopathy with no cure available. The lack of a cure makes early detection and treatment of muscular dystrophy imperative. When reviewing protocols examining muscular fatigue at submaximal isometric contractions, proper warm-up appeared to be absent and could have caused skewed results and conclusions. This study examines the effects of implementing a warm-up protocol before fatiguing trials. In this study, 10 adult subjects conducted fatiguing protocols with the right rectus at submaximal isometric contractions. The warm-up period included a light walk along with contractions at 20% and 33% of maximal voluntary isometric contraction (MVIC) levels. …

Plastic never fully disappears, but instead breaks into smaller pieces referred to as microplastics (< 5 mm length). Microplastics are common worldwide, and more studies are needed to understand the accumulation and diversity of microplastics found in various environments. In this study, six locations were sampled for one year in the Guana River Estuary, a partially impounded system with heavily urbanized headwaters. This study was conducted in conjunction with the Guana Tolomato Matanzas National Estuarine Research Reserve (GTM NERR) and the Florida Fish and Wildlife Conservation Commission (FWC) at their water sampling stations. The objectives of this study were to investigate: 1) distribution patterns across sample sites over time, 2) the color, size, and shape of microplastics found, and 3) polymer composition. Water samples were collected from surface waters in 1-L bottles, with five replicates at each site. Sampling occurred once a month, at the beginning of each month from August 2020-August 2021. Samples were analyzed for abundance and characteristics of microplastics using a dissecting microscope and the polymer composition was determined using Fourier-transform infrared spectroscopy. No pattern was found in the distribution of microplastics from north to south or over time. Variations in plastic color, size, and polymer composition suggests that there are multiple sources of pollution into the Guana River Estuary.

In this two-part study, I investigated whether the cyclic-adenosine monophosphate response element-binding (CREB) protein has the potential to be clinically modulated as a therapeutic target for the treatment of neurodegenerative diseases. Cancer remains one of the most prevalent diseases in the United States and a leading cause of death. Large prospective studies have found significant correlations between dietary intake and cancer. Chronic inflammation promotes pro-cancer inflammatory environments promoting the formation and growth of tumors while preventing effective anti-tumor responses. With over 300,000 plant species on the planet and more than 90% of them relying on pollen for reproduction, palynology, the study of pollen grains plays a vital role in many research fields. One of them is forensic palynology, which uses pollen as a proxy to link individuals or objects to a location or instance.
