Scanning Microscopy

Low volume fungicide smoke particle deposits were studied with scanning electron microscopy and energy dispersive X-ray analysis. Under greenhouse conditions, particles deposited on Rhododendron leaf surfaces and onto inert, carbon planchets adjacent to Rhododendron leaves were characterized by electron beam analysis. Angular particles containing chlorine, identified as vinclozolin deposition, were quantified on a per unit area basis. Leaf samples collected from plants immediately adjacent to the smoke applicator had more particles than leaf samples collected elsewhere. More deposition was measured on leaf surfaces than on inert planchet surfaces. This study increased our knowledge about the effectiveness of using less fungicide …

In an attempt to elucidate three dimensional information on the organisation of the nucleus, nuclei have been extracted from ascites tumour cells or tissue culture cells by a variety of biochemical techniques, and prepared for high resolution scanning electron microscopy using an osmium-thiocarbo-hydrazide infiltration procedure which has previously proved successful for analysis of chromosome structure. Nuclei were prefixed with either Methanol-Acetic acid, glutaraldehyde or formaldehyde and then extracted by a variety of detergents with the aim of a 'biochemical dissection' of their outer elements to allow surface visualisation of the nuclear lamina. Records of AI-sired cows born between 1978 and 1982 were used to form two composite production and reproduction data sets. First (second) consisted of 35,568 (26,443) first lactations of daughters of 270 (237) sires. Traits were FCM, heifer, and first parity nonreturn rates, days between calving and first insemination, and days open, with means 5075 (5280) kg, .62 (.62), .44 (.49), 81 (81) d and 110 (111) d. (Co)variance components were estimated by REML with an expectation maximization algorithm. Sire model included age, month, herd-year effects, and relationships among sires. Simultaneous genetic evaluations for bulls and cows obtained by mixed model procedures with an animal model incorporating all numerator relationships for artificially sired Holstein cows in northeastern United States were compared with separate (Northeast) mixed model evaluations of cows and sires for predicting genetic evaluations of progeny from dam, sire, and maternal grandsire evaluations. Regression coefficients for progeny (322,104 daughters and 837 sons) evaluations on dam, sire, and maternal grandsire evaluations were for daughters: Northeast evaluations; .50, .37, and -.02 compared to theoretical coefficients of .74, .52, and -.13, and simultaneous evaluations; .61, .50, and -.09 compared to theoretical coefficients …

Embryonic blood vessels have not been grown in organ culture, in a way which might easily submit them to studies of vascular organogenesis. The chick embryo dorsal aorta is easily accessible and relatively simple to explant to culture. Its organ culture may provide a model for wounding and repair of the intima and/or media and provide a model for studies of growth (or maintenance) and differentiation. Fresh dorsal aorta and its organ culture from the chick embryo was characterized morphologically by light microscopy scanning (SEM) and transmission electron microscopy (TEM) from days 4 to 18. 18 nm colloidal gold-antitubulin and 4 nm colloidal gold-antitubulin were used to label microtubules in adherent, fully spread platelets. Both sizes of marker effectively labelled microtubules in the partially extracted platelets. However only the 4 nm gold penetrated the dense microfilament matrix of the inner filamentous zone so that portions of microtubules within this cytoskeletal zone could be tracked. The gold marker could be visualized well with 1 MeV high voltage transmission EM and with 5 kV or greater secondary imaging or 20 kV backscattered imaging of carbon only coated samples. 1 kV secondary imaging permitted high resolution imaging of …

The immediate activities of the aminoglycoside antibiotic, tobramycin, were investigated in Pseudomonas aeruginosa PAO1. The influence of carbon growth substrate and the antibiotic exposure environment in the magnitude of activity were examined. Lethality by 8 ug/ml tobramycin occurred rapidly (1 to 3 minutes). The release of specific cellular component into the supernatant was associated with lethality. This material was initially detected as an increase in UV-absorbance. Magnesium in the reaction mixture provided protection against lethality and leakage, but did not reverse lethal damage after a 3 minute tobramycin treatment. Also, uptake of 3H-tobramycin was reduced in the presence of …

This senior engineering problem is the study of the mechanism of blackening and methods of measurement for blackening. The effect of blackening is the loss of color and opacity in the sheet. The loss of color results as a darker sheet. There are 3 major variables which cause blackening: moisture content, nip pressures, and roll temperatures. The increase in any of these variables results in blackening. In this experiment, the increase of moisture content produced the most significant blackening. Pulp viscosity and zero span tensile tests can be used to predict the ultimate fiber strength of pulp fibers. A project to compare the traditional test with the newer zero span tensile test was completed. In order to compare both tests for precision, bleached softwood kraft pulp was disintegrated and made into handsheets. The handsheets were exposed to ION HCL vapor from a few minutes to a few hours to get a range in fiber strength since the HCl degrades the cellulose. Four members of a family, a mother, her two sons, and the mother's second cousin, presented with a history of isolated cataplexy without excessive daytime sleepiness or other symptoms typical of narcolepsy. They were polygraphically monitored during one night followed by a day of multiple nap tests. No subject manifested objective daytime sleepiness as measured by short latencies to sleep onset during daytime naps, and no subject exhibited periods of sleep onset rapid eye movement sleep in either the night or nap recordings. The incidence of isolated cataplexy in this family was traced by history through six generations. We conclude …

Few examples of intrachromosomal insertions have been described. These usually result from deletion of a segment of chromosome material, with insertion of the deleted material elsewhere on the same chromosome. Previous insertional translocations have been identified through a proband who has either a deletion or a duplication of the inserted segment. We describe a family which has two probands, one with a duplication and one with a deletion of the inserted segment. We report the clinicopathologic features of an infant who died of a rare form of perinatal cirrhosis associated with idiopathic hepatic and extrahepatic parenchymal siderosis. The infant appeared normal at birth but soon became severely ill, following a progressively downhill course associated with hypoglycemia, metabolic acidosis, bleeding diathesis, jaundice, and shock. The infant died at 7 days of age. The manifestations were those of hepatic failure but mimicked sepsis and disseminated intravascular coagulation. Cirrhosis, giant cell transformation, and parenchymal iron deposition characteristic of perinatal idiopathic hemochromatosis, a recently emerging clinicopathologic entity of unknown etiology, were present in this infant. Serum Creatinine Concentrations In Healthy Newborns: Reference Ranges During The First Five Days Of Life, Jennie Andreson, Jaime Alfredo Zegarra, Carlos Alberto Estrada, Francis Dumler

Serum creatinine concentrations were studied in 52 healthy, full-term infants (29 males and 23 females) during the first five days of life. At birth, mean serum creatinine concentration was 0.76 ± 0.13 mg/dL. At 6 hours of life, values increased to 0.97 ± 0.11 mg/dL (P < 0.001) and remained elevated for 24 hours. Values then returned to baseline so that serum creatinine concentrations were 0.81 ± 0.15 mg/dL at 48 hours and 0.6l ± 0.15 mg/dL at 3 to 5 days of age. At birth, serum creatinine concentrations were higher in males than in females (0.80 ± 0.13 versus 0.71 ± 0.11 mg/dL, respectively; P < 0.009). However, the increases in serum creatinine concentration observed after birth were similar in both sexes. This is the first report of a prospective longitudinal study of serum creatinine concentrations between birth and 3 to 5 days of age in a Hispanic population. In addition, data were analyzed by sex. The results are a useful reference for normal serum creatinine concentrations in early life.

Ovarian Failure In A Chromosomally Competent Adolescent Female: A Case Report, T. Jann Caison-Sorey

Primary ovarian failure in adolescence is uncommon. This report depicts such a case in a 17 1/2-year old girl whose investigation revealed elevations in the follicle-stimulating hormone and luteinizing hormone levels in association with a low estrogen level. The blood leukocyte chromosome analysis with Giemsa banding revealed a 46,XX chromosomal complement. Pelvic ultrasound disclosed a rudimentary uterus and a small left ovary. Hormonal replacement has resulted in advancement of secondary sexual characteristics and monthly withdrawal bleeding.

Electronic rectal thermometry is performed routinely in most medical centers. While electronic thermometry has been validated at the oral site, rectal measurement has not. We performed a controlled study on 84 children comparing mercury-in-glass and electronic thermometry at the rectal site. No significant difference occurred in temperatures between the two methods. We conclude that the electronic thermometer is a valid instrument to measure rectal temperature.
