Lignocellulosic biomass is a valuable feedstock for the production of liquid biofuels. However, liquid biofuel production from lignocellulose is economically challenging due to the difficulties associated with separating the cellulosic sugars from lignin. In addition, the current thermochemical methods to deconstruct lignocellulose typically involve hazardous chemicals that can negatively impact the environment. Recent studies have demonstrated the efficacy of white-rot fungi as alternative pretreatment agents that can lower the costs of lignocellulose deconstruction and reduce negative impacts on the environment. Fescue toxicosis can negatively impact growth and fertility of beef cattle grazing tall fescue infected with an endophytic fungus. The fungus produces ergot alkaloids that contribute to the hardiness of the plant, but can be detrimental to the animal if consumed. A single nucleotide polymorphism (SNP) in the dopamine receptor D2 gene (DRD2) has been associated with serum prolactin concentrations and hair coat scores in cattle grazing toxic endophyte-infected (TE) tall fescue; cattle with the GG genotype had decreased serum prolactin concentrations and increased hair coat scores. Pityopsis (Asteraceae) includes seven species; one species, P. ruthii, is federally endangered. The genus exhibits a range of ploidy levels, widespread hybridization among species with overlapping ranges, and interesting adaptive traits such as fire-stimulated flowering. However, taxonomy of Pityopsis has remained unresolved. Resolving interspecific relationships can lead to a deeper understanding of the inheritance and hybridization patterns, as well as the evolution of adaptable traits. They are used to estimate the historical parameters of a population based upon the sampling of present-day genetic information. A popular approach employs Markov chain Monte Carlo (MCMC) methods. While effective, these methods are very computationally intensive, often taking weeks to run. Although attempts have been made to leverage parallelism in an effort to reduce runtimes, they have not resulted in scalable solutions. Little information is available regarding the genetic structure and diversity of sportfishes in large rivers, which can be vital to continuing the sustainable exploitation of these fisheries. Additionally, impoundments often have vast impacts on riverine systems. These impacts include altering the flow regime, changing the nutrient load, and preventing migratory fish from reaching their spawning grounds. Chronically high blood glucose levels lead to many problems, such as insulin resistance, the hallmark of Type II diabetes. Increased flux through the hexosamine biosynthesis pathway is one mechanism by which high glucose as well as glucosamine has been shown to induce insulin resistance. This study tests the effects of glucosamine induced insulin resistance on insulin regulation of the metabolic genes glucose-6-phosphate dehydrogenase (G6PDH) and fatty acid synthase (FAS) as well as insulin responsive proteins tribbles homolog (TRIB3) and sterol regulatory element binding protein (SERBP-1c) 1c.

Lucinid clams and their sulfur-oxidizing endosymbionts comprise two compartments of a three-stage, biogeochemical relationship among the clams, seagrasses, and microbial communities in marine sediments. A population of the lucinid clam, Stewartia floridana, was sampled from a subtidal seagrass bed at Bokeelia Island Seaport in Florida to test the hypotheses: (1) S. floridana, like other lucinids, are more abundant in seagrass beds than bare sediments; (2) S. floridana gill microbiomes are dominated by one bacterial operational taxonomic unit (OTU) at a sequence similarity threshold level of 97% (a common cutoff for species level taxonomy) from 16S rRNA genes; …

In this study, Phytophthora infestans and Peronospora farinosa f. sp. spinaciae populations were analyzed using single nucleotide polymorphisms (SNPs). In Ecuador and Colombia, Phytophthora infestans causes significant damage to potato and tomato and the epidemiology is known to be highly clonal. Our objective was to measure population structure within the context of this clonal epidemiology using both synonymous and nonsynonymous markers. Candidate SNP sites were selected by comparing the draft genomes of the Ecuadorian isolates EC1-3527 and EC1-3626. The ATP-binding cassette (ABC) transporter gene superfamily is ubiquitous among extant organisms. ABC transporters act to transport compounds across cellular membranes and are involved in a diverse range of biological processes and functions including cancer resistance in humans, drug resistance among vertebrates, and herbicide resistance in weeds. This superfamily of genes appears to be larger and more diverse in the plant kingdom—yet, we know relatively less about ABC transporter function in plants compared with mammals and bacteria. The range and abundance of the Louisiana black bear (Ursus americanus luteolus) were greatly diminished during the 20th century. This subspecies was reduced to 3 small, isolated subpopulations in Louisiana as bottomland hardwood habitat was converted to agriculture. These bears were listed as threatened by the U.S. Fish and Wildlife Service in 1992 and a recovery plan was published in 1995. Recovery requires estimates of population parameters to evaluate current population status and future viability. Tall fescue (Lolium arundinaceum Schreb.) is the most prevalent forage in the Midsouth United States due in part to the presence of the endophytic fungus Neotyphodium coenophialum. The fungus, while conferring hardiness to tall fescue, contributes to decreased production efficiency in cow-calf operations. A previous genome-wide association study was performed using the Illumina 50k bovine SNP chip. Twenty-four SNP were found to be associated (P < 0.05) with adjusted birth weight and adjusted 205-day weights of calves from 48 beef cows at Ames Plantation. The first objective was to validate each SNP by testing associations with several additional phenotypes. Custom Taqman genotyping assays (Applied Biosystems, Foster City, CA) were subsequently designed to genotype each SNP in beef cattle located at Tennessee Tech University (n = 654), to validate associations in a large, independent herd. The results yielded 15 associations that were significant (P < 0.05) with 6 phenotypes linked to those affected by fescue toxicosis. The Lyme borreliosis (LB) cycle, involving Ixodes scapularis and Borrelia burgdorferi sensu stricto (Bbss), is well documented in the northeastern US, where LB is becoming increasingly prevalent. In coastal North Carolina, I. affinis has been shown to have a higher incidence of Bbss than I. scapularis. My objectives were, to assess changes in prevalence of Bbss in Ixodes spp. along a transect from Virginia to Florida, and to assess the value of dogs and mesomammals as sentinels for spread of Borrelia burgdorferi sensu lato.

The subfamily Phasiinae (Diptera: Tachinidae) is composed of obligate endoparasitoid flies that attack heteropteran insects, many of which are important agricultural pests. To lay a foundation for future studies in phasiine life history and their potential for use as biological control agents, the phylogenetic relationships within Phasiinae were explored using nucleotide sequences of two genes, CAD and LGL. A total of 63 taxa for CAD and 73 taxa for LGL were included in the analyses, representing 58 genera. Genome-wide assays performed in Arabidopsis and other organisms have revealed that the translation status of mRNAs responds dramatically to different stresses and environmental and growth signals. The steroid hormone ecdysone plays vital roles during Drosophila development. Pulses of 20E during Drosophila life cycle function as temporal cues, signaling the onset of metamorphic processes, including the stage specific programmed cell death of larval tissues. Ecdysone is the critical developmental cue orchestrating the metamorphic reformation of CNS, resulting in the formation of adult-specific neural circuitry. Ecdysone signaling is transduced by a heterodimeric receptor complex formed between two nuclear receptors: EcR and Ultraspiracle (USP). A recent study using human fecal samples in conjunction with a culture-independent approach revealed immense diversity of antibiotic resistance (AR) genes in the human gut microflora. We hypothesize that food animal gut microflora also contain diverse and novel AR genes which could contribute to the emergence and transmission of AR in pathogens important in animal and human health. To test this, we examined AR reservoir in chicken gut microflora using a metagenomic, functional cloning method.
